The complexities underlying age-related macular degeneration： could amyloid beta play an important role？

e-related macular degeneration （AMD） causes irreversible loss of central vision for which there is no effective treatment. Incipient pathology is thought to occur in the retina for many years before AMD manifests from midlife onwards to affect a large proportion of the elderly. Although genetic as well as non-genetic/environmental risks are recognized, its complex aetiology makes it difficult to identify susceptibility, or indeed what type of AMD develops or how quickly it progresses in different individuals. Here we summarize the literature describing how the Alzheimer＇s-linked amyloid beta （Aβ） group of misfolding proteins accumulate in the retina. The discovery of this key driver of Alzheimer＇s disease in the senescent retina was unexpected and surprising, enabling an altogether different perspective of AMD. We argue that Aβ fundamentally differs from other substances which accumulate in the ageing retina, and discuss our latest findings from a mouse model in which physiological amounts of Aβ were subretinally-injected to recapitulate salient features of early AMD within a short period. Our discoveries as well as those of others suggest the pattern of Aβ accumulation and pathology in donor aged/AMD tissues are closely reproduced in mice, including late-stage AMD phenotypes, which makes them highly attractive to study dynamic aspects of Aβ-mediated retinopathy. Furthermore, we discuss our findings revealing how Aβ behaves at single-cell resolution, and consider the long-term implications for neuroretinal function. We propose Aβ as a key element in switching to a diseased retinal phenotype, which is now being used as a biomarker for latestage AMD.

Scholars have criticized the capacity of playgrounds to support children’s participation in public *** of childhood,such as walls,fences,and enclosures,dominate children’s"public”spatial experiences in the global *** well-established critiques of the fenced playground as a space that segregates and controls childhood experiences,this study offers a novel and nuanced perspective,emphasizing the qualities of the playground fence that support play and playful connections,on,through,and around *** an ethnographic methodology,this study includes 167h of observations in three typical urban public playgrounds in Greece and 65 semi-structured interviews with 124 *** on recursive thematic qualitative analysis,the fence emerges as a blurred boundary,that is,an element that transgresses assumptions and questions spatial classifications and *** the subject of design discourse,these findings are particularly significant in design disciplines globally and offer new understandings on the possibilities afforded by the playground *** themes,namely,indeterminacy,climbabilty,playability,and porosity,are proposed as principles to guide fenced playground design as part of a fundamental *** reconceptualization positions the fenced playground as a public space infrastructure,supporting intergenerational interaction and play as well as children’s presence and play in the public realm.

The computerized LASSI-BC Test versus the Standard LASSI-L Paper-and-Pencil Version in Community-Based-Samples

Proactive Semantic Interference (PSI) and failure to recover from PSI (frPSI), are novel constructs assessed by the LASSI-L. These measures are sensitive to cognitive changes in early Mild Cognitive Impairment (MCI) and preclinical AD determined by Aβ load using PET. The goal of this study was to compare a new computerized version of the LASSI-L (LASSI-Brief Computerized) to the standard paper-and-pencil version of the test. In this study, we examined 110 cognitively unimpaired (CU) older adults and 79 with amnestic MCI (aMCI) who were administered the paper-and-pencil form of the LASSI-L. Their performance was compared with 62 CU older adults and 52 aMCI participants examined using the LASSI-BC. After adjustment for covariates (degree of initial learning, sex, education, and language of evaluation) both the standard and computerized versions distinguished between aMCI and CU participants. The performance of CU and aMCI groups using either form was relatively commensurate. Importantly, an optimal combination of Cued B2 recall and Cued B1 intrusions on the LASSI-BC yielded an area under the ROC curve of .927, a sensitivity of 92.3% and specificity of 88.1%, relative to an area under the ROC curve of .815, a sensitivity of 72.5%, and a specificity of 79.1% obtained for the paper-and-pencil LASSI-L. Overall, the LASSI-BC was comparable, and in some ways, superior to the paper-and-pencil LASSI-L. Advantages of the LASSI-BC include a more standardized administration, suitability for remote assessment, and an automated scoring mechanism that can be verified by a built-in audio recording of responses.

Indian Ocean SST modes and Their Impacts as Simulated in BCC_CSM1.1(m) and HadGEM3

The sea surface temperature anomalies （SSTAs） in the tropical Indian Ocean （TIO） show two dominant modes at interan- nual time scales, referred to as the Indian Ocean basin mode （IOBM） and dipole mode （IOD）. Recent studies have shown that the IOBM and IOD not only affect the local climate, but also induce remarkable influences in East Asia via teleconnections. In this study, we assess simulations of the IOBM and IOD, as well as their teleconnections, using the operational seasonal pre- diction models from the Met Office （HadGEM3） and Beijing Climate Center [BCC_CSMI.I（m）]. It is demonstrated that the spatial patterns and seasonal cycles axe generally reproduced by the control simulations of BCC_CSM1.1 （m） and HadGEM3, although spectra biases exist. The relationship between the TIO SSTA and E1 Nifio is successfully simulated by both mod- els, including the persistent IOBM warming following E1 Nifio and the IOD-E1 Nifio interactions. BCC_CSMI.I（m） and HadGEM3 axe capable of simulating the observed local impact of the IOBM, such as the strengthening of the South Asian high. The influences of the IOBM on Yangtze River rainfall are also captured well by both models, although this teleconnec- tion is slightly weaker in BCC_CSM 1.1 （m） due to the underestimation of the northwestern Pacific subtropical high. The local effect of the IOD on East African rainfall is reproduced by both models. However, the remote control of the IOD on rainfall over southwestern China is not clear in either model. It is shown that the realistic simulations of TIO SST modes and their teleconnections give rise to the source of skillful seasonal predictions over China.

Background:This study examined the volume and patterns of physical activity(PA) and sedentary time(ST) across different segments of the week among boys and ***:A total of 188 children aged 7-12 years wore a wrist-mounted ActiGraph GT3 X+accelerometer for 7 *** spent in PA and ST was calculated using ActiLife *** mean number of minutes of light PA,moderate PA,vigorous PA,moderate-to-vigorous PA(MVPA),and ST were calculated per weekday(before school,during school,and after school) and per weekend day(morning and afternoon-evening).Results:After school represented the greatest accumulation of ST compared with before school and during school *** engaged in225.4 min/day of ST(95% confidence interval(CI):216-235),and girls engaged in 222.2 min/day of ST(95%CI:213-231).During school,boys engaged in significantly more MVPA than girls(46.1 min/day(95%CI:44-48) vs.40.7 min/day(95%CI:39-43)).Across the whole weekday,boys participated in significantly more MVPA than girls(103.9 min/day(95%CI:99-109) vs.95.7 min/day(95%CI:90-101)).The weekend afternoon—evening segment represented the larger accumulation of ST,where boys were significantly more sedentary than girls(367.5 min/day(95%CI:353-382) vs.339.8 min/day(95%CI:325-355),respectively).Conclusion:Our findings suggest that children are highly sedentary and spend little of their time in school in MVPA,especially *** breaks in school elicit increases in light PA and *** work should consider the use of more active breaks within school time to encourage PA and reduce ST.

Immediate lymphatic reconstruction(ILR)has become increasingly utilized for the prevention of breast cancerrelated lymphedema(BCRL).A growing body of evidence has demonstrated the long-term efficacy of ILR in reducing the rate of *** certain risk factors for BCRL are well-recognized,such as axillary lymph node dissection,regional lymph node radiation,and elevated body mass index,other potential risk factors such as age and taxane-based chemotherapeutics remain under *** experience with ILR has highlighted an additional potential risk factor for *** anatomy,specifically compensatory lymphatic channels that bypass the axilla,may play a largely underrecognized role in determining which patients will develop BCRL after *** anatomic knowledge has primarily been based on cadaveric studies that predate the twentieth *** approaches to lymphatic anatomical mapping using indocyanine green lymphography have helped to elucidate baseline lymphatic anatomy and compensatory channels,and certain variations within these channels may act as anatomic risk ***,the purpose of this review was to highlight ways in which variations in lymphatic anatomy can inform the application and improve the accessibility of this *** ILR continues to advance and evolve,anatomical mapping of the lymphatic system is valuable to both the patient and lymphatic microsurgeon and is a critical area of future study.

Numerical modelling of supercritical flow in circular conduit bends using SPH method

The capability of 1he smoothed-particle hydrodynamics （SPH） method to model supercritical flow in circular pipe bends is considered. The standard SPH method, which makes use of dynamic boundary particles （DBP）, is supplemented with the original algorithm for the treatment of open boundaries. The method is assessed through a comparison with measured free-surface profiles in a pipe bend, and already proposed regression curves for eslimation of the flow-type in a pipe bend. The sensitivity of the model to different parameters is also evaluated. It is shown that an adequate choice of the artificial viscosity coefficient and the initial particle spacing can lead to correct presentation of the flow-type in a bend. Due to easiness of its implementation, the SPH method can he efficiently used in the design of circular conduits with supercritical flow in a bend, such as tunnel spillways, and bottom outlets of dams, or storm sewers.
