differential regulation of protein synthesis in skeletal muscle and liver of neonatal pigs by leucine through an mTORC1-dependent pathway

Neonatal growth is characterized by a high protein synthesis rate that is largely due to an enhanced sensitivity to the postprandial rise in insulin and amino acids, especially leucine. The mechanism of leucine＇s action in vivo is not well understood. In this study, we investigated the effect of leucine infusion on protein synthesis in skeletal muscle and liver of neonatal pigs. To evaluate the mode of action of leucine, we used rapamycin, an inhibitor of mammalian target of rapamycin （mTOR） complex-1 （mTORC1）. Overnight-fasted 7-day-old piglets were treated with rapamycin for 1 hour and then infused with leucine （400 μmol·kg^-1·h^-1） for 1 hour. Leucine infusion increased the rate of protein synthesis, and ribosomal protein S6 kinase 1 （S6K1） and eukaryotic initiation factor （elF） 4E-binding protein-1 （4E-BP1） phosphorylation in gastrocnemius and masseter muscles （P 〈 0.05）, but not in the liver. The leucine-induced stimulation of protein synthesis and S6K1 and 4E-BP1 phosphorylation were completely blocked by rapamycin, suggesting that leucine action is by an mTORC1-dependent mechanism. Neither leucine nor rapamycin had any effect on the activation of the upstream mTQRC1 regulators, AMP-activated protein kinase and protein kinase B, in skeletal muscle or liver. The activation of elF2α and elongation factor 2 was not affected by leucine or rapamycin, indicating that these two pathways are not limiting steps of leucine-induced protein synthesis. These results suggest that leucine stimulates muscle protein synthesis in neonatal pigs by inducing the activation of mTORC1 and its downstream pathway leading to mRNA translation.

Autoantibodies against G-Protein-Coupled Receptors Modulate Heart Mast Cells

Mast cells are believed to be involved in myocardial tissue remodelling under pathophysiological conditions. We examined the effects of autoantibodies against G-protein-coupled receptors in sera of patients with heart diseases on myocardial mast cells in the cultured neonatal Sprague-dawley rat heart cells. Cells collected at day 3 and 10 of the culture were preincubated with autoantibodies against α-adrenoceptor and angiotensin Ⅱ ATl-receptor, agonist phenylephrine and angiotensin Ⅱ, and control IgG. The pretreated cultured cells were stained for selected mast cell markers tryptase, chymase and TNF-α The cultured cells were also processed for observation with electron microscopy. The autoantibodies-treatment of the 3-day cultured cells caused both increased intensity of immunofluorescence （p 〈 0.05） and their enlarged diameters of the mast cells when compared to age-matched ones. In contrast, the fluorescence of preincubated 10-day-old mast cells was decreased compared with controls （p 〈 0.01）. In control samples, the fluorescence of 10-day-old mast cells was significantly higher than that of 3-day-old ones （p 〈 0.001）. Results of electron microscopy examination demonstrated there was an increased granulation of treated 3-day-old mast cells, while a degranulation of mast cells at day 10 of application. The results suggest the modulation effect of the autoantibodies against G-protein-coupled receptors on mast cells, indicating a potential functional link between the autoantibodies against G-protein-coupled receptors and the mast cells in progression of heart disease.

Root architecture characteristics of plant inlay in live slope grating

In the experimental garden of the department of Soil Bioengineering and Landscape Construction, University of Applied Life Sciences in Vienna, Austria, coarse root systems of three different brush species were completely excavated and semiutomatically digitized. The species were Lonicera xylosteum, Ligustrum vulgare and Euonymus europaeus. The 3-d root architectures reveal different growth strategies between species, which are related to ecological characteristics and physical soil properties. The root architecture of Lonicera xylosteum and Ligustrum vulgare, planted in the under layer of the live slope grading, where the soil is very tight and the soil water content and fertility are relatively low, is shallow. However, the root distribution of E. europaeus, planted in the middle layer, where environmental conditions are better, is deeper. Most of the root biomass of the three species is concentrated in the 0-30 cm soil layer. A quarter of the root biomass ofLigustrum vulgare is distributed in the upper layer of the plant inlay. E. europaeus has a relatively even distribution in the 30-0 cm and 60-90 cm soil layer.

Three-dimensional Rotational Angiography in Congenital Heart disease:Estimation of Radiation Exposure

Objectives: There is an increasing use of three-dimensional rotational angiography (3d-RA) during catheterization of congenital heart disease. dose-area-product (dAP) measured by the angiography system and computed-tomography dose index (CTdI) do not appear practical for dose assessment. Hence, we performed real dose measurements in anthropomorphic phantoms. Methods: Three different anthropomorphic phantoms (10 kg, 19 kg and 73 kg bodyweight) equipped with thermoluminescent dosimeters (TLd) were used. We used a typical standard diagnostic program and a low-dose program. The effective dose (Ed) was calculated according to the International Commission on Radiological Protection (ICRP) 103. The 3d distribution of radiation in the body was assessed. Results: Ed for the male 10 kg phantom was 0.192 mSv in the diagnostic program and 0.050 mSv (male) in the low-dose program. The 19 kg phantom received an Ed of 0.205 mSv (male) in the diagnostic program. In the low-dose program the Ed reached 0.058 mSv (male). The male adult 73 kg phantom was exposed with an Ed of 0.730 mSv in the diagnostic program and 0.282 mSv in the low-dose program. Ed for the female phantoms was slightly higher for both acquisition-programs. dose distribution was inhomogeneous with a dose maximum in the esophageal region behind the heart, whereas in the brain, intestine and gonads we found nearly no radiation. Conclusions: 3d-RA imaging in the interventional catheter laboratory is possible with an effective dose lower than 1 mSv. With its potential to reduce fluoroscopic time and the number of control angiographies in catheterization and intervention in complex anatomy, it can decrease the radiation dose.
