Growth inhibitory effects of Phyllanthus niruri extracts in combination with cisplatin on cancer cell lines

AIM：To investigate the cytotoxic effects of spray-dried extracts of Phyllanthus niruri in combination with cis- platin on two cancer cell lines. MeTHODS： Colorectal carcinoma （HT29） and human hepatocellular carcinoma （HepG2） cells were treated with spray-dried extracts of Phyllanthus niruri （SDePN） either alone or in combination with cisplatin at differ- ent concentrations （0.5 mg/mL and 1 mg/mL） for 4 h and 24 h. To verify and quantify cancer cells treated with these products as well as identify the cell cycle stage and cell viability, we stained the cells with prop- idium iodide and assessed them by flow cytometry. The percentage of cells in different cell cycle phases was quantified and data were expressed as histo- grams. Significant differences between groups were determined using analysis of variance and Bonferroni＇s test, as indicated. A value of P 〈 0.05 was considered to be statistically significant. ReSULTS： SDePN had significantly different cyto- toxic effects on HT29 （2.81 4- 0.11 vs 3.51 4- 1.13, P 〉 0.05） and HepG2 （5.07± 0.3 vs 15.9 ± 1.04, P 〈 0.001） cells when compared to control cells for 4 h. SDePN also had significantly different cytotoxic effects on HT29 （1.91 ± 0.57 vs 4.53± 1.22, P 〉 0.05） and HepG2 （14.56 ± 1.6 vs 35.67 ± 3.94, P 〈 0.001） cells when compared to control cells for 24 h. Both cell lines were killed by cisplatin in a dose-dependent manner compared to control cells （HepG2 cells for 4 h： 10.78 ± 1.58 vs 53.89 ± 1.53, P 〈 0.001; 24 h： 8.9 ± 1.43 vs 62.78 ± 1.87, P 〈 0.001 and HT29 cells for 4 h： 9.52 ±0.913 vs 49.86 ± 2.89, P 〈 0.001; 24 h： 11.78 ± 1.05 vs 53.34 ± 2.65, P 〈 0.001）. In HT29 cells, pretreat- ment with SDePN and subsequent treatment with cis-platin resulted in a greater number of cells being killed （12.78 ± 1.01 vs 93.76 ± 1.6, P 〈 0.001）. What have we learned about the kallikrein-kinin and renin-angiotensin systems in neurological disorders?

The kallikrein-kinin system(KKS) is an intricate endogenous pathway involved in several physiological and pathological cascades in the brain. Due to the pathological effects of kinins in blood vessels and tissues, their formation and degradation are tightly controlled. Their components have been related to several central nervous system diseases such as stroke, Alzheimer's disease, Parkinson's disease, multiple sclerosis, epilepsy and others. Bradykinin and its receptors(B1R and B2R) may have a role in the pathophysiology of certain central nervous system diseases. It has been suggested that kinin B1R is up-regulated in pathological conditions and has a neurodegenerative pattern, while kinin B2R is constitutive and can act as a neuroprotective factor in many neurological conditions. The renin angiotensin system(RAS) is an important blood pressure regulator and controls both sodium and water intake. AngⅡ is a potent vasoconstrictor molecule and angiotensin converting enzyme is the major enzyme responsible for its release. AngⅡ acts mainly on the AT1 receptor, with involvement in several systemic and neurological disorders. Brain RAS has been associated with physiological pathways, but is also associated with brain disorders. This review describes topics relating to the involvement of both systems in several forms of brain dysfunction and indicates components of the KKS and RAS that have been used as targets in several pharmacological approaches.

目的探讨脂多糖诱导巨噬细胞（RAW264．7细胞）释放的外泌体在小鼠急性肺损伤中的作用及其机制。方法体外培养RAW264．7细胞，设置对照组和脂多糖刺激组，超速离心法提取两组细胞的外泌体，分别命名为对照eXO组和脂多糖eXO组。（1）体内实验：将18只C57BL／6雄性小鼠按随机数字表法随机均分成正常对照组、正常外泌体组、脂多糖刺激外泌体组各6只，造模12h后放血处死，取肺组织行He染色；行白细胞介素（IL）-1β和肿瘤坏死因子（TNF）-α的免疫组化评价；Western印迹检测各组IL-1β、TNF-α、β-链蛋白、e-钙黏蛋白、紧密连接蛋白l（ZO-1）和封闭蛋白的相对表达量；行组织切片免疫荧光对比各组间封闭蛋白的表达差异。（2）体外实验：将磷酸缓冲盐溶液（PBS）、对照eXO组和脂多糖eXO组外泌体与小鼠Ⅱ型肺泡上皮细胞（MLe-12细胞）共培养，设置空白对照组、对照eXO刺激组、脂多糖eXO刺激组。12h后检测胞中IL-1β、TNF-α和封闭蛋白的相对表达量。结果对照eXO组和脂多糖eXO组鉴定为外泌体。（1）体内实验：脂多糖刺激外泌体组肺组织出现肺泡腔及问质内广泛炎症细胞浸润，肺泡腔内有渗出液，肺泡隔增厚，肺泡及问质充血、出血，部分肺泡塌陷、不张等肺损伤表现；脂多糖刺激外泌体组肺组织中IL-18和TNF-α蛋白相对表达量均显著高于正常对照组、正常外泌体组（1．331±0．203比0．274±0．018、0．892±0．074；0．800±0．096比0．5964-0．025、0．441±0．061；均P〈0．05），脂多糖刺激外泌体组肺组织中封闭蛋白相对表达量均显著低于正常对照组、正常外泌体组（0．2514-0．021比0．862±0．029、0．453±O．013；均P〈0．05）。（2）体外实验：脂多糖eXO刺激组中IL-1β和TNF-α蛋白相对表达量均显著高于空白对照组、对照eXO刺激组（0．900±0．033比0．320±0．030、0．661±0．028；0．739±0．045比0．151±0．024、0．360±0．037；均P〈0．05），脂多糖eXO刺激组封闭蛋白相对表达量均显著低于空白对照组、对照eXO刺激组（0．585±0．082比0．941±0．090、0．732±0．083；均P〈0．05）。结论脂多糖诱导的RAW264．7细胞释放的外泌体导致小鼠发生急性肺损伤，且与肺泡Ⅱ型上皮细胞封闭蛋白的下调密切相关。

Residual effect of Zinc Application Doses and Methods on Nutrition and Productivity of Corn

Zinc is the micronutrient that presents the greatest response in corn grain production. However, the mode of application and the dose used are decisive factors with respect to fertilizer efficiency in the crop. Thus, the objective of the present study was to evaluate the residual effect of zinc application methods and rates on corn nutrition and productivity. The experiment was conducted in the experimental area of the Teaching, Research and Production Farm of the FCAV/UNeSP. Soil in the area is classified as dystrophic Dark-Red Latosol (dRL). The experimental setup consisted of a randomized complete block design with four replications. Treatments included three localized Zn doses applied to the soil (planting row);three Zn doses incorporated into the soil at depth of 0 - 20 cm;foliar application, seed application and treatment without addition of Zn. Two evaluations of growth during cultivation were performed. Grain productivity of the crop and the zinc content in soil, leaves and seeds were evaluated. Foliar spraying with Zn simultaneously promoted increased leaf Zn content and basal diameter of the plant. The Zn contents in grains and in the soil, as well as grain yield, were not affected by the Zn application methods or the applied doses. The residual effect of incorporated Zn doses was observed only in the basal diameter and number of leaves at 60 days after emergence.

在地震资料处理中,多次波压制的重点是自由表面多次波。阐述了利用反馈迭代法预测并压制自由表面多次波的基本原理和实现步骤。反馈迭代法不需要地下界面信息,能够适应较复杂地下结构的情况。设计了水平海底层状介质模型和起伏海底层状介质模型,对反馈迭代法压制自由表面多次波的效果进行了试验和分析,并与抛物线拉冬变换处理结果进行了对比。结果表明,反馈迭代法在迭代次数较少的情况下就能有效地去除合成地震记录中的自由表面多次波;抛物线拉冬变换的压制效果不太理想,处理后的结果中残留有较强的自由表面多次波。

Presence of the Genes cagA, cage, virB11 and Allelic Variation of vacA of Helicobacter pyloriAre Associated with the Activity of Gastritis

Non-atrophic active chronic gastritis (ACG) is characterized by the presence of H. pylori in the gastric epithelium, known to be one of the first steps that precede progression to gastric adenocarcinoma. Inactive chronic gastritis (ICG) suggests that the patient has H. pylori gastritis, but this diagnosis is rarely made in routine histopathology. Clinical manifestations associated with H. pylori infection are potentially due to differences in virulence between strains;however, it is unclear if the progression of ACG to ICG depends on the H. pylori strain. The aim of this study was to compare the prevalence of the virulence factors of H. pylori found in patients with ACG and ICG, and its influence on the development of ICG. A significant association was observed between H. pylori detection by histological examination and the activity of gastritis (p 1 year) was reported by 28.6% of the ACG group and 42.5% of the ICG, while no evidence of association between long-term use of PPI and decreased inflammation was found in the patients studied. The genes cagA, cage and virB11 were statistically associated with ACG (p = 0.01, p vacAs1 allele groups, ACG was associated with the most virulent group (p = 0.0015), while ICG was associated with the less virulent group (p e rate of co-infection was significantly higher in ICG than in ACG cases (p = 0.02). In conclusion, this study points to the role of virulent strains of H. pylori in the non-resolution of gastritis.

We present numerical studies on the switching characteristics of a fiber Bragg grating (FBG) with modulation in the third order nonlinear index of refraction along it’s length. The FBG is operating in a continuous wave regime (CW). This study was done taking into account the possible asymmetry brought by the non harmonic modulation of the nonlinearity, leading to different reflection and transmission characteristics, that depend on the direction of propagation along the modulated nonlinear FBG. This phenomenon may be useful for applications like an optical isolator. It was found that for a set of values of the modulation parameter, the FBG can exhibit multistable states. The numerical studies were obtained starting from the coupled-mode equations solved from the coupled-mode theory and simulated using the fourth-order Runge-Kutta method.

Use of Foliar Fertilizers for the Specific Physiological Management of Different Soybean Crop Stages

In addition to essential macronutrients and micronutrients, bioregulators, biostimulants or bioactivators are being increasingly used on agricultural crops with important outcomes. Therefore, improved knowledge on the functioning of these chemicals on plants is needed. Based on the assumption that foliar fertilizers applied at specific times promote soybean growth, development and yield, the aim of the present study was to evaluate the biometric, biomass, physiological, nutritional and grain yield parameters to increase knowledge about specific fertilizers applied alone or in combination and at different stages of soybean growth and development. The present study was performed in partnership between the Goiano Federal Institute (Instituto Federal Goiano), Campus Rio Verde, state of Goiás (GO), Brazil, and the company Tecno Nutricao Vegetal e Biotecnologia Ltda. A randomized block experimental design was used with four replicates. The biometric, physiological, nutritional and yield data were subjected to analysis of variance followed by Tukey’s test when significant differences were found at p 0.05 using the SISVAR software. In the emergency and early use of Tonik combined with Nodumax and PreventCoMo, in the vegetative phase, the Lumix combined with Vivat or VivatMn in the reproductive stage of flowering, 2 application Tripper combined with VivatB at a dose of 500 ml·ha-1 and reproductive phase of grain filling, 2 application Apport at a dose of 500 g·ha-1 gave the best results.

AIM: To evaluate changes in neurotransmission induced by a psychoactive beverage ayahuasca in the hippocampus and amygdala of naive rats. MeTHODS: The level of monoamines, their main metabolites and amino acid neurotransmitters concentrations were quantified using high performance liquid chromatography(HPLC). Four groups of rats were employed: saline-treated and rats receiving 250, 500 and 800 mg/kg of ayahuasca infusion(gavage). Animals were killed 40 min after drug ingestion and the structures stored at-80 ℃ until HPLC assay. The data from all groups were compared using Analysis of variance and Scheffé as post test and P epted as significant. ReSULTS: The results showed decreased concentrations of glycine(GLY)(0.13 ± 0.03 vs 0.29 ± 0.07, P e amygdala of rats that received 500 of ayahuasca.
