Preclinical evaluation of azathioprine plus buthionine sulfoximine in the treatment of human hepatocarcinoma and colon carcinoma

AIm: To evaluate the efficacy and the safety of azathioprine (AZA) and buthionine sulfoximine (BSO) bylocalized application into HepG2 tum.r in ***: Different hepatoma and colon carcinoma cell lines (HepG2, HuH7, Chang liver, LoVo, rKO, SW-48, SW-480) were grown in minimal essencial medium supplemented with 10% fetal bovine serum and 1% antibiotic/antimycotic solution and maintained in a humidified 37 ℃ incubator with 5% CO2. These cells were pretreated with BSO for 24 h and then with AZA for different times. We examined the effects of this combination on some proteins and on cellular death. We also studied the eff icacy and the safety of AZA (6 mg/kg per day) and BSO (90 mg/kg per day) in HepG2 tum.r growth in vivo using athymic mice. We measured safety by serological m.rkers such as aminotransferases and creatine ***: The in vitro studies revealed a new mechanism of action for the AZA plus BSO combination in the cancer cells compared with other thiopurines (6-m.rcaptopurine, 6-methylm.rcaptopurine, 6-thioguanine and 6-methylthioguanine) in combination with BSO. The cytotoxic effect of AZA plus BSO in HepG2 cells resulted from necroptosis induction in a mitochondrial-dependent manner. From kinetic studies we suggest that glutathione (GSH) depletion stimulates c-Jun amino-terminal kinase and Bax translocation in HepG2 cells with subsequent deregulation of mitochondria (cytochrome c release, loss of mem.rane potential), and proteolysis activation leading to loss of mem.rane integrity, release of lactate dehydrogenase and DNA degradation. Some of this biochemical and cellular changes could be reversed by N-acetylcysteine (a GSH replenisher). In vivo studies showed that HepG2 tum.r growth was inhibited when AZA was combined with ***: Our studies suggest that a combination of AZA plus BSO could be useful for localizedtreatment of hepatocellular carcinoma as in the currently used transarterial chemoembolization method.

The goal of our work was to locate and quantify changes that occurred in 66% of the mexican coastline, based on four land cover maps generated by the mexican mangrove monitoring System(Smmm) of the National Commission for the Knowledge and Use of Biodiversity(CONABIO) for the years 1970/81, 2005, 2010, and 2015. Our results showed overall dominance of erosion over accretion processes, beaches being the most affected coastal land cover. Emphasis was placed on identification and description of coastline sites in which land was either continuously lost(erosion) or gained(accretion) during the studied time periods. These sites were defined as continuous unidirectional dynamic sites and were compared with previous knowledge about the geodynamics of mexican coasts. Continuous unidirectional dynamic sites were distributed throughout the study area and within all land cover types, but predominantly corresponded to areas covered by mangroves in the states of Campeche and Nayarit. Finally, we found an intensification of coastal erosion-accretion processes over time;coastline change rates having duplicated between the earliest(1970/81–2005) and the two m.re recent(2005–2010, and 2010–2015) analysed time periods, with erosion rates for each corresponding period of –3 m/yr,–7.5 m/yr, and –7.3 m/yr,and accretion rates of 2.8 m/yr, 7.3 m/yr, and 6.9 m/yr, respectively.

This research is framed within the affective computing, which explains the importance of emotions in human cognition (decision making, perception, interaction and human intelligence). Applying this approach to a pedagogical agent is an essential part to enhance the effectiveness of the teaching-learning process of an intelligent learning system. This work focuses on the design of the inference engine that will give life to the interface, where the latter is represented by a pedagogical agent. The inference engine is based on an affective-motivational model. This model is implemented by using artificial intelligence technique called fuzzy cognitive maps.

Degradation of Oxo-Degradable-Polyethylene and Polylactic Acid Films Embodied in the Substrate of the Edible Fungus <i>Pleurotus ostreatus</i>

Degradable plastic mulch is being used to overcome the negative environmental impacts of burning and landfilling agricultural plastic waste. In this study P. ostreatus was used to model the capacity of a vegetal species to degrade conventional and degradable plastic films. Plastics studied were oxo-degradable polyethylene (OXO-PE), UV-irradiated oxo-degradable polyethylene (UV-OXO-PE), polylactic acid (PLA) and conventional polyethylene (C-PE). The cultivation of P. ostreatus resulted in a reduction in the median of weight (78.2% - 80.2%) and volume (56.1% - 60.1%) of the substrate (wheat straw). Degradation of the plastics embodied was evidenced by a reduction in the median of the elongation at break (OXO-PE 475% to 109%, UV-OXO-PE 23% to 8%, PLA 596% to 398% and C-PE 505% to 304%) and an increase in the median of the carbonyl index (OXO-PE 0.062 to 0.114, UV-OXO-PE 0.098 to 0.145 and PLA 0.024 to 0.034). The Kruskal-Wallis test found no statistical difference (p = 0.384) between the medians of the biological efficiency for substrates containing plastics and the substrate without plastic. In conclusion, plastics embodied in the substrates used for cultivation of P. ostreatus are degraded and the degradation of these plastics does not affect the short term growth of P. ostreatus.

Implementation of an Available Parking Space Detection System in Hectic Parking Lots

Parking automation has been developed for locating available parking spaces inside a parking lot with new technologies, such as automatic collection, capacity, or empty spaces’ detection;to both reduce search time and avoid crowds of vehicles waiting to park in it. A simple detection system of available spaces, within the Faculty and Staff parking lot of the Engineering and Chemistry Departments at the Autonomous University of Chihuahua, mexico (UACH), was set up. First, an analysis of vehicle capacity was carried out to establish the peak hours of vehicles’ entrances and compare it to the parking lots’ capacity. For one working week (6 days), it was determined that Tuesdays have the highest vehicle occupancy, and Saturdays are the days with the least traffic. Second, the geometric design of the parking lot (divided into two areas because of the terrain topography: upper and bottom levels), its dimensions and location of parking spots and marks on the pavement for traffic directions, were obtained from its blueprint, to visualize how the surveillance cameras can be oriented to cover the area. A detection system prototype was created for a specific area of the parking lot, using an artificial vision technique to perform the video extraction, while the digital image processing was developed in MATLAB. To complement and achieve the objective, Object- Oriented Programming (OOP) was used to obtain the count analysis as well as the availability/occupancy of each of the parking areas. Effect of split applications of urea on protein size distribution, physical dough properties, and baking performance of five experimental bread wheat lines

Five experimental bread wheat lines (BWL) were grown at the Yaqui Valley Experimental Station in Sonora, méxico during two consecutive growing cycles. The effect of five nitrogen fertilization (NF) treatments on protein size distribution (PSD), physical dough properties (PDP), and baking performance of the BWL was evaluated. Polym.ric and monom.ric proteins were evalu-ated by SE-HPLC. PDP were carried out by the National mixograph and the TA-XT2 Texture Analyzer. Baking performance was evaluated using the straight dough method. Protein content (PC), main protein fractions (glutenins, gliadins, albumins-globulins), and mixograph development time (mDT) were significantly influenced by NF treatment and BWL but not by their interaction. On the other hand, PDP measured by the Kieffer rig, and baking performance were significantly influenced by the main factors (NF and BWL) and their interaction. The amount and timing of fertilizer applied to the BWL modified the PC, PSD, PDP, and bread loaf volume. PDP exhibited a larger variation in comparison to the PSD of glutenins. The split application of 150 kg of urea/ha (50-50-50) to all BWL showed a better loaf volume response than the same amount of urea applied at sowing (150-0-0). The application of 300 kg of urea/ha to all BWL, either at sowing or at three split applications of 100 kg of urea/ha each, resulted in higher flour unextractable polym.ric protein (FUPP). On the other hand, the split application of 100-100-100 kg of urea/ha to three of the BWL represented the higher total unextractable polym.ric protein (TUPP). Differences on PC and PSD were reflected on differences on PDP and bread loaf volume observed among the BWL.

mollusks of the rocky Intertidal Zone at Three Sites in Oaxaca,mexico

The community of mollusks at three rocky intertidal zone in Oaxaca, mexico was analyzed. Two samplings per site were conducted. At each sampling the inspected area was 10 m2. The sampling unit was 1 m2. 5862 specimens were examined. 68 mollusks species were identified. The families better represented in species richness were: FISSUrELLIDAE, m.rICIDAE, LOTIIDAE, mYTILIDAE, CHAmIDAE, ISCHNOCHITONIDAE y CHITONIDAE. The species that had the greatest density were Lottia discors, Siphonaria palmata, Lottia acutapex, Chormoytilus palliopunctatus, Brachidontes adamsianus, Chton articulatus e Ischnochiton muscarius. The species with the widest distribution represent 11.76%. With irregular distribution was found in 58.82% of the species. The gastropod Crucibulummonticulus, the bivalve C. palliopunctatus and the polyplacophoran C. articulatus had the greatest size. The value of diversity index was H’ = 4.29 bits/ind. and J’ = 0.72. The mollusks community in the rocky intertidal zone study sites is characterized by small size, because they rarely exceed seventy millimeters. The values of H’ and J’ indicate that the mollusk community in the study area has a high diversity and high uniformity, corresponding to mature and stable communities in a tropical region.

Idiopathic gastric partial necrosis

We present the case of an elderly patient, who had orthopaedic surgery and suffered in the early post-operative period an idiopathic gastric partial necrosis. We reviewed the literature and it is an extraordinarily infrequent entity. Partial resections have already been described in case of limited necrosis, as reported in our case. Prompt diagnosis of idiopathic gastric necrosis and aggressive treatment are mandatory. Further awareness of this strange entity and its risk factors can lead to a quicker diagnosis and higher possibilities of survival for the patient.

Presence of risk Factors and Co-Factors for Acquiring HPV Infection or Cervical Cancer in Women Living in a rural Area and an Urban

Objective: Identify the presence of some risk factors and co-factors for acquiring HPV (human papilloma virus) or cervical cancer in women from a rural area and an urban area. material and methods: Cross-sectional study conducted in 2008 in the State of San Luis Potosí, méxico. Sampling of non-probabilistic convenience was applied. 99 women participated (73 from urban area and 26 rural). Two surveys were used to identify risk factors and socio-demographic data. All women underwent HPV testing;the samples were self-collected and processed by the Hybrid Capture II method?. To analyze the data, descriptive and inferential statistics were applied. results: The predominant age range was 37 to 42 years. 38.5% is in a rural community and an urban 37.3%. 72.6% began their sexual life at 19 years or m.re in urban areas and 57.6% before 18 in rural areas. In relation to the number of sexual partners reported having had only one, 73.1% and 67.1% in rural and urban area respectively. risk factors that were associated with HPV infection were the beginning of sexual activity at age 18 and under (p = 0.041) and the number of sexual partners (p = 0.001). Conclusions: There are m.re risk factors in women in urban areas but in rural areas it had the highest percentage of positive cases for infection by high-risk HPV.
