The 5-HT2c receptor gene Cys23Ser polymorphism influences the intravaginal ejaculation latency time in Dutch Caucasian men with lifelong premature ejaculation

It has been postulated that the persistent short intravaginal ejaculation latency time （IELT） of men with lifelong premature ejaculation （LPE） is related to 5-hydroxytryptamine （HT）2c receptor functioning. The aim of this study was to investigate the relationship of Cys23Ser 5-HT2c receptor gene polymorphism and the duration of IELT in men with LPE. Therefore, a prospective study was conducted in 64 Dutch Caucasian men with LPE. Baseline IELT during coitus was assessed by stopwatch over a 1-month period. All men were genotyped for Cys23Ser 5-HT2c receptor gene polymorphism. Allele frequencies and genotypes of Cys and Ser variants of 5-HT2c receptor gene polymorphism were determined. Association between Cys/Cys and Ser/Ser genotypes and the natural logarithm of the IELT in men with LPE ***. As a result, the geometric mean, median and natural mean IELT were 25.2, 27.0, 33.9s, respectively. Of all men, 20.0%, 10.8%, 23.1% and 41.5% ejaculated within 10, 10-20, 20-30 and 30-60s after vaginal penetration. Of the 64 men, the Cys/Cys and Ser/Ser genotype frequency for the Cys23Ser polymorphism of the 5-HT2c receptor gene was 81% and 19%, respectively. The geometric mean IELT of the wildtypes （Cys/Cys） is significantly lower （22.6s; 95% CI 18.3-27.8s） than in male homozygous mutants （Ser/Ser）（40.4s; 95% CI 20.3-80.4s）（P = 0.03）. It is concluded that Cys23Ser 5-HT2c receptor gene polymorphism is associated with the IELT in men with LPE. Men with Cys/Cys genotype have shorter IELTs than men with Ser/Ser genotypes.

We appreciate and thank Drs Saitz and Serefoglu for their positive and sympathetic words on our article.1 Indeed, our genetic studies have shown that 5-HTTLPR polymorphism, 5-HT1A receptor gene polymorphism and 5-HT2c receptor gene polymorphism are associated with the duration of the intravaginal ejaculatory latency time in men with lifelong premature ejaculation. In line with the remarks） we hope that our stopwatch method will be used by other investigators to investigate the role of serotonergic gene polymorphisms on the duration of the intravaginal ejaculatory latency time.

Sharing roosts but not ectoparasites:high host-specificity in bat flies and wing mites of Miniopterus schreibersii and Rhinolophus ferrumequinum(Mammalia:Chiroptera)

Schreiber’s bent-winged bat Miniopterus schreibersii and the greater horseshoe bat Rhinolophus ferrumequinum are widespread and common cavernicolous species across southern Europe that host numerous specialized ectoparasite *** objective of this study was to characterize the species assemblage,genetic diversity,and host specificity of bat flies(Nycteribiidae,Diptera)and wing mites(Spinturnicidae,Acari)found on these bat hosts in Serbia and Bosnia and ***,while bat flies lay puparia on the cave walls and can thus be transmitted indirectly,wing mites require direct body contact for *** identification and sequencing of a 710-bp fragment of cytochrome oxidase I gene of 207 bat flies yielded 4 species,3 on *** and 1 on *** of a 460-bp small subunit ribosomal RNA fragment,in all 190 collected wing mites revealed 2 species,1 per *** no case was a parasite associated with 1 host found on the other *** and genetic diversity of flies were higher in ***,likely reflecting their host’s larger colony sizes and migratory *** species of both hosts showed similarly low diversity,likely due to their faster life history and lower winter *** findings highlight a remarkably high host-specificity and segregation of ectoparasite species despite direct contact among their hosts in the roost,suggesting a defined host preference in the investigated ectoparasite ***,the differences in ectoparasite genetic diversity exemplify the interplay between host and parasite life histories in shaping parasite population genetic structure.

Melanoma of the rectum: A rare entity

A 41-year-old man presented with a 6-mo history of changed defecation and rectal bleeding. A 3-cm polypoid tumor of the lower rectum was found at rectosigmoidos- copy, which proved to be a leiomyosarcoma upon biopsy. Dissemination studies did not show any metastases. He was underwent to an abdomino-perineal resection (APR). Histopathology of the specimen showed a melanoma (S-100 stain positive). Two years after the resection, me- tastases in the abdomen and right lung were found. He died one and half years later. Primary anorectal melano- ma is a rare and very aggressive disorder. According to current data, one should always perform a S-100 stain when anorectal sarcoma is suspected. A positive S-100 stain suggests the tumour to be most likely a melanoma. Subsequently, thorough dissemination studies need to be performed. Depending on the outcome of the dissemina- tion studies, a surgical resection has to be performed. Nowadays, a sphincter-saving local excision combined with adjuvant loco-regional radiotherapy should be pre- ferred in case of small tumors. The same loco-regional control is achieved with less "loss of function" compared to non-sphincter saving surgery. Only in the case of large and obstructing tumors an abdomino-perineal resection is the treatment of choice.

GPU based techniques for deep image merging

Deep images store multiple fragments perpixel,each of which includes colour and depth,unlike traditional 2D flat images which store only a single colour value and possibly a depth ***,deep images have found use in an increasing number of applications,including ones using transparency and compositing.A step in compositing deep images requires merging per-pixel fragment lists in depth order;little work has so far been presented on fast *** paper explores GPU based merging of deep images using different memory layouts for fragment lists:linked lists,linearised arrays,and interleaved *** also report performance improvements using techniques which leverage GPU memory hierarchy by processing blocks of fragment data using fast registers,following similar techniques used to improve performance of transparency *** report results for compositing from two deep images or saving the resulting deep image before compositing,as well as for an iterated pairwise merge of multiple deep *** results show a 2 to 6 fold improvement by combining efficient memory layout with fast register based merging.

As an emerging approach to space situational awareness and space imaging,the practical use of an event-based camera(EBC)in space imaging for precise source analysis is still in its *** nature of event-based space imaging and data collection needs to be further explored to develop more effective event-based space imaging systems and advance the capabilities of event-based tracking systems with improved target measurement ***,for event measurements to be meaningful,a framework must be investigated for EBC calibration to project events from pixel array coordinates in the image plane to coordinates in a target resident space object’s reference *** this paper,the traditional techniques of conventional astronomy are reconsidered to properly utilise the EBC for space imaging and space situational *** paper presents the techniques and systems used for calibrating an EBC for reliable and accurate measurement *** techniques are vital in building event-based space imaging systems capable of real-world space situational awareness *** calibrating sources detected using the EBC,the spatiotemporal characteristics of detected sources or“event sources”can be related to the photometric characteristics of the underlying astrophysical ***,these characteristics are analysed to establish a foundation for principled processing and observing techniques which appropriately exploit the capabilities of the EBC.

Assessment of oxidative stress in chronic pancreatitis patients

AIM: To assess the levels of antioxidant capacity and oxidative damage in blood of chronic pancreatitis (CP) patients in comparison with those in healthy control sub- jects, by using several different analytical techniques. METHODS: Thirty-five CP patients and 35 healthy con- trol subjects were investigated prospectively with re- spect to plasma levels of thiols, ferric reducing ability of plasma (FRAP, i.e. antioxidant capacity), levels of protein carbonyls and thiobarbituric acid reactive substances (TBARS). Additionally, we evaluated the production of reactive oxygen species (ROS) in whole blood. RESULTS: The antioxidative thiols including cysteine, cysteinylglycine and glutathione were significantly lower in CP patients. In addition, the non-enzymatic antioxi- dant capacity was significantly lower in CP patients, which correlated with the amount of oxidative protein (protein carbonyls) and the extent of lipid damage (TBARS), both were significantly higher in CP patients. The ROS production in whole blood after stimulation with phorbol 12-myritate 13-acetaat, demonstrated a strong tendency to produce more ROS in CP patients. CONCLUSION: Oxidative stress may contribute to the pathogenesis of chronic pancreatitis by decreasing anti- oxidant capacity and increasing oxidative damage in CP patients may be a rationale for intervention with antioxi- dant therapy.

Defective recovery of QT dispersion following transcatheter aortic valve implantation： frequency, predictors and prognosis

Background Corrected QT dispersion （cQTD） has been correlated with non-uniform ventricular repolarisation and increased mortality. In patients with aortic stenosis, cQTD has been shown improved after surgical valve replacement, but the effects of transcatheter aortic valve implantation （TAVI） are unknown. Therefore, we sought to explore the frequency, predictors and prognostic effects of defective cQTD recovery at 6 months after TAVI. Methods A total of 222 patients underwent TAVI with the Medtronic-CoreValve System between November 2005 and January 2012. Patients who were on class Ⅰ or Ⅲ antiarrhythmics or on chronic haemodialysis or who developed atrial fibrillation, a new bundle branch block or became pacemaker dependent after TAVI were excluded. As a result, pre-, post- and follow-up ECG （median： 6 months） analysis was available in 45 eligible patients. Defective cQTD recovery was defined as any progression beyond the baseline cQTD at 6 months. Results In the 45 patients, the mean cQTD was 47 ± 23 ms at baseline, 45 ±17 ms immediately after TAVI and 40 ± 16 ms at 6 months （15% reduction, P = 0.049）. Compared to baseline, cQTD at 6 months was improved in 60% of the patients whereas defective cQTD recovery was present in 40%. cQTD increase immediately after TAVI was an independent predictor of defective cQTD recovery at 6 months （per 10 ms increase; OR： 1.89, 95% CI： 1.15-3.12）. By univariable analysis, defective cQTD recovery was associated with late mortality （HR： 1.52, 95% CI： 1.05-2.17）. Conclusions Despite a gradual reduction ofcQTD after TAVI, 40% of the patients had defective recovery at 6 months which was associated with late mortality. More detailed ECG analysis after TAVI may help to avoid late death.

Optimizing Growth Conditions for Digoxin Production in Digitalis lanata Ehrh

Objective: Digoxin is a therapeutic cardenolide widely used to treat various heart conditions such as atrial flutter, atrial fibrillation and heart failure in both Western as well as Chinese medicine. Digoxin is extracted from cultivated Digitalis lanata Ehrh. plants, known as Mao Hua Yang Di Huang in Chinese medicine. This manuscript presents two studies that were conducted to optimize the cultivation conditions for digoxin production in the TCM Mao Hua Yan Di Huang in a greenhouse under GAP ***: Two experiments were designed in which 4 growth conditions were compared. Levels of digoxin, gitoxin, digitoxin, α-acetyldigoxin, β-acetyl-digoxin were measured using HPLC-UV and compared between the ***: Normal soil, no CO_2 enrichment combined with a cold shock was found to be the optimal condition for producing digoxin in the first experiment. Gitoxin content was significantly lower in plants grown in this condition. Mechanical stress as well as the time of harvesting showed no statistically significant differences in the production of cardenolides. In the second experiment the optimal condition was found to be a combination of cold nights, sun screen, fertilizer use and no milled ***: This study shows that digoxin production can be increased by controlling the growth conditions of D. lanata Ehrh. The effect of cold was important in both experiments for improving digoxin production. Cultivating Chinese herbal medicines in optimized greenhouse conditions might be an economically attractive alternative to regular open air cultivation.
