Cyclosporine-Associated Nephrotoxicity

Cyclosporine (CsA) has revolutionized transplant medicine and is currently one of the most important immunosuppressive agents for a wide range of organ transplantations and of autoimmune and inflammatory diseases, such as rheumatoid arthritis, uveitis, psoriasis, and atopic dermatitis. Renal impairment represents the main limitation to CsA long-term continuous therapy. However, it has been shown that nephrotoxicity is associated with longer treatment duration, larger cumulative doses and higher daily dose of CsA. With low dose regimens (<5 mg/kg/day), stable serum creatinine levels have been observed up to 15-20 years after kidney transplantation. Intermittent therapy may offer a good therapeutic strategy to limit long-term renal dysfunction, given the fact that renal structural changes are dose- and time-dependent. The best predictor of permanent renal damage is a persistent increase in serum creatinine level one month after treatment withdrawal. In patients with autoimmune diseases, the percentage increase in serum creatinine above baseline value during CsA therapy has been shown to predict CsA-induced nephropathy. Before CsA therapy initiation, patients should undergo a thorough baseline evaluation including laboratory assessments, in particular electrolytes, serum creatinine, and urea levels. Furthermore, patients should be evaluated for factors that might increase the risk of nephrotoxicity, such as obesity, older age, hypertension, concomitant use of nephrotoxic drugs, and pre-existing renal conditions. In the present paper, CsA-induced nephropathy will be reviewed in terms of pathophysiology, pathologic and clinical findings, and strategies for prevention and management.

Comparison of Insecticide Resistance and Its Enzyme Mechanisms among Aedes aegypti Collected with Three Methods in a Dengue-Endemic City in Southern Mexico

Background: Despite the physical and chemical effort to control Aedes aegypti, the arboviruses transmission in the south of Mexico remains latent. Trying to improve the methods of entomological surveillance routinely used, whether the estimation of resistance to insecticides used for its control, as well as their enzyme mechanisms, were influenced by the phase in which the mosquitoes were collected through three different collection methods was investigated. Materials and Methods: Mosquito collections from the “5 de Febrero” neighborhood in Tapachula, Mexico were obtained by ovitraps, larvitraps, and a CDC backpack aspirator. Insecticide resistance of F1 females was determined by WHO diagnostic doses and resistance ratios (RR50), furthermore, levels of insecticide metabolism enzymes were determined by biochemical assays. Results: Overall, in mosquitoes collected by ovitraps, larvitraps, and CDC backpack aspirator respectively, the low mortalities obtained with the discriminant dose to Malathion (27.57%, 26.97%, and 26.91%), and to Bendiocarb (50.5%, 45.36%, and 54.97%) suggest resistance. However, LC50 for Malathion (0.922, 0.934, and 0.915) and for Bendiocarb (0.112, 0.109, and 0.107);and the low resistance ratios (RR50) for Malathion (3.34, 3.29, and 3.27) and for Bendiocarb (2.15, 2.1, and 2.06) does not suggest resistance. Although a slight numerical variation is observed between the three LC50 values, the overlap observed between their confidence intervals allows us to assume that there were no differences between the three methods. The study of specific differences between women and men is arousing huge interests in various fields of medicine, including dermatology. The available data on gender medicine applied to common skin diseases are unfortunately still scanty. Psoriasis is a chronic immunemediated skin disease which affects 1%-3% of most populations worldwide and can involve also the joints and entheses. The pathogenesis of the disease is very complex, resulting from the interaction between genetic predisposition and several environmental triggers. The pathogenic role of sex hormones has also been hypothesized. The analysis of gender-specific differences in psoriasis seems to suggest some interesting findings, such as an earlier age of disease onset in females, a higher probability of severe disease in men, or different tendencies in care utilization, adherence to treatment, development of psychological distress, and coping strategies. Moreover, sex-related differences have been recently described in some epidemiological and clinical features among patients with psoriatic arthritis. The objective of this article is to review briefly the available evidence regarding gender differences in various aspects of psoriasis, such as epidemiology, genetics, risk factors, associated conditions, quality of life, clinical and therapeutic aspects.

Liver Damage by the Interaction of Malathion with Cimetidine in Rat

In this study the authors aimed to evaluate the oxidative stress enzymes indicative of liver damage in rats exposed to malathion （M）, subchronic form using cimetidine （C） and cimetidine plus malathion （M ＋ C）. Malathion, widely used organophosphorus insecticide worldwide, induces oxidative liver damage type; cimetidine is an antagonist of histamine H2-receptor, it has been shown to be an inhibitor of various CYP45o isoforms. Male Wistar rats weighing 200-250 g were studied, exposed to malathion orally for 3 weeks （0.15 mg/kg/day, 2 mg/kg/day, 15 mg/kg/day） and cimetidine 10 mg/kg/day. Malathion plus cimetidine affect susceptibility to oxidative stress and possibly modifies the antioxidant defense capacity directly or indirectly.

Bacteriospermia among smallholder artificial insemination boars in the Philippines and potential associated factors

Objective:To determine the prevalence of bacteriospermia,the bacterial load,and the potential factors associated with bacterial contamination in boar semen collected by local smallholder artificial insemination ***:Fifteen individual raw semen samples were collected from locally available artificial insemination boars owned by different smallholder boar operators within the 5th district of Leyte,Philippines and were subjected to standard bacteriological culture and identification,including a survey of potentially associated *** and bacterial count were determined accordingly,while boar characteristics and collection practices were clustered following agglomerative hierarchical clustering ***:One hundred percent contamination with a bacterial count of(2.01±0.38)×10^(3) CFU/mL was *** least 73.33%of the samples were positive for Bacillus spp.,while other identified isolates included Enterobacter spp.,Staphylococcus spp.,***,Pseudomonas spp.,Citrobacter spp.,and Klebsiella ***:Despite the high prevalence of bacteriospermia,the bacterial count is ***,on-farm practices on boar health and management,semen collection,and sanitation as well as the enhancement of basic protocols to control contamination should be conscientiously considered in smallholder artificial insemination operation.

Correlation between Iron Reducibility in Natural and Iron-Modified Clays and Its Adsorptive Capability for Arsenic Removal

The study reports aspects that allowed to correlate structural and redox properties of iron species deposited on clay minerals with the capacity of geomaterials for arsenic removal. Natural ferruginous clays as well as an iron-poor clay chemically modified with Fe(III) salt (ferrihydrite species) were investigated as adsorbents of the arsenate(V) in water. The study, carried out from minerals from abundant Argentinean deposits, was conducted with the aid of different techniques such as X-ray Diffraction (XRD), Scanning Electron Microscopy (SEM-EDS), Raman Spectroscopy, ICP-AES (Inductively Coupled Plasma) chemical analysis and Temperature Programmed Reduction (TPR). This last technique allowed to detect availability of iron species in oxidic environment with different structural complexity and to determine active sites, accessible for arsenate(V) adsorption. The effect was observed through temperature dependence of the first Fe(III) reduction step (below 570°C) of iron-oxide species. The sequence of reducibility: ferrihydrite > hydrous oxide (goethite) > anhydrous oxide (hematite) > structural iron in clay was in agreement with the availability of iron active sites for the reducing process as well as for the arsenate adsorption. The important role of very high iron content in original samples was also observed. The chemical activation of iron-poor clay by a simple and feasible modification with Fe(III) solutions promoted the deposition of the ferrihydrite active phase with an increase of 2.81% (expressed as Fe2O3) respect to the original content of 1.07%, constituting an accessible and eco-friendly technological alternative to solve the environmental problem of water containing arsenic.

Influence of Temperature on Calcium Hydroxyapatite Nanopowders

The aim of this work is to investigate the effects of thermal treatment, in the range of temperature between 80oC - 900oC, on crystallinity, morphology and particles size of calcium hydroxylapatite nanopowders (HAp). A complete study was carried out applying 31P Magic Angle Spinning NMR, X-ray diffraction, nitrogen porosimetry and Transmission Electron Microscopy techniques. HAp specimens were prepared by chemical precipitation in an aqueous solution of calcium nitrate and ammonium hydrogen phosphate. The thermal treatment performed, showed the formation of crystals whose appearance has three main morphologies and different particles size. HAp treated up to 500oC showed a set of needle-like shape crystals with a low surface area value. These crystals turned into rod-like crystals with temperature increase, along with an increase in crystallite size. At 900oC was detected a hexagonal hydroxylapatite phase. Furthermore, Dynamic Light Scattering explored a stable HAp dispersion. The isopropyl alcohol dispersion resulted stable for at least two weeks, useful to create layers of determined morphology nanoparticles.
