The elevated presence of metal(loid)s in the environment significantly impacts ecosystems and human health and is generally largely due to industrial and mining ***,in the current study,we investigated and proposed an environmentally friendly method(phytomanagement)aimed at reducing the negative impacts associated with metal(loid)pollution through the use of soil amendments(biochar and compost)to permit Ailanthus altissima growth on a highly contaminated mining Technosol,with arsenic(As)and lead(Pb)contents of 539.06 and 11453 mg kg^(-1),*** objective was to examine the impacts of three biochars and compost on i)the physicochemical characteristics of soil,ii)metal(loid)immobilization in soil,and iii)*** *** revealed that the application of biochar as a soil amendment improved soil conditions by increasing soil electrical conductivity,pH,and water-holding ***,concomitantly,we observed a large reduction(99%)in Pb mobility and availability following application of the hardwood biochar in combination with compost(HBCP).Thus,this combined soil amendment was most effective in promoting *** *** addition,the HBCP treatment prevented As translocation in the upper parts of plants,although soil pore water As concentration was not diminished by amendment application.

Sharing sensitive data is a specific challenge for research infrastructures in the field of life sciences. For that reason a toolbox has been developed, providing resources for researchers who wish to share and use sensitive data, to support the workflows for handling these kinds of digital objects. Common and community approved annotations are required to be compliant with FAIR principles(Findability, Accessibility,Interoperability, Reusability). The toolbox makes use of a tagging(categorisation) system, allowing consistent labelling and categorisation of digital objects, in terms relevant to data sharing tasks and activities. A pilot study was performed within the Horizon 2020 project EOSC-Life, in which 2 experts from 6 life sciences research infrastructures were recruited to independently assign tags to the same set of 10 to 25 resources related to sensitive data management and data sharing(in total 110). Summary statistics of agreement and observer variation per research infrastructure are provided. The pilot study has shown that experts were able to attribute tags but in most cases with a considerable observer variation between experts. In the context of CWFR(Canonical Workflow Frameworks for Research), this indicates the necessity for careful definition,evaluation and validation of parameters and processes related to workflow descriptions. The results from this pilot study were used to tackle this issue by revising the categorisation system and providing an updated version.

Cardiac resynchronization therapy(CRT)has been shown to improve symptoms,left ventricle(LV)performance and long-term survival in patients with heart failure with re-duced ejection fraction(HFrEF)and delayed ventricular activation.[1]CRT is currently recom-mended in symptomatic patients with LV ejection fraction of≤35%,and ventricular conduction dis-order manifested as left bundle branch block(LBBB)>149 ms,and should be considered in LBBB>129 ms or non-LBBB QRS duration>150 ms.

Precision medicine is defined by the administration of drugs based on the tumor's particular genetic characteristics. It is developing quickly in the field of cancer therapy. For example, KRAS, NRAS and BRAF genetic testing demonstrates its efficiency for precision medicine in colorectal cancer(CRC). Besides for these well-known mutations, the purpose of performing larger genetic testing in this pathology is unknown. Recent reports have shown that using the poly ADP ribose polymerase(PARP) inhibitor olaparib in patients with homologous repair enzyme deficiency gave positive clinical results in breast, ovarian and prostate cancers. We have reported here the cases of 2 patients with multi-treated metastatic CRC who underwent somatic and constitutional exome analyses. The analyses revealed a loss of function mutation in a homologous repair enzyme resulting in the loss of heterozygosity for both patients(Check2 for the first patient and RAD51 C for the second one). Both patients were treated with off-label usage of olaparib. While the first patient showed clinical benefit, reduction of carcinoembryonic antigen tumor marker and radiologic response, the second patient quickly presented a progression of the tumor. Additional genetic analyses revealed a frameshift truncating mutation of the TP53BP1 gene in the patient who progressed. Interestingly, deficiency in TP53BP1 was previously described to confer resistance to olaparib in mice breast cancer models. Our findings suggest that exome analysis may be a helpful tool to highlight targetable mutations in CRC and that olaparib may be efficient in patients with a homologous repair deficiency.

AIM: To investigate cellular 5-HT4(-h/+h) receptor distribution, particularly in the epithelial layer, by laser mi-crodissection and polymerase chain reaction (PCR) in porcine gastrointestinal (GI) tissues. METHODS: A stepwise approach was used to evaluate RNA quality and to study cell-specific 5-HT4 receptor mRNA expression in the porcine gastric fundus and colon descendens. After freezing, staining and laser microdissection and pressure catapulting (LMPC), RNA quality was evaluated by the Experion automated electrophoresis system. 5-HT4 receptor and glyceral-dehyde-3-phosphate dehydrogenase (GAPDH) expressions were examined by endpoint reverse transcription (RT)-PCR in mucosal and muscle-myenteric plexus (MMP) tissue fractions, in mucosal and MMP parts of hematoxylin and eosin (HE) stained tissue sections andin microdissected patches of the epithelial and circular smooth muscle cell layer in these sections. Pig gastric fundus tissue sections were also stained immunohisto-chemically (IHC) for enterochromaffin cells (EC cells; MAB352); these cells were isolated by LMPC and examined by endpoint RT-PCR. RESULTS: After HE staining, the epithelial and circular smooth muscle cell layer of pig colon descendens and the epithelial cell layer of gastric fundus were identified morphologically and isolated by LMPC. EC cells of pig gastric fundus were successfully stained by IHC and isolated by LMPC. Freezing, HE and IHC staining, and LMPC had no influence on RNA quality. 5-HT4 recep-tor and GAPDH mRNA expressions were detected in mucosa and MMP tissue fractions, and in mucosal and MMP parts of HE stained tissue sections of pig colon descendens and gastric fundus. In the mucosa tissue fractions of both GI regions, the expression of h-exon containing receptor [5-HT4(+h) receptor] mRNA was significantly higher (P<0.01) compared to 5-HT4(-h) re-ceptor expression, and a similar trend was obtained in the mucosal part of HE stained tissue sections. Quantifying Forest Loss in the Mbalmayo Forest Reserve (Center Region, Cameroon)

This study was carried out in the forest reserve of Mbalmayo, Center Region of Cameroon, with the aim to analyze the dynamics of land use/land cover (LULC) changes over the past 30 years. The databases used were made up of 3 Landsat satellite images (5TM of 1990, 7ETM+ of 2005 and 8OLI of 2020). The satellite images were processed using ArcGIS and Erdas Imagine software. Ethnobotanical surveys were conducted to investigate the relationships between the local population and the forest reserve. The Mbalmayo forest reserve is consisted of seven main land cover namely: mature secondary forest, young secondary forest, savanna, fallows and crops, built, bare soil and water surface. The dynamics of the forest cover undergoes more significant changes between the years 1990-2005 with losses estimate at 4762 ha compared to the years 2005-2020 (2231 ha), with a marked decrease in the area of dense forests. This regression is much more important in dense forest vegetation. The forest cover lost 6993 ha (26.92%) of its surface, which corresponds to an average rate of deforestation of 233.1 ha/year. The survey revealed the need for NTFPs for the Mbalmayo reserve forest populations. Indeed, all respondents agreed that they use non-timber forest products from the forest, mainly for medication (37%), wood (34%) and food (24%). Therefore, it becomes urgent to redefine a management plan for the Mbalmayo forest reserve which will define how the forest must be managed in order to avoid the loss of large areas of forests disappearing each year under the human action, which exerts pressure on forest species, thus leading to their possible disappearance.

Modelling of massive particulates for breakwater engineering using coupled FEMDEM and CFD

The seaward slope of many breakwaters consists of thousands of interlocking units of rock or concrete comprising a massive granular system of large elements each weighing tens of tonnes. The dumped quarry materials in the core are protected by progressively coarser particulates. The outer armour layer of freely placed units is intended to both dissipate wave energy and remain structurally stable as strong flows are drawn in and out of the particulate core. Design guidance on the mass and shape of these units is based on empirical equations derived from scaled physical model tests. The main failure mode for armour layers exposed to severe storms is hydraulic instability where the armour units of concrete or rock are subjected to uplift and drag forces which can in turn lead to rocking, displacement and collisions sufficient to cause breakage of units. Recently invented armour unit designs making up such granular layered system owe much of their success to the desirable emergent properties of interlock and porosity and how these combine with individual unit structural strength and inertial mass. Fundamental understanding of the forces governing such wave-structure interaction remains poor. We use discrete element and combined finite-discrete element methods to model the granular solid skeleton of randomly packed units coupled to a CFD code which resolves the wave dynamics through an interface tracking technique. The CFD code exploits several methods including a compressive advection scheme, node movement, and general mesh optimization. We provide the engineering context and report progress towards the numerical modelling of instability in these massive granular systems.

Calibration and Validation of the GR2M Hydrologic Model in the Kouilou-Niari Basin in Southwestern Congo-Brazzaville

The hydrologic simulation of a catchment area, described as the transformation of rainfall into runoff, generally uses hydrologic model. This work opts for the global conceptual hydrologic model GR2M, a monthly time step model, to study the Kouilou-Niari basin, the second most important ones of the Republic of Congo. This includes two parameters to model the hydrologic behavior of a catchment area. The choice of the conceptual model GR2M is justified by the reduced number of parameters and the monthly time scale. The objective of this study is to determine the characteristic parameters of the GR2M model, by a calibrating and a validating procedure. The use of these parameters enables to follow the evolution of the water resources from the climatic variables. It has been first carried out a characterization of some physical, geological and climatic factors governing the flow, by dealing with the main climatic variables which constitute the inputs of the hydrologic model. Then, a hydrologic rainfall-runoff modeling allows to calibrate and validate the model at monthly time scale. Taking into account the number of parameters involved in hydrologic processes and the complexity of the cathment area, this model gives acceptable results throughout the Kouilou-Niari basin. The values of the Nash-Sutcliffe criterion and those of the correlation coefficient obtained are greater than 80% in validation, which explains the performance and robustness of the GR2M model on this basin.
