Combination immunotherapy with Survivin and luteinizing hormone-releasing hormone fusion protein in murine breast cancer model

AIM To investigate the therapeutic potential of two recombinant proteins, Survivin and luteinizing hormone-releasing hormone (LHRH) fusion protein [LHRH(6 leu)-LTB] for immunotherapy of breast *** Murine 4 T-1 breast cancer model was used to evaluate the efficacy of recombinant proteins in vivo. Twenty four Balb/c mice were divided into 4 groups of 6 mice each. Recombinant Survivin and LHRH fusion protein, alone or in combination, were administered along with immunomodulator Mycobacterium indicus pranii (MIP) in Balb/c mice. Unimmunized or control group mice were administered with phosphate buffer saline. Each group was then challenged with syngeneic 4 T-1 cells to induce the growth of breast tumor. Tumor growth was monitored to evaluate the efficacy of immune-response in preventing the growth of cancer *** Preventive immunization with 20 μg recombinant Survivin and MIP was effective in suppressing growth of 4 T-1 mouse model of breast cancer (P = 0.04) but 50 μg dose was ineffective in suppressing tumor growth. However, combination of Survivin and LHRH fusion protein was more effective in suppressing tumor growth (P = 0.02) as well as metastasis in vivo in comparison to LHRH fusion protein as vaccine antigen *** Recombinant Survivin and MIP suppress tumor growth significantly. Combining LHRH fusion protein with Survivin and MIP enhances tumor suppressive effects marginally which provides evidence for recombinant Survivin and LHRH fusion protein as candidates for translating the combination cancer immunotherapy approaches.

Ankle Fractures: A Literature Review of Current Treatment Methods

Ankle fracture is one of the most common lower limb fractures for they account for 9% of all fractures representing a significant portion of the trauma workload. Ankle fractures usually affect young men and older women, however, below the age of 50;ankle fractures are the commonest in men. Two commonly used classification systems for ankle fractures include the danis weber AO classification and the Lauge-Hansen classification. There is biomechanical evidence that posterior non-locking plates are superior in stability than laterally placed plates;however there is little clinical evidence. There are several different methods of ankle fracture fixation, however the goal of treatment remains a stable anatomic reduction of talus in the ankle mortise and correction of the fibula length as a 1 mm lateral shift of the talus in the ankle mortise reduces the contact area by 42%, and displacement (or shortening) of the fibula more than 2 mm will lead to significant increases in joint contact pressures. Further research both biomechanically and clinically needs to be undertaken in order to clarify a preferable choice of fixation.

The antitumor capabilities of agonistic anti-4-1BB mAbs have made them an attractive target for tumor ***,the adverse side effects associated with agonist antibodies have hindered their clinical ***,we aimed to study the immune-related adverse events of repeated doses and long-term use of agonistic anti-4-1BB *** show that chronic activation of 4-1BB signals induced the accumulation of IFN-γ-producing PD-1^(+)CD8^(+)T cells in the secondary lymphoid organs of tumor-bearing mice by increasing the number of dividing CD8^(+)T cells,which was beneficial for suppressing tumor growth in the early phase of anti-4-1BB ***,repeated exposure to anti-4-1BB mAbs led to granuloma development in tumor-draining lymph nodes(TDLNs)of mice due to recruitment and accumulation of macrophages via the CD8^(+)T cell-IFN-γ*** was accompanied by excessive lymph node swelling,which impaired the sequential activation of CD8^(+)T *** data provide insights into the immune-related adverse events of long-term agonist 4-1BB antibody dosing,which should be considered during the clinical development of immunomodulating therapy.

Right vocal cord paralysis in our present case was diagnosed on clinical and radiological examination which is precipitated by an anomalous right aortic arch with diverticulum. This is a very uncommon vascular etiology of hoarseness and is extremely rare. Because of this rarity, the practicing otolaryngologist may miss this finding while evaluating a case of idiopathic right vocal cord paralysis. Thus, the authors feel that idiopathic or unexplained right vocal cord paralysis should be routinely investigated with a CT or MRI of neck and chest with or without contrast to avoid such shortcomings. There is only one such case of right vocal cord paralysis by right aortic which has been reported earlier in literature.

A Novel Hybrid Tunicate Swarm Naked Mole-Rat Algorithm for Image Segmentation and Numerical Optimization

This paper provides a new optimization algorithm named as tunicate swarm naked mole-rat algorithm(TSNMRA)which uses hybridization concept of tunicate swarm algorithm(TSA)and naked mole-rat algorithm(NMRA).This newly developed algorithm uses the characteristics of both algorithms(TSA and NMRA)and enhance the exploration abilities of *** from the hybridization concept,important parameter of NMRA such as mating factor is made to be self-adaptive with the help of simulated annealing(sa)mutation operator and there is no need to define its value *** evaluating the working capabilities of proposed TSNMRA,it is tested for 100-digit challenge(CEC 2019)test problems and real multi-level image segmentation *** the results obtained for CEC 2019 test problems,it can be seen that proposed TSNMRA performs well as compared to original TSA and *** case of image segmentation problem,comparison of TSNMRA is performed with multi-threshold electro magnetism-like optimization(MTEMO),particle swarm optimization(PSO),genetic algorithm(GA),bacterial foraging(BF)and found superior results for TSNMRA.

Fermented functional food products are among the major segments of food processing *** imparts several characteristic effects on foods including the enhancement of organoleptic characteristics,increased shelf-life,and production of novel health beneficial ***,in addition to macronutrients present in the food,secondary metabolites such as polyphenols are also emerging as suitable fermentable *** the traditional antimicrobial view of polyphenols,accumulating research shows that polyphenols exert differential effects on bacterial communities by suppressing the growth of pathogenic microbes while concomitantly promoting the proliferation and survival of probiotic ***,probiotic bacteria not only survive among polyphenols but also induce their fermentation which often leads to improved bioavailability of polyphenols,production of novel metabolic intermediates,increased polyphenolic content,and thus enhanced functional capacity of the fermented *** addition,selective fermentation of combinations of polyphenol-rich foods or fortification with polyphenols can result in novel functional *** present narrative review specifically explores the potential of polyphenols as fermentable substrates in functional *** discuss the emerging bidirectional relationship between polyphenols and probiotic bacteria with an aim at promoting the development of novel functional foods based on the amalgamation of probiotic bacteria and polyphenols.

As far as the present state is concerned in detecting the behavioral pattern of humans(subject)using morphological image processing,a considerable portion of the study has been conducted utilizing frontal vision data of human *** present research work had used a side vision of human-face data to develop a theoretical framework via a hybrid analytical model *** this example,hybridization includes an artificial neural network(ANN)with a genetic algorithm(GA).We researched the geometrical properties extracted from side-vision human-face *** additional study was conducted to determine the ideal number of geometrical characteristics to pick while *** close vicinity ofminimum distance measurements is done for these clusters,mapped for proper classification and decision process of behavioral *** identify the data acquired,support vector machines and artificial neural networks are utilized.A method known as an adaptiveunidirectional associative memory(AUTAM)was used to map one side of a human face to the other side of the same *** behavioral pattern has been detected based on two-class problem classification,and the decision process has been done using a genetic algorithm with best-fit *** developed algorithm in the present work has been tested by considering a dataset of 100 subjects and tested using standard databases like FERET,Multi-PIE,Yale Face database,RTR,CASIA,*** complexity measures have also been calculated under worst-case and best-case situations.
