Transcriptional responses of Brassica nigra to feeding by specialist insects of different feeding guilds

Plants show phenotypic changes when challenged with herbivorous insects. The mechanisms underlying these changes include the activation of transcriptional responses, which are dependent on the attacking insect. Most transcriptomic studies on crucifer-insect interactions have focused on the model plant Arabidopsis thaliana, a species that faces low herbivore pressure in nature. Here, we study the transcriptional responses of plants from a wild black mustard （Brassica nigra） population to herbivores of different feeding guilds using an A. thaliana-based whole-genome microarray that has previously been shown to be suitable for transcriptomic analyses in Brassica. Transcriptional responses of B. nigra after infestation with either Pieris rapae caterpillars or Brevicoryne brassicae aphids are analyzed and compared. Additionally, the insect-induced expression changes of some individual genes are analyzed through quantitative real-time polymerase chain reaction. The results show that feeding by both insect species results in the accumulation of transcripts encoding proteins involved in the detoxification of reactive oxygen species, defensive proteins and glucosinolates and this is correlated with experimental evidence in the literature on such biochemical effects. Although genes encoding proteins involved in similar processes are regulated by both insects, there was little overlap in the induction or repression of individual genes. Furthermore, P. rapae and B. brassicae seem to affect different phytohormone signaling pathways. In conclusion, our results indicate that B. nigra activates several defense-related genes in response to P rapae or B. brassicae feeding, but that the response is dependent on the attacking insect species.

Myzus persicae has severe economic impact on pepper(Capsicum)***,we identified two populations of ***,NL and SW,that were avirulent and virulent,respectively on *** accession *** transcriptomics approach used in the current study,which is the first study to explore the pepper−aphid interaction at the whole genome gene expression level,revealed genes whose expression is differentially regulated in pepper accession PB2013071 upon infestation with these *** *** NL population induced ROS production genes,while the SW population induced ROS scavenging genes and repressed ROS production *** also found that the SW population can induce the removal of ROS which accumulated in response to preinfestion with the NL population,and that preinfestation with the SW population significantly improved the performance of the NL *** paper supports the hypothesis that *** can overcome the resistance in accession PB2013071 probably because of its ability to manipulate plant defense response especially the ROS metabolism and such ability may benefit avirulent conspecific aphids.

Dense genetic maps create a base for QTL analysis of important traits and future implementation of marker-assisted *** tetraploid rose,the existing linkage maps includeers to cover 28 linkage groups(4 homologous sets of 7 chromosomes).Here we used the 68k WagRhSNP Axiom single-nucleotide polymorphism(SNP)array for rose,in combination with SNP dosage calling at the tetraploid level,to genotype offspring from the garden rose cultivar‘Red New Dawn’.The offspring proved to be not from a single bi-parental *** rose breeding,crosses with unintended parents occur *** developed a strategy to separate progeny into putative populations,even while one of the parents was unknown,using principle component analysis on pairwise genetic distances based on sets of selected SNP markers that were homozygous,and therefore uninformative for one *** of the inferred populations was consistent with self-fertilization of‘Red New Dawn’.Subsequently,linkage maps were generated for a bi-parental and a self-pollinated population with‘Red New Dawn’as the common maternal *** densest map,for the selfed parent,had 1929 SNP markers on 25 linkage groups,covering 1765.5 cM at an average marker distance of 0.9 *** with the strawberry(Fragaria vesca)genome was *** ICM1 corresponded to *** pseudochromosome 7(Fv7),ICM4 to Fv4,ICM5 to Fv3,ICM6 to Fv2 and ICM7 to *** ICM2 corresponded to parts of *** pseudochromosomes 1 and 6,whereas ICM3 is syntenic to the remainder of Fv6.

Quantitative trait loci(QTL)mapping approaches rely on the correct ordering of molecular markers along the chromosomes,which can be obtained from genetic linkage maps or a reference genome *** apple(Malus domestica Borkh),the genome sequence v1 and v2 could not meet this need;therefore,a novel approach was devised to develop a dense genetic linkage map,providing the most reliable marker-loci order for the highest possible number of *** approach was based on four strategies:(i)the use of multiple full-sib families,(ii)the reduction of missing information through the use of HaploBlocks and alternative calling procedures for single-nucleotide polymorphism(SNP)markers,(iii)the construction of a single backcross-type data set including all families,and(iv)a two-step map generation procedure based on the sequential inclusion of *** map comprises 15417 SNP markers,clustered in 3 K HaploBlock markers spanning 1267 cM,with an average distance between adjacent markers of 0.37 cM and a maximum distance of 3.29 ***,chromosome 5 was oriented according to its homoeologous chromosome *** map was useful to improve the apple genome sequence,design the Axiom Apple 480 K SNP array and perform multifamily-based QTL *** collinearity with the genome sequences v1 and v3 are *** our knowledge,this is the shortest published SNP map in apple,while including the largest number of markers,families and *** result validates our methodology,proving its value for the construction of integrated linkage maps for any outbreeding species.

利用发根土壤杆菌(Agrobacterium rhizogenes)在甘蓝、花椰菜、抱子甘蓝和油菜的下胚轴切段上进行遗传转化。在这些植物基因型中,花椰菜的转化培养获得了最高的根诱导频率,而甘蓝的转化培养得到了最高的转化频率。同菌株LBA9365和LBA8490相比,发根土壤杆菌LBA9402诱导的根数较多。沿着下胚轴依次截取切段进行转化培养,结果发现从子叶下切段到根系上面的切段,其根诱导频率呈阶梯状上升趋势。在切段上的单根转移培养之前,截取下胚轴切段(此切段仍保留有所有的根)上1～2mm长的顶端部分,转移到固体培养基上培养,结果大大提高了转化频率。

The prevalence of type 2 diabetes mellitus(T2DM)is increasing rapidly *** of the limited success of generic interventions,the focus of the disease study has shifted toward personalized strategies,particularly in the early stages of the *** Chinese medicine(TCM)is based on a systems view combined with personalized strategies and has improved our knowledge of personalized *** a systems biology perspective,the understanding of personalized diagnostics can be improved to yield a biochemical basis for such strategies;for example,metabolomics can be used in combination with other system-based diagnostic methods such as ultra-weak photon emission(UPe).In this study,we investigated the feasibility of using plasma metabolomics obtained from 44 pre-T2DM subjects to stratify the following TCM-based subtypes:Qi-Yin deficiency,Qi-Yin deficiency with dampness,and Qi-Yin deficiency with *** studied the relationship between plasma metabolomics and UPe with respect to TCM-based subtyping in order to obtain biochemical information for further interpreting disease *** component analysis of plasma metabolites revealed differences among the TCM-based pre-T2DM *** high levels of lipids(e.g.,cholesterol esters and triglycerides)were important discriminators of two of the three subtypes and may be associated with a higher risk of cardiovascular *** metabolomics data indicate that the lipid profile is an essential component captured by UPe with respect to stratifying subtypes of *** results suggest that metabolic differences exist among different TCM-based subtypes of pre-T2DM,and profiling plasma metabolites can be used to discriminate among these *** metabolomics thus provides biochemical insights into system-based UPe measurements.
