Tumor-induced neurogenesis and immune evasion as targets of innovative anti-cancer therapies

Normal cells are hijacked by cancer cells forming together heterogeneous tumor masses immersed in aberrant communication circuits that facilitate tumor growth and *** the well characterized angiogenic effect of some tumor-derived factors;others,such as BDNF,recruit peripheral nerves and *** neurogenic switch,activated by tumor-derived neurotrophins and extracellular vesicles,attracts adjacent peripheral fibers(autonomic/sensorial)and neural progenitor ***,tumor-associated nerve fibers can guide cancer cell ***,IL-1β,CCL2,PGE2,among other chemotactic factors,attract natural immunosuppressive cells,including T regulatory(Tregs),myeloid-derived suppressor cells(MDSCs),and M2 macrophages,to the tumor *** leukocytes further exacerbate the aberrant communication circuit releasing factors with neurogenic ***,cancer cells directly evade immune surveillance and the antitumoral actions of natural killer cells by activating immunosuppressive mechanisms elicited by heterophilic complexes,joining cancer and immune cells,formed by PD-L1/PD1 and CD80/CTLA-4 plasma membrane ***,nervous and immune cells,together with fibroblasts,endothelial,and bone-marrow-derived cells,promote tumor growth and enhance the metastatic properties of cancer *** by the demonstrated,but restricted,power of anti-angiogenic and immune cell-based therapies,preclinical studies are focusing on strategies aimed to inhibit tumor-induced *** we discuss the potential of anti-neurogenesis and,considering the interplay between nervous and immune systems,we also focus on anti-immunosuppression-based *** molecules,antibodies and immune cells are being considered as therapeutic agents,aimed to prevent cancer cell communication with neurons and leukocytes,targeting chemotactic and neurotransmitter signaling pathways linked to perineural invasion and metastasis.

The selective retinoic acid receptor-α agonist AM580 fails to control autoimmune neuroinflammation

Environmental cues provided by products from the diet,gut microbiota metabolism,and exposure to sunlight are sensed by specific receptors that drive a specific genomic profile in immune cells and influence the outcome of multiple sclerosis(MS),an autoimmune disease of the central nervous system(CNS),1 which affects approximately 2.3 million people worldwide.2 Among these receptors,the ligand-activated transcription factor retinoic acid receptorα(RAR-α)senses retinoic acid and other ligands to promote the transcription of genes that have retinoic acid receptor elements in their promoter region.3 Supplementation with all-trans retinoic acid(ATRA),a metabolite of vitamin A,ameliorates inflammation in MS and the corresponding animal model,experimental autoimmune encephalomyelitis(EAE),by shifting the balance between Th17/regulatory T cells(Tregs)and inducing tolerogenic dendritic cells.4,5 To overcome the weaknesses associated with ATRA agonists,e.g.,instability,poor bioavailability and nonselective binding to a broad range of retinoid receptors,a variety of synthetic agonists specifically targeting RAR have been developed.6 One of them,AM80,inhibits Th17 cells and suppress acute neuroinflammation;however,continuous AM80 treatment is ineffective,most likely because AM80 also inhibits Tregs and IL-10 production.6 AM580,a stable benzoic derivative of retinoic acid and a selective RAR-αagonist,has been recently shown to significantly reduce the production of Th1 cytokines but promote Th2 cytokines in human PBMCs7 and to inhibit microglial activation,thus acting beneficially in Alzheimer’s disease treatment.8 In addition,AM580 protects retinal cells against diabetes-induced apoptosis by inducing neurotrophic factors.9 However,whether it has a protective effect on neuroinflammation is unknown.

New technologies as a strategy to decrease medication errors: how do they affect adults and children differently?

Background:Medication error can occur throughout the drug treatment process,with special relevance in children given the risk of adverse effects resulting from a medication error is more prevalent than in *** significance of medication error in children is also greater because small error that would be tolerated in adults can cause significant damage in ***,the likelihood of injury is higher than in *** sources:Based on the data published,most medication errors take place in prescribing and administration stages in both *** in account that child's risk factors are different from those of adults,with some specific causes to pediatrics,we have reviewed available data about new technologies as a strategy to reduce pediatric medication ***:Even though there is a lack of standardized definitions and terminology that makes studies difficult to compare,we checked that new technologies have proven to be effectives in reducing medication errors,mainly computerized physician order entry(CPOE)and platforms to aid ***,we also observed that the use of these informatic tools can also generate new ***:Implementation of CPOE programs for pediatrics,communication improvement between healthcare professionals taking care of admitted children and the knowledge of these programs should be the mayor priorities for the safety of hospitalized children.

Induction of apoptosis on human hepatocarcinoma cell lines by an alkyl resorcinol isolated from Lithraea molleoides

AIM: To study the mechanism of cytotoxicity of a new active 5-alkyl resorcinol [1, 3-dihydroxy-5- (tridec-4’, 7’ -dienyl) benzene] isolated from Lithraea molleoides leaves on liver tumor cells. METHODS: Human hepatocarcinoma cell lines (HepG2 and Hep3B) in culture were treated with inhibitory concentrations, 50% of the compound, for 24 h. The induction of apoptosis was detected in treated cells by analysis of DNA fragmentation, DNA content, and acridine orange and propidium iodide staining. RESULTS: After 24 h of 5-alkyl resorcinol treatment, both cell lines showed: (1) the typical morphological alterations of apoptosis; (2) DNA fragmentation, detected by laddering and appearance of a subG0 population by flow cytometry; and (3) condensed and fragmented nuclei by acridine orange-propidium iodide staining. CONCLUSION: Based on the results, this compound exerts its cytotoxic effect in both hepatocellular cell lines through apoptotic cell death. For Hep3B, cells with mutated p53 and Fas, apoptosis would proceed by p53- or Fas-independent pathways.

Bucaramanga, with a population of about one million, is the capital of the Department of Santander （Colombia）, and has the Rio Surata as its main source of drinking water. In the upper reaches of the river, small-scale gold mining has been going onfor many centuries （Vetas-California mining district）. Geologically, the Vetas-California mining district is part of the Santander Massif which is composed of pre-Devonian metamorphic rocks and Mesozoic igneous rocks. The mineralisation is restricted to structurally controlled veins and is often associated with hydrothermal alteration. There is also disseminated mineralisation, but presently only the vein-type is mined. The processing residues are discharged into the nearby watercourses, contaminating the waters of the Surata river basin. As part of a joint Colombian-German technical cooperation project, with the purpose of controlling the negative environmental effects of gold mining pollution, an integrated distillation system for the recovery of mercuury. has been designed and tested.

The construction of the concept of Latin America has been through different processes regarding its structure and acceptance,from the colonial period to the automatic realignment to the United States,with rare exceptions,and a reorganization with the emergence of multilateralism and the multipolar *** abstraction,hitherto almost an axiom in the subcontinent,that it is a western region,leads us to the elaboration of this article,which seeks to present Latin America's post-western and de-westernized character in an environment of restructuration of its political economy,its actors,scenarios and,finally,***,we present here a review of the state of the art political economy of Latin America and its perspectives in this shifting world,especially in the context of the COVID-19 pandemic.

Compression Behaviors of Parallel Bamboo Strand Lumber Under Static Loading

In order to investigate the influence of length and compression directions upon behaviour of parallel bamboo strand lumber(PBSL)specimens,240 axial compression tests have been *** three similar one different typical failure modes,the mechanical performance for PBSL specimens under compression parallel to grain and perpendicular to grain are different as a *** the point of the characteristic values,the compression strength parallel to grain is 2.1 times of the compression strength perpendicular to *** elastic modulus for compression parallel to grain is 3.64 times of the compression strength perpendicular to *** the compression ratios along two compression directions are equal to each *** bigger Poisson ratios for one typical side surface is 3.93 times of that for another typical side surface for PBSL specimens under compression perpendicular to grain,and the bigger value is equal to that for PBSL specimens under compression parallel to *** can influence the mechanical properties of the PBSL *** size 50 mm×50 mm×100 mm should be good choice for the standard or code to measure the compression *** materials have better ductility under compression parallel to grain than that under compression perpendicular to ***-strain relationship models were proposed for PBSL specimens under compression parallel to grain and perpendicular to grain,*** proposed models gave a good agreement with the test results.

BACKGROUND Psychosocial and physical trauma are known risk factors for irritable bowel syndrome(IBS),including in war veterans,whereas war exposure in civilians is *** experienced two wars,1970-1990:The Sandinistas Revolution(1970s)and The Contra War(1980s).Our aim was to investigate the role of exposure to war trauma in the subsequent development of IBS in the context of an established health surveillance system(11000 households).AIM To investigate in a civilian population the relationship between exposure to war trauma and events and the subsequent development of IBS in the context of an established public health and demographic surveillance system in western *** We conducted a nested population-based,cross-sectional study focused on functional gastrointestinal disorders based on Rome II criteria.1617 adults were randomly *** Spanish Rome II Modular Questionnaire and Harvard Trauma Questionnaire were validated in *** exposure was assessed with 10 measures of direct and indirect war trauma and post-war *** exposures were defined by≥3 *** The prevalence of IBS was 15.2%[Female(F)17.1%,Male(M)12.0%],war exposure 19.3%(F 9.3%,M 36.7%),and post-traumatic stress disorder(PTSD)5.6%(F 6.4%,M 4.3%).Significant associations with IBS in the civilian population were observed(adjusted by gender,age,socioeconomic status,education):physical and psychological abuse[adjusted odds ratio(aOR):2.25;95%confidence interval:1.1-4.5],witnessed execution(aOR:2.4;1.1-5.2),family member death(aOR:2.2;1.2-4.2),and multiple exposures(aOR:2.7;1.4-5.1).PTSD was independently associated with IBS(aOR:2.6;1.2-5.7).CONCLUSION An enduring association was observed in the Nicaragua civilian population between specific civil war-related events and subsequent *** populations in regions with extended armed conflict may warrant provider education and targeted interventions for patients.
