Can non-typeable Haemophilus influenzae carriage surveillance data infer antimicrobial resistance associated with otitis media?

Importance:In remote communities of the Northern Territory,Australia,children experience high rates of otitis media(OM),commonly caused by non-typeable Haemophilus influenzae(NTHi).Few data exist on antibiotic susceptibility of NTHi from ***:To determine whether population-level nasopharyngeal NTHi antibiotic susceptibility data could inform antibiotic treatment for ***:NTHi isolates(n=92)collected from ear discharge between 2003 and 2013 were selected to time-and age-match NTHi isolates from the nasopharyngeal carriage(n=95).Antimicrobial susceptibility were *** trees and a genome-wide association study(GWAS)were performed to determine the similarity of nasopharyngeal and ear isolates at a population ***:Among 174 NTHi isolates available for antimicrobial susceptibility testing,10.3%(18/174)were resistant to ampicillin and 9.2%(16/174)were resistant to *** numbers of isolates(≤3)were resistant to tetracycline,chloramphenicol,or amoxicillin-clavulanic *** was no statistical difference in the proportion of ampicillin-resistant(P=0.11)or trimethoprim-sulfamethoxazole-resistant isolates(P=0.70)between ear discharge and nasopharynx-derived NTHi *** multi-drug resistant NTHi isolates were *** trees showed no clustering of 187 Haemophilus influenzae isolates based on anatomical niche(nasopharynx or ear discharge),and no genetic variations that distinguished NTHi derived from ear discharge and nasopharyngeal carriage were evident in the ***:In this population-level study,nasopharyngeal and ear discharge isolates did not represent distinct microbial *** results support tracking of population-level nasopharyngeal NTHi antibiotic resistance patterns to inform clinical management of OM in this population.

Derived, still living cockroach genus Cariblattoides （Blattida： Blattellidae） from the Eocene sediments of Green River in Colorado, USA

Cariblattoides labandeirai sp.n. from the Eocene sediments of Green River in Colorado, USA bear only two plesiomorphies, but also several significant autapomorphies within the advanced and highly derived living cockroach genus. Thus, Cariblattoides with extant occurrence in the Caribbean and South America was historically common in the Nearctic, and represents important evidence for the occurrence of derived living genera of cockroaches -50 Ma ago. Generally, the vast majority of living genera were absent during the Palaeocene, thus the diversification of most living cockroach lineages near the Palaeocene/Eocene boundary must have been extremely rapid. Females of living C. suave, the type species, have identical （sophisticated） coloration ofpronotum, but the most related living taxa are C. piraiensis and *** from Brazil （supported by phylogenetical analysis）.

Hepatic intra-arterial infusion of yttrium-90 microspheres in the treatment of recurrent hepatocellular carcinoma after liver transplantation: A case report

Hepatocellular carcinoma （HCC） recurs with a reported frequency of 12%-18% after liver transplantation. Recurrence is associated with a mortality rate exceeding 75%. Approximately one-third of recurrences develop in the transplanted liver and are therefore amenable to local therapy. A variety of treatment modalities have been reported including resection, transarterial chemoembolization （TACE）, radiofrequency ablation （RFA）, ethanol ablation, cryoablation, and external beam irradiation. Goals of treatment are tumor control and the minimization of toxic effect to functional parenchyma. Efficacy of treatment is mitigated by the need for ongoing immunosuppression. Yttrium-90 microspheres have been used as a treatment modality both for primary HCC and for pre-transplant management of HCC with promising results. Twenty-two months after liver transplantation for hepatitis C cirrhosis complicated by HCC, a 42-year old man developed recurrence of HCC in his transplant allograft. Treatment of multiple right lobe lesions with anatomic resection and adjuvant chemotherapy was unsuccessful. Multifocal recurrence in the remaining liver allograft was treated with hepatic intra-arterial infusion of yttrium-90 microspheres （SIR-Spheres, Sirtex Medical Inc., lake Forest, Il, USA）. Efficacy was demonstrated by tumor necrosis on imaging and a decrease in alpha-fetoprotein （AFP） level. There were no adverse consequences of initial treatment.

Folate binding protein—Outlook for drug delivery applications

Serum proteins represent an important class of drug and imaging agent delivery vectors. In this mini- review, key advantages of using serum proteins are discussed, followed by the particular advantages and challenges associated with employing soluble folate binding protein. In particular, approaches employing drugs that target folate metabolism are reviewed. Additionally, the slow-onset, tight- binding interaction of folate with folate binding protein and the relationship to a natural oligomerization mechanism is discussed. These unique aspects of folate binding protein suggest interesting applications for the protein as a vector for further drug and imaging agent development.

Is analysis of lower esophageal sphincter vector volumes of value in diagnosing gastroesophageal reflux disease?

AIM:With successful surgical treatment of gastroesophageal reflux disease(GERD),there is interest in understanding the anti-reflux barrier and its mechanisms of *** date, the potential use of vector volumes to predict the DeMeester score has not been adequately explored. METHODS:627 patients in the referral database received esophageal manometry and ambulatory 24-hour pH *** data included lES resting pressure(lESP), overall lES length(Ol)and abdominal length(Al),total vector volume(TVV)and intrabdominal vector volume(IVV). RESUlTS:In cases where lESP,TVV or IVV were all below normal,there was an 81.4%probability of a positive DeMeester *** cases where all three were normal, there was an 86.9%probability that the DeMeester score would be ***-operating characteristics(ROC) for lESP,TVV and IVV were nearly identical and indicated no useful cut-off *** regression demonstrated that lESP and IVV had the strongest association with a positive DeMeester score;however,the regression formula was only 76.1%accurate. CONClUSION:While the indices based on TVV,IVV and lESP are more sensitive and specific,respectively,than any single measurement,the measurement of vector volumes does not add significantly to the diagnosis of GERD.

Biotemplated metal nanoclusters have garnered much attention owing to their wide range of potential applications in biosensing, bioimaging, catalysis, and nanomedicine. Here, we report the synthesis of stable, biocompatible, watersoluble, and highly fluorescent bovine serum albumin-templated cadmium nanoclusters （CdNcs） through a facile one-pot green method. We covalently conjugated hyaluronic acid （HA） to the CdNcs to form a pH-responsive, tumor- targeting theranostic nanocarrier with a sustained release profile for doxorubicin （DOX）, a model anticancer drug. The nanocarrier showed a DOX encapsulation efficiency of about 75.6%. DOX release profiles revealed that 74% of DOX was released at pH 5.3, while less than 26% of DOX was released at pH 7.4 within the same 24-h period. The nanocarrier selectively recognized MCF-7 breast cancer cells expressing CD44, a cell surface receptor for HA, whereas no such recognition was observed with HA receptor-negative HEK293 cells. Biocompatibility of the nanocarrier was evaluated through cytotoxicity assays with HEK293 and MCF-7 ceils. The nanocarrier exhibited very low to no cytotoxicity, whereas the DOX-loaded nanocarrier showed considerable cellular uptake and enhanced MCF-7 breast cancer cell-killing ability. We also confirmed the feasibility of using the highly fluorescent nanoconjugate for bioimaging of MCF-7 and Hela cells. The superior targeted drug delivery efficacy, cellular imaging capability, and low cytotoxicity position this nanoconjugate as an exciting new nanoplatform with promising biomedical applications.

The Effects of Antibiotics, Metals, and Biotic Interactions on the Assembly of Taxonomically Diverse Single and Mixed Species Biofilms

To better understand the assembly of the sturgeon egg microbiome, we purified six bacterial isolates from eggs and characterized their ability to form biofilms under the stress of tobramycin, with and without exogenous protein. In experiments with single species biofilms, tobramycin reduced the metabolic activity of all isolates and increased biofilm biomass of three. The addition of exogenous protein to the assay countered the inhibition of biofilm and metabolic activity by tobramycin of Pseudomonas sp., Brevundimonas sp., Flavobacterium columnare and mixed biofilms of Pseudomonas-F. columnare and Brevundimonas-Hydrogenophaga. Two of the isolates (Pseudomonas spp.) that produced antimicrobial activity, were effective at reducing biofilm formation by Brevundimonas, but enhanced biofilm formation in other isolates. Increasing concentrations of Mg2+ had no effect on biofilm formation but Ca2+ enhanced biofilm formation of Pseudomonas aeruginosa PA01 (positive control) and Brevundimonas. Biofilm assembly by these two bacteria was inhibited by low concentrations of Ni2+. Mixed biofilms of Brevundimonas and Hydrogenophage consistently produced more robust biofilm than the strains in isolation, suggesting synergism. Established Brevundimonas biofilm appeared adept at recruiting pelagic Acidovorax and Hydrogenophaga into biofilm, suggesting that it plays an important role in the selection of species into the microbiome.
