Disruption of colonic barrier function and induction of mediator release by strains of campylobacter jejuni that invade epithelial cells

AIM: To study the mechanisms by which campylobacter jejuni (c. jejuni) causes inflammation and diarrhea. In particular, direct interactions with intestinal epithelial cells and effects on barrier function are poorly under- stood. METHODS: To model the initial pathogenic effects of c. jejuni on intestinal epithelium, polarized human colonic HcA-7 monolayers were grown on permeabilized filters and infected apically with clinical isolates of c. jejuni. Integrity of the monolayer was monitored by changes in monolayer resistance, release of lactate dehydrogenase, mannitol fluxes and electron microscopy. Invasion of HcA-7 cells was assessed by a modified gentamicin protection assay, translocation by counting colony forming units in the basal chamber, stimulation of mediator release by immunoassays and secretory responses in monolayers stimulated by bradykinin in an Ussing chamber. RESULTS: All strains translocated across monolayers but only a minority invaded HcA-7 cells. Strains that invaded HcA-7 cells destroyed monolayer resistance over 6 h, accompanied by increased release of lactate dehydrogenase, a four-fold increase in permeability to [3H] mannitol, and ultrastructural disruption of tight junctions, with rounding and lifting of cells off the filter membrane. Synthesis of interleukin (IL)-8 and prostaglandin E2 was increased with strains that invaded the monolayer but not with those that did not. cONcLUSION: These data demonstrate two distinct effects of c. jejuni on colonic epithelial cells and provide an informative model for further investigation of initial host cell responses to c. jejuni.

Myocarditis is a disease process that every emergency physician fears *** severity can be mild to life-threatening,and many cases are likely undetected because they are subclinical with nonspecifi c signs.[1]Subtle cardiac signs may be overshadowed by systemic symptoms of the underlying infectious ***,myalgias,lethargy,symptoms commonly associated with viral syndrome,can mask the life-threatening myocarditis that may be *** fact,in the United States Myocarditis Treatment Trial,almost 90%of patients reported symptoms consistent with a viral prodrome.[2]Ammirati et al[3]reported that 27%of patients with myocarditis had either reduced left ventricular ejection fraction,ventricular arrhythmias,or low cardiac ***,we present a case report,in which handheld point-of-care ultrasound was utilized at the bedside to aid in the critical diagnosis of *** the additional information provided through this imaging modality,this patient was able to be transferred to the appropriate tertiary care facility in an expeditious manner and receive possible defi nitive treatment.

Magnetic resonance imaging(MRI) is a well established technique that has revolutionized diagnostic radiology. Until recently, the impact that MRI has had in the assessment of gastrointestinal motor function and bowel fluid distribution in health and in disease has been more limited, despite the novel insights that MRI can provide along the entire gastrointestinal tract. MRI biomarkers include intestinal motility indices, small bowel water content and whole gut transit time. The present review discusses new developments and applications of MRI in the upper gastrointestinal tract, the small bowel and the colon reported in the literature in the last 5 years.

capsicum is a nutritious vegetable and its cultivation in farms is getting popular in Bangladesh. Although many efforts have lain to explore better yielding and nutritionally rich cultivars with suitable modern cultivation techniques but still have to find the desired outcome. Thus, it’s necessary to conduct further research to identify the high-yielding and nutritious capsicum cultivars in Bangladesh. An experiment was conducted from July 2021 to June 2022 at the Bangladesh Institute of Research and Training on Applied Nutrition (BIRTAN) research field with three cultivars of capsicum: B0 = california Wonder, B1 = BARI Misti Morich-1 and B2 = BARI Misti Morich-2 and three mulching: T0 = No mulching, T1 = Water hyacinth, T2 = Poly Mulching in randomized complete block design with three replications to identify better quality capsicum cultivar and suitable mulching material. Among cultivars the BARI Misti Morich-2 (B2) showed increased agronomic parameters like number of branches and effective branches per plant, leaves length and width, consequently yield and yield contributing traits were also enhanced like fruits per plant, fruit length, fruit diameter and yield per plant (25.97%, 4.54%, 3.64% and 21.43%, respectively). Poly Mulching (T2) increased agronomic traits, yield traits and yield (0.61 kg) than BARI Misti Morich-1 (T1). The combined effect of B2T2 increased the number of branches per plant, effective branches per plant, leaves length and breadth by 40%, 90%, 15.57% and 26.22%, respectively, hence resulting in an increased yield of 20%. BARI Misti Morich-2 cultivar showed an increase in Fe, Zn and Vitamin-c content of 26.24% and 23.10%, 8.82% and 5.14%, and 6.03% and 5.74% than B0 and B1 cultivars, respectively. Total temporomandibular joint(TMJ)replacement is recommended only when there is irreversible damage to the joint and no conservative treatment can provide functional *** stock and custom-made TMJ implants have been made available;however,retrospective and comparative studies were unable to find significant differences between the two types of *** introduction of additive manufacturing(AM)techniques in medical practice allows for a greater freedom of design and a higher degree of device *** combination of AM with structural optimisation may streamline development and provide the key for fabricating biomechanic ally enhanced TMJ *** this study,structural optimis ation techniques were applied to develop and numerically validate a patient-specific TMJ *** biomechanical behaviour of each intermediate TMJ design was assessed under four different nominal and maximum biting tasks using finite element *** addition,a new set of metrics were proposed to compare each design regarding biomechanical performance and implant *** results suggest that 55-82%of the natural/intact strain patterns can be recovered with the finally selected TMJ *** represents an increase of 15%in biomechanical performance for incisor biting,15%for right molar biting,17%for left molar biting and a decrease of 2%for left group biting compared with the initial *** results also suggest that load transfer at the proximal ramus reduces the implant’s impact on the mandible’s strain ***,structural optimisation allows for a volume reduction of up to 44%with a minimum loss of implant safety and biomechanical performance.

Purpose:This study aimed to determine the accuracy of a 4 split time modelling method to generate velocity-time and velocity-distance variables in elite male 100-m sprinters and subsequently to assess the roles of key sprint parameters with respect to 100-m sprint ***,this study aimed to assess the differences between faster and slower sprinters in key sprint variables that have not been assessed in previous ***:Velocity-time and velocity-distance curves were generated using a mono-exponential function from 4 split times for 82 male sprinters during major athletics *** race variables-maximum velocity,the acceleration time constant(τ),and percentage of velocity lost(vLoss)-were derived for each *** were divided into tertiles,based on 100-m time,with the first and third tertiles considered to be the faster and slower groups,respectively,to facilitate further ***:Modelled split times and velocities displayed excellent accuracy and close agreement with raw measures(range of mean bias was-0.2%to 0.2%,and range of intraclass correlation coefficients(Iccs)was 0.935 to 0.999)except for 10-m time(mean bias was 1.6%±1.3%,and the Icc was 0.600).The 100-m sprint performance time and all 20-m split times had a significant near-perfect negative correlation with maximum velocity(r≥-0.90)except for the 0 to 20-m split time,where a significantly large negative correlation was found(r=-0.57).The faster group had a significantly higher maximum velocity andτ(pcant difference was found for vLoss(p=0.085).conclusion:coaches and researchers are encouraged to utilize the 4 split time method proposed in the current study to assess several key race variables that describe a sprinter’s performance capacities,which can be subsequently used to further inform training.
