SYNGR4 and PLEKHB1 deregulation in motor neurons of amyotrophic lateral sclerosis models: potential contributions to pathobiology

Amyotrophic lateral sclerosis is the most common adult-onset neurodegenerative disease affecting motor neurons. Its defining feature is progressive loss of motor neuron function in the cortex, brainstem, and spinal cord, leading to paralysis and death. Despite major advances in identifying genes that can cause disease when mutated and model the disease in animals and cellular models, it still remains unclear why motor symptoms suddenly appear after a long pre-symptomatic phase of apparently normal function. One hypothesis is that age-related deregulation of specific proteins within key cell types, especially motor neurons themselves, initiates disease symptom appearance and may also drive progressive degeneration. Genome-wide in vivo cell-type-specific screening tools are enabling identification of candidates for such proteins. In this minireview, we first briefly discuss the methodology used in a recent study that applied a motor neuron-specific RNASeq screening approach to a standard model of TAR DNA-binding protein-43(TDP-43)-driven amyotrophic lateral sclerosis. A key finding of this study is that synaptogyrin-4 and pleckstrin homology domain-containing family B member 1 are also deregulated at the protein level within motor neurons of two unrelated mouse models of mutant TDP-43 driven amyotrophic lateral sclerosis. Guided by what is known about molecular and cellular functions of these proteins and their orthologs, we outline here specific hypotheses for how changes in their levels might potentially alter cellular physiology of motor neurons and detrimentally affect motor neuron function. Where possible, we also discuss how this information could potentially be used in a translational context to develop new therapeutic strategies for this currently incurable, devastating disease.

Proton exchange membrane(PEM)water electrolysis represents a promising technology for green hydrogen production,but its widespread deployment is greatly hindered by the indispensable usage of platinum group metal catalysts,especially iridium(Ir)based materials for the energy-demanding oxygen evolution reaction(OER).Herein,we report a new sequential precipitation approach to the synthesis of mixed Ir-nickel(Ni)oxy-hydroxide supported on antimony-doped tin oxide(ATO)nanoparticles(IrNiyO_(x)/ATO,20 wt.%(Ir+Ni),y=0,1,2,and 3),aiming to reduce the utilisation of scarce and precious Ir while maintaining its good acidic OER *** tested in strongly acidic electrolyte(0.1 M HClO_(4)),the optimised IrNi1Ox/ATO shows a mass activity of 1.0 mAµgIr^(−1) and a large turnover frequency of 123 s^(−1) at an overpotential of 350 mV,as well as a comparatively small Tafel slope of 50 mV dec^(−1),better than the IrOx/ATO control,particularly with a markedly reduced Ir loading of only 19.7µgIr cm^(−2).Importantly,IrNi1O_(x)/ATO also exhibits substantially better catalytic stability than other reference catalysts,able to continuously catalyse acidic OER at 10 mA cm^(−2) for 15 h without obvious *** in-situ synchrotron-based x-ray absorption spectroscopy confirmed that the Ir^(3+)/Ir^(4+)species are the active sites for the acidic ***,the performance of IrNi1Ox/ATO was also preliminarily evaluated in a membrane electrode assembly,which shows better activity and stability than other reference *** IrNi1Ox/ATO reported in this work is a promising alternative to commercial IrO_(2) based catalysts for PEM electrolysis.

Mg-Zn-Zr(ZK)alloys exhibit notably high mechanical strength amongst all magnesium alloy ***,due to the formation of low melting point Mg_3Zn_7-precipitates,these alloys are susceptible to hot cracking,thus compromising their metallurgical *** addition of rare earths to ZK alloys is an alternative to form higher melting point intermetallic compounds,speed up dynamic recrystallization,refine grain size,enhance corrosion resistance and extend the service temperature due to improved creep *** work deals with the effect of Ce-base mischmetal addition on the hot rolling behaviour of as-cast ZK60 *** microstructure investigation conducted using electron microscopy and X-Ray diffraction shows that precipitation of Mg_(7)Zn_(3) intermetallics occur during hot rolling,whereas no further precipitation is observed for the ZK60-Mm *** fragmentation of the intermetallic compounds occur during hot rolling and finer particles of Mg_(7)Zn_(3) are observed for the ZK60,whereas Mg_(7)Zn_(3) and Mg Zn_(2) Ce intermetallics are formed in the alloy modified with mischmetal addition.A higher fraction of dynamically recrystallized grains is observed for the ZK60-Mm in comparison to the *** recrystallization takes place in ZK60 with the formation of sub-grains near to the intermetallics and the addition of mischmetal promotes the occurrence of discontinuous recrystallization with the nucleation of new grains close to the *** mechanical strength and,in particular,the ductility of the hot-rolled alloys are notably improved when compared to the same alloys in the as-cast *** mechanical strength is,however,higher for the ZK60 *** solid solution strengthening,softer Mg Zn_(2) Ce intermetallics and more extensive recrystallization contribute to reduce the mechanical strength of *** in both alloys are initiated at coarse intermetallics and propagate through intermetallic-rich regions.

Methionine sources and genotype affect embryonic intestinal development, antioxidants, tight junctions, and growth-related gene expression in chickens

Methionine(Met) is an essential and first limiting amino acid in the poultry diet that plays a significant role in chicken embryonic development and growth. The present study examined the effect of in ovo injection of DL-Met and L-Met sources and genotypes on chicken embryonic-intestinal development and health. fertilized eggs of the two genotypes, TETRA-SL layer hybrid(TSL)—commercial layer hybrid and Hungarian Partridge colored hen breed(HPC)—a native genotype, were randomly distributed into four treatments for each genotype. The treatment groups include the following: 1) control non-injected eggs(NoIn); 2) saline-injected(SaIn); 3) DL-Met injected(DLM); and 4) L-Met injected(LM). The in ovo injection was carried out on 17.5 d of embryonic development; after hatching, eight chicks per group were sacrificed, and the jejunum was extracted for analysis. The results showed that both DLM and LM groups had enhanced intestinal development as evidenced by increased villus width, villus height, and villus area(P ficantly reduced crypt depth, glutathione(GSH) content, glutathione S-transferase 3 alpha(GST3), occludin(OCLN) gene expression and increased villus height to crypt depth ratio in the TSL genotype than the LM group(P factor 1(IGf1) gene, tricellulin(MD2), occludin(OCLN),superoxide dismutase 1(SOD1), and GST3 genes than the TSL genotype(P findings showed that in ovo injection of Met enhanced intestinal development, and function, with genotypes responding differently under normal conditions. Genotypes also influenced the expression of intestinal antioxidants, tight junction, and growth-related genes.

The SARS-CoV-2 Omicron variants are notorious for their transmissibility,but little is known about their subgenomic RNA(sgRNA)*** study applied RNA-seq to delineate the quantitative and qualitative profiles of canonical sgRNA of 118 respiratory samples collected from patients infected with Omicron BA.2 and compared with 338 patients infected with non-variant of concern(non-VOC)-D614G.A unique characteristic profile depicted by the relative abundance of 9 canonical sgRNAs was reproduced by both BA.2 and non-VOCD614G regardless of host gender,age and presence of ***,such profile was lost in samples with low viral load,suggesting a potential application of sgRNA pattern to indicate viral activity of individual patient at a specific time point.A characteristic qualitative profile of canonical sgRNAs was also reproduced by both BA.2 and *** presence of a full set of canonical sgRNAs carried a coherent correlation with crude viral load(AUC¼0.91,95%CI 0.88–0.94),and sgRNA ORf7b was identified to be the best surrogate marker allowing feasible routine application in characterizing the infection status of individual *** potentials in using sgRNA as a target for vaccine and antiviral development are worth pursuing.

Perovskite solar cells(PSCs)have made great advances in terms of power conversion efficiency(PCE),yet their subpar stability continues to hinder their *** interface between the perovskite layer and the charge-carrier transporting layers plays a crucial role in undermining the stability of *** this work,we propose a strategy to stabilize high-performance PSCs with PCE over 23%by introducing a cesium-doped graphene oxide(GO-Cs)as an interlayer between the perovskite and hole-transporting *** GO-Cs treated PSCs exhibit excellent operational stability with a projected T80(the time where the device PCE reduces to 80%of its initial value)of 2143 h of operation at the maximum powering point under one sun illumination.

Preserving the literary past, looking to the future: the first Hong Kong Literature Database

In the last two decades of the 20th century, there has been an increasing interest in and emphasis on the study of the Hong Kong literature in both the academic and general public in Hong Kong. Recognizing the emergent need of the resources on Hong Kong literature, the University Library System of the Chinese University of Hong Kong set up the Hong Kong Literature Database （the “Database”）, which was the first Chinese literature database in the Internet in 2000. The paper will examine how the database is constructed using XML technology andometadata schema, The database also employs Unicode UTf-8 as the internal code. A mapping table for traditional and simplified Chinese characters was created based on Unihan and is used behind the scene so that a user can either input traditional or simplified Chinese characters and retrieval will give both traditional and simplified Chinese characters. Currently 65% of journals use OCR technology so that full-text searching is possible. The Chinese OCR technology will be examined in greater detail. Special features of the Database such as, page-by-page browse mode, position-highlight for full-page newspaper, linking Table-Of-Contents and book jackets from the Library catalogue, etc. are described. The paper will also bring out the problem of massive downloading and compare the state-of-the-art technology and their shortcomings. This paper shows how the Hong Kong Literature Database facilitates future collaboration and data exchange by using open standard, shareable structure and the latest technology.

Superconductivity in antiperovskites

We present a comprehensive theoretical study of conventional superconductivity in cubic antiperovskites materials with composition XYZ_(3) where X and Z are metals,and Y is H,B,C,N,O,and *** starting point are electron–phonon calculations for 397 materials performed with density-functional perturbation *** 43%of the materials are dynamically unstable,we discovered 16 compounds close to thermodynamic stability and with T_(c) higher than 5 *** these results to train interpretable machine-learning models,leads us to predict a further 57(thermodynamically unstable)materials with superconducting transition temperatures above 5 K,reaching a maximum of 17.8 K for ***,the models give us an understanding of the mechanism of superconductivity in *** combination of traditional approaches with interpretable machine learning turns out to be a very efficient methodology to study and systematize whole classes of materials and is easily extendable to other families of compounds or physical properties.

Heusler compounds attract a great deal of attention from researchers thanks to a wealth of interesting properties,among which is *** we perform an extensive study of the superconducting and elastic properties of the cubic(full-)Heusler family using a mixture of ab initio methods,as well as interpretable and predictive machine-learning *** analyzing the statistical distributions of these properties and comparing them to anti-perovskites,we recognize universal behaviors that should be common to all conventional superconductors while others turn out to be specific to the material *** total,we discover a total of eight hypothetical materials with critical temperatures above 10 K to be compared with the current record of T_(c)=4.7 K in this ***,we expect most of these materials to be highly ductile,making them potential candidates for the manufacture of wires and tapes for superconducting magnets.
