Iron increases HMOX1 and decreases hepatitis C viral expression in HCV-expressing cells

AIM： To investigate effects of iron on oxidative stress, heme oxygenase-1 （HMOX1） and hepatitis C viral （HCV） expression in human hepatoma ceils stably expressing HCV proteins. METHODS： Effects of iron on oxidative stress, HMOX1, and HCV expression were assessed in CON1 cells. Measurements included mRNA by quantitative reverse transcription-polymerase chain reaction, and protein levels by western blots. RESULTS： Iron, in the form of ferric nitrilotriacetate,increased oxidative stress and upegulated HMOX1 gene expression. Iron did not affect mRNA or protein levels of Bach1, a repressor of HMOXl. Silencing the up-regulation of HMOXl nuclear factor-erythroid 2-related factor 2 （Nrf2） by Nrf2-siRNA decreased FeNTA-mediated up-regulation of HMOXl mRNA levels. These iron effects were completely blocked by deferoxamine （DFO）. Iron also significantly decreased levels of HCV core mRNA and protein by 80%-90%, nonstructural 5A mRNA by 90% and protein by about 50% in the Con1 full length HCV replicon cells, whereas DFO increased them. CONCLUSION： Excess iron up-regulates HMOXl and down-regulates HCV gene expression in hepatoma cells. This probably mitigates liver injury caused by combined iron overload and HCV infection.

Macrophages,which are key cellular components of the liver,have emerged as essential players in the maintenance of hepatic homeostasis and in injury and repair processes in acute and chronic liver *** liver injury,resident Kupffer cells(KCs)sense disturbances in homeostasis,interact with hepatic cell populations and release chemokines to recruit circulating leukocytes,including monocytes,which subsequently differentiate into monocyte-derived macrophages(MoMφs)in the *** KCs and MoMφs contribute to both the progression and resolution of tissue inflammation and injury in various liver *** diversity of hepatic macrophage subsets and their plasticity explain their different functional responses in distinct liver *** this review,we highlight novel findings regarding the origins and functions of hepatic macrophages and discuss the potential of targeting macrophages as a therapeutic strategy for liver disease.

The simplest picture of excitons in materials with atomic-like localization of electrons is that of Frenkel excitons,where electrons and holes stay close together,which is associated with a large binding ***,using the example of the layered oxide V_(2)O_(5),we show how localized charge-transfer excitations combine to form excitons that also have a huge binding energy but,at the same time,a large electron-hole distance,and we explain this seemingly contradictory *** anisotropy of the exciton delocalization is determined by the local anisotropy of the structure,whereas the exciton extends orthogonally to the chains formed by the crystal ***,we show that the bright exciton goes together with a dark exciton of even larger binding energy and more pronounced *** findings are obtained by combining first principles many-body perturbation theory calculations,ellipsometry experiments,and tight binding modelling,leading to very good agreement and a consistent *** explanation is general and can be extended to other materials.

＜正＞High-resolution seismic reflections have been used effectively to investigate sinkholes formed from the dissolution of a bedded salt unit found throughout most of central Kansas. Surface subsidence can have devastating effects on transportation structures. Roads and rails, bridges, and pipelines can be adversely affected by even minor ground instability. Areas susceptible to rapid surface subsidence represent a potential risk to public safety. Significantly larger subsurface expressions relative to surface depressions are a consistent observation on seismic images recorded over sinkholes in Kansas. It appears until subsidence reaches the ground surface, failure is controlled by faults with reverse orientation. Once a surface depresssion begins to form and dissolution of the salt slows or stops, subsidence along rock layers manifests itself with a more normal fault geometry. Detecting areas of rapid subsidence potential, pre-surface failure, is the ultimate goal of any geotechnical survey where the ground surface is susceptible to settling. Seismic reflection images have helped correlate present subsidence to paleofeatures, project the total horizontal growth of active sinkholes, and appraise the risk of catastrophic failure. It is likely active dissolution and the associated fluid movement within the salt interval correlates to collapse geometries observed in salt overburden rocks. Geologic structures play a significant role in both controlling sinkhole expansion and in the susceptibility for sinkholes to form in areas above the Hutchinson Salt in Kansas.

Changes in tumor-antigen expression profile as human small-cell lung cancers progress

Objective： Our group has previously observed that in patients with small-cell lung cancers （SCLCs）, the expression of a tumor antigen, glioma big potassium （gBK） ion channel, is higher at the time of death than when the cancer is first treated by surgical resection. This study aimed to determine whether this dichotomy was common in other potential lung tumor antigens by examining the same patient samples using our more extensive profile analysis of tumor-antigen precursor protein （TAPP）. we then tested the hypothesis that therapeutic intervention may inadvertently cause this increased gBK production. Methods： SCLC samples （eight surgical resections and three autopsy samples） and three control lungs were examined by quantitative real-time polymerase chain reaction for 42 potential TAPPs that represent potential T-cell-mediated immunological targets. Results： Twenty-two TAPP mRNAs displayed the same profile as gBK, i.e., more mRNAs were expressed at autopsy than in their surgical counterparts. B-cyclin and mouse double minute 2, human homolog of PS3-binding protein were elevated in both autopsy and surgical specimens above the normal-lung controls. when HTB119 cells were incubated with doxorubicin, gBK was strongly induced, as confirmed by intracellular flow cytometry with a gBK-specific antibody. Conclusion： Our findings suggested that more immunological targets became available as the tumor responded to chemotherapy and proceeded toward its terminal stages.

Solid Aneurysmal Bone Cyst of the Distal Metatarsus in a Horse

A horse was presented with a slowly growing mass of the distal metatarsal region of the right hind leg. Radiographic examination revealed an eccentric cyst-like lesion with distortion of the lateral margin of the distal metatarsus. The lesion involved approximately half of the metatarsal width, exhibited fine septa, a sclerosed margin towards unaffected bone and a smooth rim of thin compact bone at the periphery. Longitudinal sectioning of the distal metatarsus revealed a solid pale yellow mass with reddish foci and margin. Histologically, the mass consisted of a cell-rich, well vascularized fibrous stroma, containing numerous irregularly shaped trabeculae of woven bone with regular osteocytes. Osteoblasts were observed around the osteoid, and there were foci of numerous osteoclast-like giant cells. The lesion was diagnosed solid variant of an aneurismal bone cyst.
