The structure of the irreducible collective spaces of the group Sp(12,R),which many-particle nuclear states are classified according to the chain Sp(12,R)⊃U(6)⊃SO(6)⊃SU_(pn)(3)⊗SO(2)⊃SO(3)of the proton-neutron symplectic model(PNSM),is considered in *** chain of the PNSM was recently shown to correspond to a microscopic shell-model version of the Bohr-Mottelson collective *** construction of the relevant shell-model representations of the Sp(12,R)group along this chain is considered for three nuclei with varying collective properties and from different mass *** is shown that the SU_(pn)(3)basis states of the Sp(12,R)representations belonging to the SO(6)irreps with seniorityυ≥υ0,withυ0 denoting the maximal seniority SO(6)irrep contained in the Sp(12,R)bandhead,are always Pauli allowed,but organized in a different way into different SO(6)*** is in contrast to the case of filling the levels of the standard three-dimensional harmonic oscillator and using the plethysm *** the SU_(pn)(3)multiplets withinυhe present work are important for the practical application of the microscopic version of the Bohr-Mottelson collective model.

Detailed sedimentological analysis of the Pokaran Boulder Bed,representing the most basal unit of the Neoproterozoic-Lower Cambrian Marwar Supergroup,clearly indicates its glacial *** glacial sediments are interpreted as an ice-contact submarine fan *** on the detrital and inherited zircon population of the Marwar Supergroup sediments and interlayered pyroclastic deposits,a Marinoan cryochron of the Cryogenian Period is envisaged for the Pokaran Boulder *** well-preserved Ediacaran elements in the post-glacial sedimentary succession also support a Cryogenian to Early Cambrian age of the Marwar *** glacial deposition at the base of the Marwar Supergroup strengthens the regional correlation between the studied sediments with the Lesser himalayan Blaini-Krol-Tal sediments and the haqf Supergroup of *** available zircon ages and paleomagnetic data of the Malani Igneous suite,along with the Marinoan glacial deposits,detrital zircon ages,and Ediacaran fossil elements of the Marwar Supergroup add significant information to the Neoproterozoic Earth history.

Autosomal recessive spinocerebellar ataxias（SCARs） are one of the most common neurodegenerative diseases characterized by progressive ataxia. Although SCARs are known to be caused by mutations in multiple genes, there are still many cases that go undiagnosed or are misdiagnosed. In this study, we presented a SCAR patient, and identified a probable novel pathogenic mutation（c.1A>G, p.M1V） in the AFG3L2 start codon. The proband's genotype included heterozygous mutations of the compound AFG3L2(p.[M1V]; [R632X]（c.[1A>G];[1894.C>T]）), which were inherited from the father（c.1A>G, p.M1V） and mother（c.1894C>T, p.R632X）. Functional studies performed on hiPSCs（human induced pluripotent stem cells） generated from the patients and hEK293T cells showed that the mutations impair mitochondrial function and the unbalanced expression of AFG3L2 mRNA and protein levels. Furthermore, this novel mutation resulted in the degradation of the protein and the reduction of the stability of the AFG3L2 protein, and MCU（mitochondrial calcium uniporter） complex mediated Ca2+overload.

Metabolic flexibility has emerged as a critical determinant of CD8+T-cell antitumor activity,yet the mechanisms driving the metabolic flexibility of T cells have not been *** this study,we investigated the influence of the nuclear cap-binding complex(CBC)adaptor protein ARS2 on mature T *** doing so,we discovered a novel signaling axis that endows activated CD8+T cells with flexibility of glucose ***2 upregulation driven by CD28 signaling reinforced splicing factor recruitment to pre-mRNAs and affected approximately one-third of T-cell activation-induced alternative splicing *** these effects,the CD28-ARS2 axis suppressed the expression of the M1 isoform of pyruvate kinase in favor of PKM2,a key determinant of CD8+T-cell glucose utilization,interferon gamma production,and antitumor effector ***,PKM alternative splicing occurred independently of CD28-driven PI3K pathway activation,revealing a novel means by which costimulation reprograms glucose metabolism in CD8+T cells.

A phase Ⅰ study of hemay022, an irreversible dual EGFR/hER2 tyrosine kinase inhibitor in Chinese patients with hER2-positive advanced breast cancer

Objective: hemay022 is a novel small-molecule and an irreversible tyrosine kinase inhibitor with the target of epidermal growth factor receptor(EGFR)/human epidermal growth factor receptor 2(hER2), which demonstrated anti-tumor activity in preclinical studies. This first-in-human study evaluated the safety, pharmacokinetics,tolerability and preliminary anti-tumor activity of hemay022 in hER2-positive advanced breast cancer ***: heavily pretreated patients with hER2-positive advanced breast cancer were assigned to eight dose cohorts in a 3+3 dose-escalation pattern at doses of 50-600 mg QD and 300 mg BID. Eligible patients were given a single dose of hemay022 on d 1 in week 0, followed by once daily continuous doses for four weeks in 28-day *** samples were obtained on d 1 and d 28. Clinical responses were assessed every eight ***: Twenty-eight patients with advanced breast cancer were treated with hemay022. The most frequently reported drug-related adverse events were diarrhoea(85.7%), vomiting(28.6%), nausea(25.0%) and decreased appetite(17.9%). No grade 4 drug-related adverse events were reported. At 50-600 mg doses, steady state areas under the concentration-time curve and peak concentrations increased with doses. One patient achieved complete response(CR), and three achieved partial response(PR). The objective response rate(ORR) and disease control rate(DCR) were 14.3% and 46.4% in 28 patients, respectively. The median progression-free survival(PFS) was3.98 ***: hemay022 at the dose of 500 mg once daily was well tolerated. The pharmacokinetic properties and encouraging anti-tumor activities of hemay022 in advanced breast cancer patients warranted further evaluation of hemay022 for treating breast cancer patients in the current phase Ⅲ trial(No. NCT05122494).

Objective:The aim of this study was to review findings from a large prospective national database of chemosensory disturbances associated with coronavirus disease 2019(COVID-19)*** Sources:The Virginia Commonwealth University Smell and Taste Center national database of COVID-19 chemosensory ***:A series of online surveys,first opened on April 10,2020,was made accessible nationwide to any adult with sudden chemosensory dysfunction since January *** received subsequent follow-up surveys 14 days,1 month,3 months,and 6 months after *** additional survey was sent to all participants on May 28,2022 to assess long-term *** pertaining to demographics,symptoms,comorbidities,treatments,and life impact was ***:Of 363 participants who reported complete smell recovery,51.2%recovered within 1 month,70%within 3 months,and 79%within 6 months,while 8.8%took over 1 year to completely *** all participants,7.5%had no smell *** predictors of recovery included agehe presence of nasal *** predictors included difficulty breathing and prior head *** participants reported a decrease in quality of life and the presence of potential safety hazards associated with decreased smell ***:Most subjects with COVID-19-related chemosensory dysfunction recover,with the majority noting complete recovery within weeks of *** aged over 40 years and female gender were associated with lower rates of recovery.A considerable number of participants reported significant impact on quality of life and safety.

This paper investigates the problem of collecting multidimensional data throughout time(i.e.,longitudinal studies)for the fundamental task of frequency estimation under Local Differential Privacy(LDP)*** to frequency estimation of a single attribute,the multidimensional aspect demands particular attention to the privacy ***,when collecting user statistics longitudinally,privacy progressively ***,the“multiple”settings in combination(i.e.,many attributes and several collections throughout time)impose several challenges,for which this paper proposes the first solution for frequency estimates under *** tackle these issues,we extend the analysis of three state-of-the-art LDP protocols(Generalized Randomized Response–GRR,Optimized Unary Encoding–OUE,and Symmetric Unary Encoding–SUE)for both longitudinal and multidimensional data *** the known literature uses OUE and SUE for two rounds of sanitization(***),i.e.,L-OUE and L-SUE,respectively,we analytically and experimentally show that starting with OUE and then with SUE provides higher data utility(i.e.,L-OSUE).Also,for attributes with small domain sizes,we propose Longitudinal GRR(L-GRR),which provides higher utility than the other protocols based on unary ***,we also propose a new solution named Adaptive LDP for LOngitudinal and Multidimensional FREquency Estimates(ALLOMFREE),which randomly samples a single attribute to be sent with the whole privacy budget and adaptively selects the optimal protocol,i.e.,either L-GRR or *** shown in the results,ALLOMFREE consistently and considerably outperforms the state-of-the-art L-SUE and L-OUE protocols in the quality of the frequency estimates.
