The impacts of weather and climate-related disasters are increasing,and climate change can exacerbate many *** communicating climate risk and integrating science into policy requires scientists and stakeholders to work *** dialogue between scientists and policymakers can be challenging given the inherently multidimensional nature of the issues at stake when managing climate *** on the growing use of serious games to create dialogue between stakeholders,we present a new game for policymakers called Climate Attribution Under Loss and Damage:Risking,Observing,Negotiating(CAULDRON).CAULDRON aims to communicate understanding of the science attributing extreme events to climate change in a memorable and compelling way,and create space for dialogue around policy decisions addressing changing risks and loss and damage from climate *** describe the process of developing CAULDRON,and draw on observations of players and their feedback to demonstrate its potential to facilitate the interpretation of probabilistic climate information and the understanding of its relevance to informing *** looking to engage with stakeholders can learn valuable lessons in adopting similar innovative *** suitability of games depends on the policy context but,if used appropriately,experiential learning can drive coproduced understanding and meaningful dialogue.

The factors that determine fibrosis progression or normal tissue repair are largely *** previously demonstrated that autophagy inhibition-mediated epithelial-mesenchymal transition(EMT)in human alveolar epithelial type Il(ATIl)cells augments local myofibroblast differentiation in pulmonary fibrosis by paracrine ***,we report that liver kinase B1(LKB1)inactivation in ATIl cells inhibits autophagy and induces EMT as a *** IPF lungs,this is caused by the down-regulation of CAB39L,a key subunit within the LKB1 complex.3D co-cultures of ATIl cells and MRC5 lung fibroblasts coupled with RNA sequencing(RNA-seq)confirmed that paracrine signaling between LKB1-depleted ATIl cells and fibroblasts augmented myofibroblast ***,these data suggest that reduced autophagy caused by LKB1 inhibition can induce EMT in ATIl cells and contribute to fibrosis via aberrant epithelial-fibroblast crosstalk.

Objective: Hemay022 is a novel small-molecule and an irreversible tyrosine kinase inhibitor with the target of epidermal growth factor receptor(EgFR)/human epidermal growth factor receptor 2(HER2), which demonstrated anti-tumor activity in preclinical studies. This first-in-human study evaluated the safety, pharmacokinetics,tolerability and preliminary anti-tumor activity of Hemay022 in HER2-positive advanced breast cancer ***: Heavily pretreated patients with HER2-positive advanced breast cancer were assigned to eight dose cohorts in a 3+3 dose-escalation pattern at doses of 50-600 mg QD and 300 mg BID. Eligible patients were given a single dose of Hemay022 on d 1 in week 0, followed by once daily continuous doses for four weeks in 28-day *** samples were obtained on d 1 and d 28. Clinical responses were assessed every eight ***: Twenty-eight patients with advanced breast cancer were treated with Hemay022. The most frequently reported drug-related adverse events were diarrhoea(85.7%), vomiting(28.6%), nausea(25.0%) and decreased appetite(17.9%). No grade 4 drug-related adverse events were reported. At 50-600 mg doses, steady state areas under the concentration-time curve and peak concentrations increased with doses. One patient achieved complete response(CR), and three achieved partial response(PR). The objective response rate(ORR) and disease control rate(DCR) were 14.3% and 46.4% in 28 patients, respectively. The median progression-free survival(PFS) was3.98 ***: Hemay022 at the dose of 500 mg once daily was well tolerated. The pharmacokinetic properties and encouraging anti-tumor activities of Hemay022 in advanced breast cancer patients warranted further evaluation of Hemay022 for treating breast cancer patients in the current phase Ⅲ trial(No. NCT05122494).

The prevalence of diabetes mellitus and its associated complications,particularly diabetic foot pathologies,poses significant healthcare challenges and economic burdens *** review synthesises current evidence on the surgical management of the diabetic foot,focusing on the interplay between neuropathy,ischemia,and infection that commonly culminates in ulcers,infections,and,in severe cases,*** escalating incidence of diabetes mellitus underscores the urgency for effective management strategies,as diabetic foot complications are a leading cause of hospital admissions among diabetic patients,significantly impacting morbidity and mortality *** review explores the pathophysiological mechanisms underlying diabetic foot complications and further examines diabetic foot ulcers,infections,and skeletal pathologies such as Charcot arthropathy,emphasising the critical role of early diagnosis,comprehensive management strategies,and interdisciplinary care in mitigating adverse *** addressing surgical interventions,this review evaluates conservative surgeries,amputations,and reconstructive procedures,highlighting the importance of tailored approaches based on individual patient profiles and the specific characteristics of foot *** integration of advanced diagnostic tools,novel surgical techniques,and postoperative care,including offloading and infection control,are discussed in the context of optimising healing and preserving limb function.

Birch has long suffered from a lack of active forest management,leading many researchers to use mate-rial without a detailed management *** collected from three birch(Betula pendula Roth,*** Ehrh.)sites in southern Sweden were analyzed using regression analysis to detect any trends or differences in wood proper-ties that could be explained by stand history,tree age and stem *** sites were genetics trials established in the same *** of acoustic velocity(AV)from non-destructive testing(NDT)and predicted AV had a higher correlation if data was pooled across sites and other stem form factors were considered.A subsample of stems had radial profiles of X-ray wood density and ring width by year created,and wood density was related to ring number from the pith and ring *** seemed likely that wood density was negatively related to ring width for both birch *** models had slight improvements if site and species were included,but only the youngest site with trees at age 15 had both birch *** paper indicated that NDT values need to be considered separately,and any predictive models will likely be improved if they are specific to the site and birch species measured.

The increasing amount of user traffic on Internet discussion forums has led to a huge amount of unstructured natural language data in the form of user *** modern recommendation systems rely on manual tagging,relying on administrators to label the features of a class,or story,which a user comment corresponds *** common approach is to use pre-trained word embeddings to compare class descriptions for textual similarity,then use a distance metric such as cosine similarity or Euclidean distance to find top k ***,neither approach is able to fully utilize this user-generated unstructured natural language data,reducing the scope of these recommendation *** paper studies the application of domain adaptation on a transformer for the set of user comments to be indexed,and the use of simple contrastive learning for the sentence transformer fine-tuning process to generate meaningful semantic embeddings for the various user comments that apply to each *** order to match a query containing content from multiple user comments belonging to the same class,the construction of a subquery channel for computing class-level similarity is *** channel uses query segmentation of the aggregate query into subqueries,performing k-nearest neighbors(KNN)search on each individual *** achieves state-of-the-art performance,outperforming other state-of-the-art models in accuracy,precision,recall,and F1 score for classifying comments between four and eight classes,*** outperforms the most precise state-of-the-art model(distilRoBERTa)in precision by 6.97%for matching comments between eight classes.

Compact flame-holders for afterburners are an increasing requirement for modern aero ***,flame-holder design is non-trivial since high inlet temperatures,velocities,and elaborate structures induce complex turbulence,combustion,and spray coupling in modern *** this work,the LES-pdf and stochastic fields-Lagrangian particle spray methods are used to investigate methane and aviation kerosene combustion structures formed by new-type concave *** flow pattern,combustion mode,and flame structure of gaseous and liquid fuel around a concave flame-holder are analyzed,discussed,and compared with experimental *** reveal that the flame stability of a concave flame-holder is better than that of the non-concave ***,when using liquid fuel,the concave flame-holder forms a stable and compact *** results suggest concave flame-holders are a promising design for compact afterburners.
