Therapy with bone marrow cells reduces liver alterations in mice chronically infected by Schistosoma mansoni

AIM： To investigate the potential of bone marrow mononuclear cells （BM-MCs） in the regeneration of hepatic lesions induced by Schistosoma mansoni （***） chronic infection. METHODS： Female mice chronically infected with *** were treated with BM-MCs obtained from male green fluorescent protein （GFP） transgenic mice by intravenous or intralobular injections. Control mice received injections of saline in similar conditions, Enzyme-linked immunosorbent assay （ELISA） assay for transforming growth factor-beta （TGF-β）, polymerase chain reaction （PCr） for GFP DNA, immunofluorescence and morphometric studies were performed. resULTS： Transplanted GFP^＋ cells migrated to granuloma areas and reduced the percentage of liver fibrosis. The presence of donor-derived cells was confirmed by Fluorescence in situ hybridization （FISH） analysis for detection of cells bearing Y chromosome and by PCr analysis for detection of GFP DNA. The levels of TGF-β, a cytokine associated with fibrosis deposition, in liver fragments of mice submitted to therapy were reduced. The number of oval cells in liver sections of ***-infected mice increased 3-4 fold after transplantation. A partial recovery in albumin expression, which is decreased upon infection with ***, was found in livers of infected mice after cellular therapy. CONCLUSION： In conclusion, transplanted BMCs migrate to and reduce the damage of chronic fibrotic liver lesions caused by ***.

background:Leptospirosis morbidity and mortality rates in China have decreased since the *** analyses of the spatiotemporal and demographic changes occurring in the last decade and its implication on estimates of disease burden are required to inform intervention *** this study,we quantified the epidemiological shift and geographical heterogeneity in the burden of leptospirosis during 2005-2015 in ***:We used reported leptospirosis case data from 1st January 2005 to 31st of December 2015 that routinely collected by the China Information System for Disease Control and Prevention(CISDCP)to analyze the epidemiological trend and estimate the burden in terms of disability-adjusted life-years(DALYs)over space,time,and demographical ***:A total of 7763 cases were reported during *** which,2403(31%)cases were the laboratoryconfirmed *** 2005,the notified incidence rate was gradually decreased(Prelatively stable during 2011-2015(P>0.05).During 2005-2015,we estimated a total of 10313 DALYs were lost due to leptospirosis comprising a total of 1804 years-lived with disability(YLDs)and 8509 years-life lost(YLLs).Males had the highest burden of disease(7149 DALYs)compared to females(3164 DALYs).The highest burden estimate was attributed to younger individuals aged 10-19 years who lived in southern provinces of *** 2005-2015,this age group contributed to approximately 3078 DALYs corresponding to 30%of the total DALYs lost in ***,our analysis indicated a declining trend in burden estimates(Premained relatively low during *** burden estimates have been identified in the endemic regions where infections principally *** of the changes in DALY estimates were driven by changes in ***:In the last 11-years,the burden estimates of leptospirosis have shown a declining trend across the country;however,leptospirosis should not be neglected as it re

background： Beef cattle breeding programs in Brazil have placed greater emphasis on the genomic study of reproductive traits of males and females due to their economic importance. In this study, genome-wide associations were assessed for scrotal circumference at 210 d of age, scrotal circumference at 420 d of age, age at first calving, and age at second calving, in Canchim beef cattle. Data quality control was conducted resulting in 672,778 SNPs and 392 ***： Associated SNPs were observed for scrotal circumference at 420 d of age（435 SNPs）, followed by scrotal circumference at 210 d of age（12 SNPs）, age at first calving（six SNPs）, and age at second calving（four SNPs）. We investigated whether significant SNPs were within genic or surrounding regions. Biological processes of genes were associated with immune system, multicellular organismal process, response to stimulus, apoptotic process, cellular component organization or biogenesis, biological adhesion, and ***： Few associations were observed for scrotal circumference at 210 d of age, age at first calving, and age at second calving, reinforcing their polygenic inheritance and the complexity of understanding the genetic architecture of reproductive traits. Finding many associations for scrotal circumference at 420 d of age in various regions of the Canchim genome also reveals the difficulty of targeting specific candidate genes that could act on fertility; nonetheless,the high linkage disequilibrium between loci herein estimated could aid to overcome this issue. Therefore, al relevant information about genomic regions influencing reproductive traits may contribute to target candidate genes for further investigation of causal mutations and aid in future genomic studies in Canchim cattle to improve the breeding program.
