Organic and Inorganic Pigments from Commercial Inks Used in Eyebrows’ Microblading: Approach for Assessing the Safety Use by Bacterial Reverse Mutation Assay

Background and Objective: The aesthetic products’ safety should not be neglected to the detriment of the market. This study aimed to evaluate the mutagenic potential of commercial inks. It was formulated with organic (P1) and inorganic (P2) pigments for eyebrows microblading using a validated method by regulatory agencies, the Salmonella/microsome assay, to assure the safety of use. Methods: The assay was carried out in three steps: preliminary toxicity, medium without (-S9), and presence (+S9) of metabolic activation. The strains, auxotrophic to histidine (His-) TA97a, TA98, TA100, and TA102, were exposed to both types of inks, in triplicate, compared to negative and positive controls. Data were statistically analyzed, and values with mutagenic index ≥ 2.0 were indicative of mutagenicity. Results: The inks with organic (P1) and inorganic (P2) pigments were not toxic to TA98 and TA100 S. typhimurium tester strains, even at concentrations applied in humans. Both inks were not mutagenic either in the absence or presence of metabolic activation in the tested concentrations, including that applied in humans. The assay showed that P1 and P2 were not direct (-S9) or indirect (+S9) mutagens as commercially formulated. Conclusions: These results indicate that applying these inks on organisms with microsomal enzymes, including humans, is safe since no compound in the inks became more toxic to induce the bacterial reverse mutation.

Rilmenidine prevents blood pressure increase in rats with compromised nitric oxide production

AIM: To search tools of high blood pressure in the model of nitric oxide (NO)-defective hypertension, and the study focused on the effect of rilmenidine, agonist of imidazoline receptors, which was suggested to modulate central sympathetic outflow. METHODS: Three experimental groups, each consisting of 7 rats, were used: (I) rats with inhibition of NO synthase (NOS) by NG-nitro-L-arginine methyl ester (L-NAME) 40 mg·kg-1·d-1 for 4 weeks in drinking water, (II) rats with inhibited NOS as in group I , plus agonist of imidazoline receptors rilmenidine 3 mg·kg-1·d-1 for 4weeks by gavage, and(III) control rats. Systolic blood pressure wasmeasured weekly noninvasively. At the end of experiment aortic ring isometric tension was followed, NOS expression (aorta, left ventricle), and NOS activity (left ventricle and brain) were determined. RESULTS: In the group I systolic blood pressure in- creased significantly, aortic ring relaxation to acetylcholine was significantly attenuated. Rilmenidine administered simultaneously with L-NAME (group II) prevented the increase of blood pressure which did not differ significantly from control values; aortic ring relaxation to acetylcholine did not differ from control. No change in NOS expres- sion (aorta and left ventricle) was found in groups I and II. Significant decline in NOS activity (left ventricle and brain) wasfound in groups I and II. CONCLUSION: Rilmenidinehas aremarkable role inNO-defective hypertension, possibly by inhibiting central sympathetic outflow and by affecting receptors in vascular smooth muscle also. The prime cause of hypertension in this experimental model - the compromised production of NO due to inhibition of NOS - was not affected by rilmenidine.

Fault Location Approach to Distribution Networks Based on Custom State Estimator

This paper presents a properly designed branchcurrent based state estimator(BCBSE)used as the main core ofan accurate fault location approach(FLA)devoted to distribution *** to the approaches available in the literature,it uses only a limited set of conventional measurementsobtained from smart meters to accurately locate faults at busesor branches without requiring measurements provided by phasor measurement units(PMUs).This is possible due to themethods used to model the angular reference and the faultedbus,in addition to the proper choice of the weights in the stateestimator(SE).The proposed approach is based on a searchingprocedure composed of up to three stages:①the identificationof the faulted zones;②the identification of the bus closest tothe fault;and③the location of the fault itself,searching onbranches connected to the bus closest to the ***,this paper presents a comprehensive assessment of the proposedapproach,even considering the presence of distributed generation,and a sensitivity study on the proper weights required bythe SE for fault location purposes,which can not be found inthe *** show that the proposed BCBSE-basedFLA is robust,accurate,and aligned with the requirements ofthe traditional and active distribution networks.

Novel therapeutic strategies targeting mitochondria as a gateway in neurodegeneration

In recent years, multiple disciplines have focused on mitochondrial biology and contributed to understanding its relevance towards adult-onset neurodegenerative disorders. These are complex dynamic organelles that have a variety of functions in ensuring cellular health and homeostasis. The plethora of mitochondrial functionalities confers them an intrinsic susceptibility to internal and external stressors(such as mutation accumulation or environmental toxins), particularly so in long-lived postmitotic cells such as neurons. Thus, it is reasonable to postulate an involvement of mitochondria in aging-associated neurological disorders, notably neurodegenerative pathologies including Alzheimer’s disease and Parkinson’s disease. On the other hand, biological effects resulting from neurodegeneration can in turn affect mitochondrial health and function, promoting a feedback loop further contributing to the progression of neuronal dysfunction and cellular death. This review examines state-of-the-art knowledge, focus on current research exploring mitochondrial health as a contributing factor to neuroregeneration, and the development of therapeutic approaches aimed at restoring mitochondrial homeostasis in a pathological setting.

Ultrastructural Analysis of Chitosan Antibacterial Activity against Clinical Isolates of <i>Staphylococcus aureus</i>and <i>Escherichia coli</i>

The continuous search for an antimicrobial agent led to the identification of potential antimicrobial biomaterials based on polymers naturals, such as chitosan (CS). However, the mechanism of action of antibacterial activity of CS for gram-positive and gram-negative bacteria was not completely elucidated. The aim of this work is to report the antibacterial activity of CS through ultrastructural analyses of the clinical isolates Staphylococcus aureus and Escherichia coli by Transmission Electron Microscopy. The CS has a bactericidal action against S. aureus and E. coli which alters its cellular ultrastructure, such as with collapsed cell walls, condensed chromatin and the increase of intracellulares structures like vacuoles and cell debris. In this way, the CS represents a potential model for the future design of antibacterial in order to control bacterial resistance of patients in hospital settings.

Series solution of unsteady peristaltic flow of a Carreau fluid in small intestines

The study of peristaltic flow of a Carreau fluid in a non-uniform tube under the con- sideration of long wavelength is presented. The flow is investigated in a wave frame of reference moving with velocity of the wave e. Numerical integration has been used to obtain the graphical results for pressure rise and frictional forces. The effects of various emerging parameters are investigated through graphs.

Low-level laser therapy enhances the number of osteocytes in calvaria bone defects of ovariectomized rats

Background : Osteoporosis can make bone repair difficult. Low-level laser therapy( LLLT) has been shown to be a promising tool for bone neoformation. This study aimed to analyze the effect of LLLT on calvaria bone defects of ovariectomized rats using stereology. Methods : Fifty-four Wistar rats were subjected to bilateral ovariectomy, and bone defects were created in calvaria after 150 days. The animals were divided into nine groups(n = ?6 per group), and 24 hours after the bone defects were created they received three, six or 12 sessions of LLLT at 0, 20 or 30 J/cm 2, using a 780-nm low-intensity GaAlAs laser. One-way ANOVA followed by Tukey ' s post hoc test was used for data processing. A difference of P < 0.05 was considered statistically significant. The parameters evaluated were osteocyte density( Nv_(ost)), total osteocyte number( Nto ost), trabecular surface density( Sv_t), and trabecular surface area( Sa_t). Results : Data obtained showed that Nto ost, Sv t, and Sa t in group G2 rats were significantly different from G1(0 J/cm^2)( P < 0.05). Compared to group G4, G5 presented higher values for the parameters Sv t and Sa t, and G6 presented significantly higher values for almost all the analyzed parameters( Nv _(ost), Nto_(ost), Sv_t, and Sa t)( P < 0.05). Compared to group G7, G8 showed a higher value only for the parameter Sa t, and G9 showed significantly higher values for parameters Nv ost, Nto ost, Sv_t, and Sa_t. Conclusion : We conclude that LLLT stimulated bone neoformation and contributed to an increase in the total number of osteocytes, especially with a laser energy density of 30 J/cm^2 given for six and 12 sessions.

Region-dependent effects of diabetes and insulin-replacement on neuronal nitric oxide synthase-and heme oxygenase-immunoreactive submucous neurons

AIM To investigate the intestinal segment-specific effects of diabetes and insulin replacement on the density of different subpopulations of submucous neurons. METHODS Ten weeks after the onset of type 1 diabetes samples were taken from the duodenum, ileum and colon of streptozotocin-induce diabetic, insulin-treated diabetic and sex-and age-matched control rats. Whole-mount preparations of submucous plexus were prepared from the different gut segments for quantitative fluorescent immunohistochemistry. The following double-immunostainings were performed: neuronal nitric oxide synthase(n NOS) and Hu C/D, heme oxygenase(HO) 1 and peripherin, as well as HO2 and peripherin. The density of n NOS-, HO1-and HO2-immunoreactive(IR) neurons was determined as a percentage of the total number of submucous neurons. RESULTS The total number of submucous neurons and the proportion of n NOS-, HO1-and HO2-IR subpopulations were not affected in the duodenal ganglia of control, diabetic and insulin-treated rats. While the total neuronal number did not change in either the ileum or the colon, the density of nitrergic neurons exhibited a 2-and 3-fold increase in the diabetic ileum and colon, respectively, which was further enhanced after insulin replacement. The presence of HO1-and HO2-IR submucous neurons was robust in the colon of controls(38.4%-50.8%), whereas it was significantly lower in the small intestinal segments(0.0%-4.2%, P < 0.0001). Under pathophysiological conditions the only alteration detected was an increase in the ileum and a decrease in the colon of the proportion of HO-IR neurons in insulin-treated diabetic animals. CONCLUSION Diabetes and immediate insulin replacement induce the most pronounced region-specific alterations of n NOS-, HO1-and HO2-IR submucous neuronal density in the distal parts of the gut.

Tensile and fracture properties of an Mg-RE-Zn alloy at elevated temperatures

Magnesium alloy EZ10 （Mg-RE-Zn） was deformed in tension at temperatures from 20 up to 520℃. A rapid decrease of the yield and tensile strength with temperature was observed at temperatures higher than 300 ℃. On the other hand, ductility of sam-ples rapidly increased in the same temperature range. Light microscopy and scanning electron microscopy was used to reveal the rea-son for these behaviours. Intermetallic particles in grain boundaries are responsible for excellent mechanical properties at lower tem-peratures. Diffusional processes occurring at temperatures higher than 300 ℃ significantly influenced the deformation mechanism as well as the fracture character.

Using the Mini Nutritional Assessment to Evaluate the Profile of Elderly Patients in a Geriatric Outpatient Clinic and in Long-Term Institutions

INTRODUCTION: The Mini Nutritional Assessment (MNA) is a scale developed to evaluate the risk of malnutrition in senior citizens and to identify those who might benefit from early intervention. The objective of this study was to verify the nutritional profile of elderly individuals through the application of the MNA in three different locations: at the Geriatric outpatient clinic and two long-term institutions. METHODS: Through transversal observation study the MNA was applied to 90 elderly people of both genders over 60 years of age. The MNA version modified by Rubenstein et al. [4] and translated into Portuguese was used to evaluate the patients for this study. The descriptive analysis of variables is shown as the average ± one standard deviation (SD). RESULTS: At the AG, the risk of malnutrition was found to be 72.73% ± 3.77%, while 27.27% ± 3.77% of the patients were eutrophic;no malnourished elderly patients were found. At the ASVP, 15.38% ± 11.28% of elderly patients were found to be malnourished, 35.90% ± 15.10% were found to be at risk of malnutrition and 48.72% ± 15.72% were eutrophic. At the PMI, 42.50% ± 15.30% of the elderly patients were found to be malnourished, 25% ± 13.40% were at risk of malnutrition and 32.50% ± 14.50% were eutrophic. CONCLUSIONS: Due to the high prevalence of elderly patients that were malnourished or at risk of malnutrition, a more detailed evaluation of their nutritional states is required, as well as a regular follow up of those patients and a dietary intervention to reverse the situation.
