Optimal concentration of necrostatin-1 for protecting against hippocampal neuronal damage in mice with status epilepticus

Hippocampal neurons undergo various forms of cell death after status ***-1 specifically inhibits necroptosis mediated by receptor interacting protein kinase 1 (riP1) and riP3 ***,there are no reports of necroptosis in mouse models of status ***,in this study,we investigated the effects of necrostatin-1 on hippocampal neurons in mice with status epilepticus,and,furthermore,we tested different amounts of the compound to identify the optimal concentration for inhibiting necroptosis and apoptosis.A mouse model of status epilepticus was produced by intraperitoneal injection of kainic acid,12 mg/*** concentrations of necrostatin- 1 (10,20,40,and 80 μM) were administered into the lateral ventricle 15 minutes before kainic acid *** damage was assessed by hematoxylin-eosin staining 24 hours after the model was successfully *** deoxynucleotidyl transferase-mediated dUTP nick end labeling staining,western blot assay and immunohistochemistry were used to evaluate the expression of apoptosis-related and necroptosis-related ***-1 alleviated damage to hippocampal tissue in the mouse model of *** 40 μM concentration of necrostatin-1 significantly decreased the number of apoptotic cells in the hippocampal CA1 ***,necrostatin-1 significantly downregulated necroptosis-related proteins (MLKL,riP1,and riP3) and apoptosis-related proteins (cleaved-Caspase-3,Bax),and it upregulated the expression of anti-apoptotic protein *** together,our findings show that necrostatin-1 effectively inhibits necroptosis and apoptosis in mice with status epilepticus,with the 40 μM concentration of the compound having an optimal *** experiments were approved by the Animal Ethics Committee of Fujian Medical University,China (approval No.2016-032) on November 9,2016.

BACKGROUND Autophagy is associated with hippocampal injury following status epilepticus(SE)and is considered a potential therapeutic ***,an emerging multitherapeutic drug,has shown neuroprotective effects in patients with nervous system diseases due to its antioxidant *** To investigate the potential role of autophagy in LiCl-pilocarpine-induced *** The drugs were administered 30 min before *** staining showed that Baicalin attenuated hippocampal injury and reduced neuronal death in the *** blotting and terminal deoxynucleotidyl transferase dUTP nick end labeling assay confirmed that Baicalin reversed the expression intensity of cleaved caspase-3 and apoptosis in hippocampal CA1 following ***,western blotting and immunofluorescence staining were used to measure the expression of autophagy markers(p62/SQSTM1,Beclin 1,and LC3)and apoptotic pathway markers(cleaved caspase-3 and Bcl-2).RESULTS Baicalin significantly upregulated autophagic activity and downregulated mitochondrial apoptotic pathway ***,3-methyladenine,a commonly used autophagy inhibitor,was simultaneously administered to inhibit the Baicalin-induced autophagy,abrogating the protective effect of Baicalin on the mitochondrial apoptotic *** We illustrated that Baicalin-induced activation of autophagy alleviates apoptotic death and protects the hippocampus of SE rats.

Securing offshore resources development: A mathematical investigation into gas leakage in long-distance flexible pipes

Gas flexible pipes are critical multi-layered equipment for offshore oil and gas development. Under high pressure conditions, small molecular components of natural gas dissolve into the polymer inner liner of the flexible pipes and further diffuse into the annular space, incurring annular pressure build-up and/or production of acidic environment, which poses serious challenges to the structure and integrity of the flexible pipes. Gas permeation in pipes is a complex phenomenon governed by various factors such as internal pressure and temperature, annular structure, external temperature. In a long-distance gas flexible pipe, moreover, gas permeation exhibits non-uniform features, and the gas permeated into the annular space flows along the metal gap. To assess the complex gas transport behavior in long-distance gas flexible pipes, a mathematical model is established in this paper considering the multiphase flow phenomena inside the flexible pipes, the diffusion of gas in the inner liner, and the gas seepage in the annular space under varying permeable properties of the annulus. In addition, the effect of a variable temperature is accounted. A numerical calculation method is accordingly constructed to solve the coupling mathematical equations. The annular permeability was shown to significantly influence the distribution of annular pressure. As permeability increases, the annular pressure tends to become more uniform, and the annular pressure at the wellhead rises more rapidly. After annular pressure relief followed by shut-in, the pressure increase follows a convex function. By simulating the pressure recovery pattern after pressure relief and comparing it with test results, we deduce that the annular permeability lies between 123 and 512 m D. The results help shed light upon assessing the annular pressure in long distance gas flexible pipes and thus ensure the security of gas transport in the emerging development of offshore resources.

Experimental study on the effect of flexible joints of a deep-buried tunnel across an active fault under high in-situ stress conditions

During dislocation,a tunnel crossing the active fault will be damaged to varying degrees due to its permanent stratum *** previous studies did not consider the influence of the tunnel’s deep burial and the high in-situ stress,so the results were not entirely *** this paper,the necessity of solving the anti-dislocation problem of deep-buried tunnels is systemically *** the model test of tunnels across active faults,the differences in failures between deep-buried tunnels and shallow-buried tunnels were compared,and the dislocation test of deep-buried segmental tunnels was carried out to analyze the external stress change,lining strain,and failure mode of *** results are as follows.(1)The overall deformation of deep-buried and shallow-buried tunnels is both *** failure mode of deep-buried tunnels is primarily characterized by shear and tensile failure,resulting in significant compressive deformation and a larger damaged *** contrast,shallow-buried tunnels mainly experience shear failure,with the tunnel being sheared apart at the fault crossing,leading to more severe damage.(2)After the segmental structure design of the deep-buried tunnel,the‘‘S”deformation pattern is transformed into a‘‘ladder”pattern,and the strain of the tunnel and the peak stress of the external rock mass are reduced;therefore,damages are significantly mitigated.(3)Through the analysis of the distribution of cracks in the tunnel lining,it is found that the tunnel without a segmental structure design has suffered from penetrating failure and that cracks affect the entire *** cracks in a flexible segmental tunnel affect about 66.6%of the entire length of the tunnel,and cracks in a tunnel with a short segmental tunnel only affect about 33.3%of the entire length of the ***,a deep-buried tunnel with a short segmental tunnel can yield a better anti-dislocation effect.(4)By comparing the shallow-buried segmental tunnel in previous

An analytical transient coal permeability model:Sorption non-equilibrium index-based swelling switch

Long-term permeability experiments have indicated that sorption-induced swelling can switch from internal to bulk depending on the evolutive sorption ***,this sorption swelling switch mechanism has not been considered in current analytical permeability *** study introduces a normalized sorption non-equilibrium index(SNEI)to characterize the sorption status,quantify the dynamical variations of matrix swelling accumulation and internal swelling partition,and formulate the sorption swelling switch *** incorporation of this index into the extended total effective stress concept leads to an analytical transient coal permeability *** results show that the sorption swelling switch itself results in the permeability switch under stress-constrained conditions,while the confined bulk swelling suppresses the permeability recovery to the continuous reduction under displacement-constrained *** verifications show that current experimental observations correspond to the early stages of the transient process,and they could be extended to the whole process with these *** study demonstrates the importance of the sorption swelling switch in determining permeability evolution using simple boundary *** provides new insights into experimentally revealing the sorption swelling switch in the future,and underscores the requirement of a rigorous model for complex coupled processes in large-scale coal seams.

To improve the wear performance of CoCrAlYTa coating,part of the carbon nanotubes(CNTs)chemically reacted with Ta to form reinforcement phase(TaC),while the other CNTs were retained as lubrication ***,the CoCrAlYTa-xCNTs(x=0,1,2,and 4;wt%)composite coatings were prepared by laserinduction hybrid cladding(LIHC),and the microstructure and wear resistance of coatings were systematically *** show that the coatings are mainly composed of TaC,y-(Co,Cr)andβ-(Co,Cr)*** the CNTs content increases from 0 wt%to 4 wt%,the volume fraction of TaC increases from 13.11 vol%to 16.12 vol%.Meanwhile,the nano-hardness ofγ-(Co,Cr)andβ-(Co,Cr)Al are improved from 7.49 and 9.72 to 9.36 and 11.19 GPa,*** a result,the microhardness of coating increases from HV536.25 to HV 738.16,the wear rate decreases from32.4×10^(-3)to 6.1×10^(-3)mg·m^(-1),and the average friction coefficient decreases from 0.55 to *** good wear performance of the coating is attributed to the formation of TaC and the existence of remained CNTs lubricant film.

Attaining synergetic equilibrium of electrical conductivity and tensile strength in GQDs@GN/Cu composites through multi-scale intragranular and intergranular reinforcements

The configuration and quality of reinforcements, as well as the robustness of interfacial bonding,holding a critical significance in determining the concurrence between electrical conductivity and mechanical strength in metal matrix composites. In this study, citric acid was employed as the precursor for synthesizing multiscale carbon nanomaterials(graphene quantum dots and graphene, abbreviated as GQDs and GN). The GQDs@GN/Cu composites were fabricated through a segmented ball milling process in conjunction with subsequent spark plasma sintering(SPS). The intragranular GQDs and intergranular GQDs@GN had synergistically reinforced Cu composites through Orowan strengthening, load transfer strengthening and refinement strengthening. Furthermore,the robust interface bonding between GQDs@GN and Cu effectively mitigated interfacial impedance stemming from electron-boundary scattering. The yield strength and ultimate tensile strength of the GQDs@GN/Cu composites were recorded as 270 and 314 MPa, respectively, representing an improvement of 92 and 28% over pure Cu, while maintaining electrical conductivity at a level comparable to that of pure Cu. This study advances the understanding of the possibility of realizing a synergistic compatibility between electrical conductivity and mechanical strength in Cu composites.

A low-melting-point metal doping strategy for the synthesis of small-sized intermetallic Pt_(5)Ce fuel cell catalysts

Carbon-supported platinum-lanthanum(Pt-Ln)intermetallic compound(IMC)nanoparticles with high activity and robust stability have been demonstrated as promising cathode catalysts for proton-exchange membrane fuel ***,the preparation of Pt-Ln IMC catalysts needs high-temperature annealing treatment that inevitably causes nanoparticle sintering,resulting in significant reduction of the electrochemical surface area and mass-based ***,we prepare small-sized M-doped Pt_(5)Ce(M=Ga,Cd,and Sb)IMCs catalysts via a low-melting-point metal doping *** speculate that the doping of low-melting-point metals can facilitate the generation of vacancies in the crystal lattice through thermal activation and thus reduce the kinetic barriers for the formation of intermetallic Pt_(5)Ce *** prepared Ga-doped Pt_(5)Ce catalyst exhibits a higher electrochemical active surface area(81 m^(2)·gPt^(-1))and a larger mass activity(0.45 A·mgPt^(-1)at 0.9 V)over the undoped Pt_(5)Ce and commercial Pt/C *** the membrane electrode assembly test,the Ga-doped Pt_(5)Ce cathode delivers a power density of 0.98 W·cm^(-2)at 0.67 V,along with a voltage loss of only 27 mV at 0.8 A·cm^(-2)at the end of accelerated stability test.

Full sexual maturity-cycle exposure to environmentally relevant concentrations of 17β-estradiol decreases reproductive capacity of zebrafish

17β-Estradiol(E2)has been widely detected in natural water and treatment with E2 induces potential endocrine disrupting effects in ***,effects on fish fecundity and steroid system after treatment with environmentally relevant concentrations of E2 for the full sexual maturation cycle remain *** this study,zebrafish were treatment with 0,10 or100 ng/L E2 from embryo to adult stage,and effects on gonadal development and differentiation,steroid hormone levels,transcription of genes associated with the hypothalamic-pituitary-gonadal-liver(HPG)axis in adults and fertilization rate of offspring were *** results showed that treatment with E2 lead to increased number of feminization in *** females,E2 decreased cumulative amount of spawning and inhibited the maturation of *** males,E2 inhibited the maturation and motility of sperm,as well as decreased the movement speed of *** adverse effects on sperm might be responsible for the reduced fertilization observed in *** addition,treatment with E2 changed the levels of steroid hormones in zebrafish gonad and altered the transcriptional levels of genes associated with HPG axis,which is responsible for the regulation of germ cells maturation and gonadal development in ***,these results suggested that treatment with environmentally relevant concentrations of E2 for the full sexual maturity cycle resulted in adverse effects on reproduction in zebrafish.
