Celecoxib attenuates 5-fluorouracil-induced apoptosis in HCT-15 and HT-29 human colon cancer cells

AIM： To investigate the combined chemotherapeutic effects of celecoxib when used with 5-FU in vitro. METHODS： Two human colon cancer cell lines （HCT-15 and HT-29） were treated with 5-FU and celecoxib, alone and in combination. The effects of each drug were evaluated using the MTT （3-（4, 5-dimethylthiazol-2-yl）-2, 5-diphenyltetrazolium bromide） assay, flow cytometry, and western blotting. RESULTS： 5-FU and celecoxib showed a dosedependent cytotoxic effect. When treated with 10-3 mol/L 5-FU （IC50） and celecoxib with its concentration ranging from 10.8 mol/L to 10.4 mol/L of celecoxib, cells showed reduced cytotoxic effect than 5-FU （10.3 mol/L） alone. Flow cytometry showed that celecoxib attenuated 5-FU induced accumulation of cells at subG1 phase. Western blot analyses for caspase-3 and poly （ADP-ribose） polymerase （PARP） cleavage showed that celecoxib attenuated 5-FU induced apoptosis. Western blot analyses for cell cycle molecules showed that G2/M arrest might be possible cause of 5-FU induced apoptosis and celecoxib attenuated 5-FU induced apoptosis via blocking of cell cycle progression to the G2/M phase, causing an accumulation of cells at the GI/S phase. CONCLUSION： We found that celecoxib attenuated cytotoxic effect of 5-FU. Celecoxib might act via inhibition of cell cycle progression, thus preventing apoptosis induced by 5-FU.

To understand and engineer plant metabolism, we need a comprehensive and accurate annotation of all metabolic information across plant species. As a step towards this goal, we generated genome-scale metabolic pathway databases of 126 algal and plant genomes, ranging from model organisms to crops to medicinal plants(https://***).Of these, 104 have not been reported *** systematically evaluated the quality of the databases, which revealed that our semi-automated validation pipeline dramatically improves the quality. We then compared the metabolic content across the 126 organisms using multiple correspondence analysis and found that Brassicaceae,Poaceae, and Chlorophyta appeared as metabolically distinct groups. To demonstrate the utility of this resource, we used recently published sorghum transcriptomics data to discover previously unreported trends of metabolism underlying drought tolerance. We also used single-cell transcriptomics data from the Arabidopsis root to infer cell typespecific metabolic pathways. This work shows the quality and quantity of our resource and demonstrates its wide-ranging utility in integrating metabolism with other areas of plant biology.

Cancer stem cells(CSCs) are maintained by theirsomatic stem cells and are responsible for tumor initiation, chemoresistance, and metastasis. Evidence for the CSCs existence has been reported for a number of human cancers. The CSC mitochondria have been shown recently to be an important target for cancer treatment, but clinical significance of CSCs and their mitochondria properties remain unclear. Mitochondriatargeted agents are considerably more effective compared to other agents in triggering apoptosis of CSCs, as well as general cancer cells, via mitochondrial dysfunction. Mitochondrial metabolism is altered in cancer cells because of their reliance on glycolytic intermediates, which are normally destined for oxidative phosphorylation. Therefore, inhibiting cancer-specific modifications in mitochondrial metabolism, increasing reactive oxygen species production, or stimulating mitochondrial permeabilization transition could be promising new therapeutic strategies to activate cell death in CSCs as well, as in general cancer cells. This review analyzed mitochondrial function and its potential as a therapeutic target to induce cell death in CSCs. Furthermore, combined treatment with mitochondriatargeted drugs will be a promising strategy for the treatment of relapsed and refractory cancer.

For more than 20 years nuclear physicists have used the GEANT code to simulate particle-matter interaction. In most recent version, GEANT4 is a toolkit for simulating the passage of particles though matter, which contains a complete range of functionality including tracking, geometry, physics models, and hits. In this article, an attempt to use GEANT4 to model a double-gap resistive plate chamber （RPC） with its improved efficiency is presented. The efficiencies of the double-gap RPC have been evaluated as a function of gamma energy range 0.005-1000MeV. A comparison to available previous simulation package GEANT3 data is also performed.

Background There are 103 million displaced people worldwide,41%of whom are *** on the provision of surgery in humanitarian settings are *** scarcer is literature on pediatric surgery performed in humanitarian settings,particularly protracted humanitarian *** We reviewed patterns,procedures,and indications for pediatric surgery among children in Nyarugusu Refugee Camp using a 20-year retrospective *** A total of 1221 pediatric surgical procedures were performed over the study *** between the ages of 12 and 17 years were the most common age group undergoing surgery(n=991;81%).A quarter of the procedures were performed on local Tanzanian children seeking care in the camp(n=301;25%).The most common procedures performed were cesarean sections(n=858;70%),herniorrhaphies(n=197;16%),and exploratory laparotomies(n=55;5%).Refugees were more likely to undergo exploratory laparotomy(n=47;5%)than Tanzanian children(n=7;2%;p=0.032).The most common indications for exploratory laparotomy were acute abdomen(n=24;44%),intestinal obstruction(n=10;18%),and peritonitis(n=9;16%).Conclusions There is a significant volume of basic pediatric general surgery performed in the Nyarugusu *** are used by both refugees and local *** hope this research will inspire further advocacy and research on pediatric surgical services in humanitarian settings worldwide and illuminate the need for including pediatric refugee surgery within the growing global surgery movement.

The CarbonTracker(CT) model has been used in previous studies for understanding and predicting the sources, sinks, and dynamics that govern the distribution of atmospheric CO_2 at varying ranges of spatial and temporal scales. However, there are still challenges for reproducing accurate model-simulated CO_2 concentrations close to the surface, typically associated with high spatial heterogeneity and land cover. In the present study, we evaluated the performance of nested-grid CT model simulations of CO_2 based on the CT2016 version through comparison with in-situ observations over East Asia covering the period 2009–13. We selected sites located in coastal, remote, inland, and mountain areas. The results are presented at diurnal and seasonal time periods. At target stations, model agreement with in-situ observations was varied in capturing the diurnal cycle. Overall, biases were less than 6.3 ppm on an all-hourly mean basis, and this was further reduced to a maximum of 4.6 ppm when considering only the daytime. For instance, at Anmyeondo, a small bias was obtained in winter, on the order of 0.2 ppm. The model revealed a diurnal amplitude of CO_2 that was nearly flat in winter at Gosan and Anmyeondo stations, while slightly overestimated in the summertime. The model's performance in reproducing the diurnal cycle remains a challenge and requires improvement. The model showed better agreement with the observations in capturing the seasonal variations of CO_2 during daytime at most sites, with a correlation coefficient ranging from 0.70 to 0.99. Also, model biases were within-0.3 and 1.3 ppm, except for inland stations(7.7 ppm).

Experimental Study for the Determination of the Material Diameter of the Riprap Bed Protection

Bed protection in apron downstream was installed to use riprap or gabion mattress. In the case of bed protection using riprap, the decision on the riprap diameter is very important because riprap diameter means the capacity to withstand the flow. Initial formulas for the decision on the riprap diameter applied velocity and weight as main factors. Since the main factors gradually expanded to depth, bed slope, and turbulence intensity, decision formulas for the riprap diameter were detailed. Because turbulence intensity was considered to be a main factor of the formula, the decision formulas for the riprap diameter could be expanded to apply to bed protection around hydraulic structures. Escarameia and May conducted a study on the riprap diameter of a strong turbulence area around the downstream of hydraulic structures. For the formula of Escarameia and May, the basis of the formula was the Izbash type, and the main factor was turbulence intensity. This study was an experimental study for deciding the riprap diameter installed around apron downstream and was based on the study results of Escarameia and May. The experiment measured the velocity upon change of discharge and riprap diameter to the installed weir model and analyzed the correlation of threshold velocity by diameter. The experimental formula in this study expanded the turbulence intensity limits of the Escarameia and May formula and increased application to turbulence intensity in weir downstream.
