Identification and immune analysis of antimicrobial peptides from the cigarette beetle(Lasioderma serricorne)

Antimicrobial peptides(AMPs)in insects are endogenous peptides that are effector components of the innate defense system of the *** may serve as antimicrobial agents because of their small molecular weight and broad-spectrum antimicrobial *** this study,we performed transcriptome analysis of cigarette beetle(Lasioderma serricorne)larvae,parasitized by the ectoparasitic wasp,Anisopteromalus *** AMP genes were significantly upregulated following *** parasitism,postulating the hypothesis that the parasitization enhanced the host's resistance against pathogenic microorganisms through the regulation of host AMP ***,3 AMP genes(LsDef1,LsDef2,and LsCole)were significantly upregulated and we studied their immune function in *** challenge and functional analysis showed that LsCole was responsible for the immune response against Gram-negative and Gram-positive bacteria,while LsDef1 and LsDef2 were involved in insect defense against Gram-positive *** recombinant LsCole exhibited antimicrobial activities against the Gram-negative bacterium Escherichia coli and the Gram-positive bacterium Staphylococcus ***2 showed an antibacterial effect against *** and LsDef2 exhibited antibiofilm activity against *** 2 AMPs disrupted cell membranes and caused leakage of *** cell *** results indicated that the 3 AMPs in *** are involved in the innate immunity of this pest *** AMPs may have potential as antimicrobial agents for bacterial infection ***,data are discussed in relation to new control strategies with greater biosafety against pest insects with use of microbial biocontrol agents in combination with RNA interference against the insect's defensive AMP genes.

Maximizing conditional independence for unsupervised domain adaptation

Unsupervised domain adaptation(UDA) studies how to transfer a learner from a labeled source domain to an unlabeled target domain with different distributions. Existing methods mainly focus on matching marginal distributions of the source and target domains, which probably leads to a misalignment of samples from the same class but different domains. In this paper, we tackle this misalignment issue by achieving the class-conditioned transferring from a new perspective. Specifically, we propose a method named maximizing conditional independence(MCI) for UDA, which maximizes the conditional independence of feature and domain given class in the reproducing kernel Hilbert spaces. The optimization of conditional independence can be viewed as a surrogate for minimizing class-wise mutual information between feature and domain. An interpretable empirical estimation of the conditional dependence measure is deduced and connected with the unconditional case. Besides, we provide an upper bound on the target error by taking the class-conditional distribution into account, which provides a new theoretical insight for class-conditioned transferring. Extensive experiments on six benchmark datasets and various ablation studies validate the effectiveness of the proposed model in dealing with UDA.

Sliding behaviors of the trapezoidal roof rock block under a lateral dynamic disturbance

The surrounding rock of underground space is always affected by external dynamic disturbance from the side position,such as blasting vibration from a stope at the same level or seismic waves from adjacent strata.A series of laboratory tests,numerical simulations and theoretical analyses were carried out in this study to disclose the sliding mechanism of roof rock blocks under lateral ***,the experiments on trapezoidal key block under various clamping loads and disturbance were conducted,followed by numerical simulations using the fast Lagrangian analysis of continua(FLAC3D).Then,based on the conventional wave propagation model and the classical shear-slip constitutive model,a theoretical model was proposed to capture the relative displacement between blocks and the sliding displacement of the key *** results indicate that the sliding displacement of the key block increased linearly with the disturbance energy and decreased exponentially with the clamping load when the key block was disturbed to slide(without instability).Meanwhile,when the key block was disturbed to fall,two types of instability process may appear as immediate type or delayed *** addition,the propagation of stress waves in the block system exhibited obvious low-velocity and lowfrequency characteristics,resulting in the friction reduction effect appearing at the contact interface,which is the essential reason for the sliding of rock *** results can be applied to practical underground engineering and provide valuable guidance for the early detection and prevention of rockfalling disasters.

Effects of acid mine drainage on photochemical and biological degradation of dissolved organic matter in karst river water

Dissolved organic matter(DOM)can be removed or transformed by photochemical and biological processes,producing the negative effect of transforming organic carbon into inorganic carbon,which plays a vital role in the karst carbon ***,acid mine drainage(AMD)will affect this process,so the degradation of DOM in karst river water(KRW)needs to be studied in this *** this study,to reveal the evolution processes of DOM under photochemical and biological conditions in AMD-impacted KRW,AMD and KRW were mixed in different ratios under conditions of visible light irradiation(VL),biodegradation(BD),ultraviolet irradiation(UV)and ultraviolet irradiation+biodegradation(UV+BD).The average DOC concentrations in samples after mixing AMD and KRW in different proportions decreased significantly(by 23%)in UV+BD,which was 1.2–1.4 times higher than under the other conditions and would lead to a significant release of inorganic *** analysis of the fluorescence parameters via parallel factor analysis(PARAFAC)revealed that the DOM fluorescence components in AMD comprised mainly protein-like substances derived from autochthonous components,while the DOM fluorescence components in KRW were mainly humic-like substances with both autochthonous and allochthonous ***,AMD could promote both the photochemical and biological degradation of DOM in karst receiving streams,resulting in the conversion of DOC to inorganic *** results showed that the synergistic effects of UV+BD and AMD accelerated the degradation of DOM and the release of inorganic carbon in KRW,thus affecting the stability of the karst carbon cycle.

Negative capacitance(NC)has the potential to enable low power microelectronics beyond the fundamental thermionic limit,and it has been theorized that the thermodynamically unstable NC of ferroelectrics can be stabilized by linear dielectric,making negative capacitance ferroelectric field effect transistors(NC-FeFET)***,the validity of NC as a physical concept for ferroelectrics remain contentious despite numerous theoretical and experimental investigations,and the intrinsic ferroelectric NC with suppressed polarization has not been demonstrated except locally at vortex *** NC-FeFET with subthreshold swing(SS)lower than 60 mV/dec limit has been reported,such device characteristics has not been directly connected to suppressed polarization at materials’level,and alternative mechanisms other than NC have also been *** we demonstrate stable sub-60 mV/dec SS with hysteresis free Isingle bondV in NC-FeFET based on SrTiO_(3)/Pb(Zr_(0.1)Ti_(0.9))O_(3)/SrTiO_(3) heterostructure,and observe its suppressed polarization at both macroscopic and microscopic *** intrinsic ferroelectric NC thus is experimentally confirmed and directly connected to NC-FeFET performance,and the mica-based device is also highly flexible and robust under cyclic bending as well as extended heating.

A feature extraction framework for discovering pan-cancer driver genes based on multi-omics data

The identification of tumor driver genes facilitates accurate cancer diagnosis and treatment,playing a key role in precision oncology,along with gene signaling,regulation,and their interaction with protein *** tackle the challenge of distinguishing driver genes from a large number of genomic data,we construct a feature extraction framework for discovering pan-cancer driver genes based on multi-omics data(mutations,gene expression,copy number variants,and DNA methylation)combined with protein–protein interaction(PPI)*** a network propagation algorithm,we mine functional information among nodes in the PPI network,focusing on genes with weak node information to represent specific cancer *** these functional features,we extract distribution features of pan-cancer data,pan-cancer TOPSIS features of functional features using the ideal solution method,and SetExpan features of pan-cancer data from the gene functional features,a method to rank pan-cancer data based on the average inverse *** features represent the common message of ***,we use the lightGBM classification algorithm for gene *** results show that our method outperforms existing methods in terms of the area under the check precision-recall curve(AUPRC)and demonstrates better performance across different PPI *** indicates our framework’s effectiveness in predicting potential cancer genes,offering valuable insights for the diagnosis and treatment of tumors.

Defects in phospholipase C zeta cause polyspermy and low fertilization after conventional IVF:not just ICSI failure

Phospholipase C zeta(PLC)is a key sperm-borne oocyte-activating factor that triggers Ca^(2+)oscillations and the subsequent block to polyspermy following gamete *** in PLCZ1,the gene encoding PLCζ,cause male infertility and intracytoplasmic sperm injection(ICSI)fertilization failure;and PLCζ expression and localization patterns are significantly correlated with ICSI fertilization rate(FR).However,in conventional in vitro fertilization(cIVF),whether and how sperm PLCζ affects fertilization remain ***,we identified one previously reported and two novel PLCZ1 mutations associated with polyspermy in vitro that are characterized by excessive sperm-zona binding and a delay in pronuclei(PN)*** staining and oocyte activation testing revealed that virtually all spermatozoa from patients lacked functional PLCζ and were thus unable to evoke Ca^(2+) *** with an artificial oocyte activation treatment successfully rescued the polyspermic phenotype and resulted in a live ***,we analyzed PLCζ in an additional 58 males after cIVF treatment in the Reproductive and Genetic Hospital of CiTiC-Xiangya(Changsha,China)between February 2019 and January *** found that the proportion of spermatozoa that expressed PLCζ was positively correlated with both 2PN rate and total *** optimal cutoff value below which males were likely to experience low FR(total FR≤30%)after clVF was 56.7%for the proportion of spermatozoa expressing *** study expands the mutation and the phenotypic spectrum of PLCZ1 and further suggests that PLCζ constitutes a promising biomarker for identifying low FRs cases in cIVF due to sperm-related oocyte activation deficiency and that sperm PLCζ analysis may benefit the widermale population and not onlymen with IcsI failure.

Molecular Mechanisms of Intracellular Delivery of Nanoparticles Monitored by an Enzyme‑Induced Proximity Labeling

Achieving increasingly finely targeted drug delivery to organs,tissues,cells,and even to intracellular biomacromolecules is one of the core goals of *** the delivery destination is refined to cellular and subcellular targets,it is essential to explore the delivery of nanomedicines at the molecular ***,due to the lack of technical methods,the molecular mechanism of the intracellular delivery of nanomedicines remains unclear to ***,we develop an enzyme-induced proximity labeling technology in nanoparticles(nano-EPL)for the real-time monitoring of proteins that interact with intracellular ***(lactic-co-glycolic acid)nanoparticles coupled with horseradish peroxidase(HRP)were fabricated as a model(HRP(+)-PNPs)to evaluate the molecular mechanism of nano delivery in *** adding the labeling probe biotin-phenol and the catalytic substrate H_(2)O_(2)at different time points in cellular delivery,nano-EPL technology was validated for the real-time in situ labeling of proteins interacting with ***-EPL achieves the dynamic molecular profiling of 740 proteins to map the intracellular delivery of HRP(+)-PNPs in macrophages over *** on dynamic clustering analysis of these proteins,we further discovered that different organelles,including endosomes,lysosomes,the endoplasmic reticulum,and the Golgi apparatus,are involved in delivery with distinct participation *** importantly,the engagement of these organelles differentially affects the drug delivery efficiency,reflecting the spatial–temporal heterogeneity of nano delivery in *** summary,these findings highlight a significant methodological advance toward understanding the molecular mechanisms involved in the intracellular delivery of nanomedicines.

Landscape ecological risk assessment and its driving factors in the Weihe River basin,China

Weihe River basin is of great significance to analyze the changes of land use pattern and landscape ecological risk and to improve the ecological basis of regional *** on land use data of the Weihe River basin in 2000,2010,and 2020,with the support of Aeronautical Reconnaissance Coverage Geographic Information System(ArcGIS),GeoDa,and other technologies,this study analyzed the spatial-temporal characteristics and driving factors of land use pattern and landscape ecological *** showed that land use structure of the Weihe River basin has changed significantly,with the decrease of cropland and the increase of forest land and construction *** the past 20 a,cropland has decreased by 7347.70 km2,and cropland was mainly converted into forest land,grassland,and construction *** fragmentation and dispersion of ecological landscape pattern in the Weihe River basin were improved,and land use pattern became more ***,landscape ecological risk of the Weihe River basin has been *** landscape ecological risk area decreased by 19,177.87 km2,high landscape ecological risk area decreased by 3904.35 km2,and moderate and low landscape ecological risk areas continued to *** is worth noting that landscape ecological risks in the upper reaches of the Weihe River basin are still relatively serious,especially in the contiguous areas of high ecological risk,such as Tianshui,Pingliang,Dingxi areas and some areas of Ningxia Hui Autonomous *** ecological risk showed obvious spatial dependence,and high ecological risk area was *** the driving factors,population density,precipitation,normalized difference vegetation index(NDVI),and their interactions are the most important factors affecting the landscape ecological risk of the Weihe River *** findings significantly contribute to our understanding of the ecological dynamics in the Weihe River basin,providing crucial insights for sustainable mana
