建立大熊猫的精子库,可以进行远距离圈养大熊猫种群间的人工授精和遗传物质的转运,以维持遗传多样性,这是目前大熊猫遗传管理的优先方法。要成为最有效的工具,精子库保存的精子解冻后的活力必须是最好的。本项研究包括:1)不同解冻速度对解冻后精子的影响;2)使用化学试剂Pentyoxyfilline(PF)对解冻后精子活率和活力的影响。试验用的精液采自11只成年大熊猫,精液冷冻速度为-40--100℃/min。试验口:把冷冻精液放入3种不同温度的水浴中解冻:(1)22℃(慢速解冻);(2)37℃(中速解冻)和(3)50℃(快速解冻)。把冷冻前(78.1±2.9%)和解冻后的平均精子活力进行比较,快速解冻后的精子活力(57.5±5.4%)显著地降低(P0.05)。在1mMPF中孵育24h的精子活力没有变化(P>0.05)。在5和10mM PF中孵育过的精子活力(5mM:24.0±4.7;10mM 19.5±3.6%)比没有加PF的对照组的精子活力(38.3±5.2%)显著地低(P<0.05)。而且,在10mM PF孵育4小时的精子的顶体正常率(56.3±2.8)也比对照组的顶体正常率低(71.0±3.3%P<0.05)。这两个大熊猫精子的试验表明大熊猫精子通过快速冷冻制成的颗粒冻精在解冻时用慢速解冻的效果更好。虽然,大熊猫精子对常用激活剂PF是敏感的,但激活是短暂的,几小时后反而会降低精子的活力,在高浓度下还会损坏精子的顶体。

Homogeneous Distribution of <i>escherichia coli</i>Measured within the Vertical Water Column of Small, Freshwater Streams

Sampling for coliform bacterial indicators such as escherichia coli (e. coli) provides a universally accepted gauge of the microbiologic quality of fresh surface waters worldwide. Protocols for the capture, preservation, and analysis of indicator bacteria collected from moving waters (e.g., rivers, streams, canals, etc.) parallel those for collecting bacteria from standing waters (e.g., ponds, lakes, and impoundments). Strict depth- and width-integrated rules established for testing moving waters are likely a result of the historical precedence of our knowledge of bacterial stratification in standing waters. Sampling protocols for indicator bacteria in freshwater streams recommend capture and retrieval of samples from the mid-water column directed into the current and within the deepest portion of the channel to prevent collection of either benthic particles or surface films. Chi-square analyses of multiple stratified samples captured on the same date and time reveal that variability in sampling position at specified depths within the main stream column or within randomly chosen locations within the main stream channel has no effect (p ≥ 0.25) upon such indicator bacteria numbers. Additionally, these data are the first to show that concentrations of the common bacterial indicator, e. coli, are homogeneously distributed throughout both lateral area and vertical water column within/near a single sampling location of a moving water body up to 245 cubic feet per second (cfs) discharge. Moreover, one data point (bacterial sample) appears to represent the overall bacterial concentration of a small freshwater stream obtained from any single sampling location within/near the main channel for a given date and time. These findings suggest some latitude in sampling strategies for assessing small freshwater streams for indicator bacteria such as e. coli for workers in both environmental and public health fields.

Baseline Clinical Profile of a “Normal Popu-lation” of HIV exposed Sero-Negative (HeSN) Partners May Impact HIV Biomedical Prevention Studies Negatively

The burden and viral diversity of HIV in Nigeria makes it suitable to evaluate biomedical prevention strategies including HIV vaccines. We document baseline clinical characteristics of a cohort of HIV exposed Sero-Negative (HeSN) partners to highlight background morbidities that might impact the interpretation of research findings especially in low income countries where “normal control” is based on physical appearance and self-reporting. We established a 2-year prospective cohort of HeSN, and obtained, at baseline medical history, general physical examination findings and safety laboratory tests results. Among 534 participants with a mean age of 37 ± 9 years, the commonest symptoms were headache (25.5%) and fever (20%), which occurred more among females. Also, 13% reported a history of urethral/vaginal discharge and genital ulcer. High blood pressure (HBP) (i.e. blood pressure ≥ 140/90 mmhg) was a major abnormal examination finding, which occurred more among males (53.2% vs 26.2%, p e female participants had abnormal laboratory results with 31% having low hemoglobin concentration (elevated alanine transaminase level (p = 0.019) and 28% with abnormal blood urea nitrogen level (p = 0.093). HBP and abnormal safety laboratory are significant findings among so called “normal population” that could affect interpretation of research findings of HIV biomedical prevention studies in Nigeria.

Background： Canopy structure, defined by leaf area index （LAI）, fractional vegetation cover （FCover） and fraction of absorbed photosynthetically active radiation （fAPAR）, regulates a wide range of forest functions and ecosystem services. Spatially consistent field-measurements of canopy structure are however lacking, particularly for the tropics. Methods： Here, we introduce the Global LAI database： a global dataset of field-based canopy structure measurements spanning tropical forests in four continents （Africa, Asia, Australia and the Americas）. We use these measurements to test for climate dependencies within and across continents, and to test for the potential of anthropogenic disturbance and forest protection to modulate those dependences. Results： Using data collected from 887 tropical forest plots, we show that maximum water deficit, defined across the most arid months of the year, is an important predictor of canopy structure, with all three canopy attributes declining significantly with increasing water deficit. Canopy attributes also increase with minimum temperature, and with the protection of forests according to both active （within protected areas） and passive measures （through topography）. Once protection and continent effects are accounted for, other anthropogenic measures （e.g. human population） do not improve the model. Conclusions： We conclude that canopy structure in the tropics is primarily a consequence of forest adaptation to the maximum water deficits historically experienced within a given region. Climate change, and in particular changes in drought regimes may thus affect forest structure and function, but forest protection may offer some resilience against this effect.

Using quality improvement methods to increase use of pain prevention strategies for childhood vaccination
