Development of a panel of unigene-derived polymorphic EST–SSR markers in lentil using public database information

Lentil(Lens culinaris Medik.), a diploid(2n = 14) with a genome size greater than 4000 Mbp, is an important cool season food legume grown worldwide. The availability of genomic resources is limited in this crop species. The objective of this study was to develop polymorphic markers in lentil using publicly available curated expressed sequence tag information(ESTs). In this study, 9513 ESTs were downloaded from the National Center for Biotechnology Information(NCBI) database to develop unigene-based simple sequence repeat(SSR) markers. The ESTs were assembled into 4053 unigenes and then analyzed to identify 374 SSRs using the MISA microsatellite identification tool. Among the 374 SSRs, 26 compound SSRs were *** pairs for these SSRs were designed using Primer3 version 1.14. To classify the functional annotation of ESTs and EST–SSRs, BLASTx searches(using E-value 1 × 10-5) against the public UniP rot(http://***/) and NCBI(http://***/) databases were performed. Further functional annotation was performed using PLAZA(version3.0) comparative genomics and GO annotation was summarized using the Plant GO slim category. Among the synthesized 312 primers, 219 successfully amplified Lens DNA. A diverse panel of 24 Lens genotypes was used to identify polymorphic markers. A polymorphic set of 57 markers successfully discriminated the test genotypes. This set of polymorphic markers with functional annotation data could be used as molecular tools in lentil breeding.

Pregnane X receptor(PXR)is a ligand-activated nuclear receptor that transcriptionally upregulates drug-metabolizing enzymes[e.g.,cytochrome P4503A4(CYP3A4)]and *** the regulation of PXR target genes is well-characterized,less is known about the regulation of PXR protein *** screening an RNAi library,we identified the F-box-only protein 44(FBXO44)as a novel E3ligase for *** abundance increases upon knockdown of FBXO44,and,inversely,decreases upon overexpression of *** analysis revealed that FBXO44 interacts with PXR,leading to its ubiquitination and proteasomal degradation,and we determined that the F-box associated domain of FBXO44 and the ligand binding domain of PXR are required for the functional *** summary,FBXO44 regulates PXR protein abundance,which has downstream consequences for CYP3A4 levels and drug-drug *** results of this study provide new insight into the molecular mechanisms that regulate PXR protein level and activity and suggest the importance of considering how modulating E3ubiquitin ligase activities will affect PXR-mediated drug metabolism.

A transgenic pig model expressing a CMV-ZsGreen1 reporter across an extensive array of tissues

Since genetic engineering of pigs can benefit both biomedicine and agriculture,selecting a suitable gene promoter is critically *** cytomegalovirus(CMV)promoter,which can robustly drive ubiquitous transgene expression,is commonly used at present,yet recent reports suggest tissue-specific activity in the *** objective of this study was to quantify ZsGreen1 protein(in lieu of CMV promoter activity)in tissues from pigs harboring a CMV-ZsGreen1 transgene with a single integration *** samples(n=35)were collected from neonatal hemizygous(n=3)and homozygous(n=3)piglets and ZsGreen1 abundance was determined via ***1 was detected in all tissues,except hypothalamus,kidney cortex and *** expression patterns of homozygous and hemizygous piglets were similar(P>0.05).However,quantification revealed that ZsGreen1 protein levels were *** neural/endocrine tissues,ZsGreen1 abundance was highest in the anterior pituitary gland,intermediate in the cerebellum and lowest in the cerebrum,spinal cord and posterior pituitary(P<0.05).In the digestive system,ZsGreen1 was more abundant in the salivary gland than esophagus,stomach,pancreas,duodenum,jejunum,ileum,spleen,colon,gallbladder and liver(P<0.05).Interestingly,ZsGreen1 amounts also differed within an organ(i.e.,the right ventricle had 3-fold higher levels than the other heart chambers;P<0.05).These results provide useful information for the use of the CMV promoter to drive transgene expression in the ***,this swine model represents a novel resource of ZsGreen1-labeled organs and a valuable tool to advance genome editing research.

Introduction:As challenges to biodiversity mount,land-use policies have been implemented to balance human needs and the integrity of ecological *** such program,Payments for Ecosystem Services(PES),incentivizes resource users to protect ecosystem services and has been implemented around the world to reduce soil erosion,create or improve wildlife habitats,and improve water quality and other environmental *** PES policy,at its core,is a concept that aims to capture the reciprocal relationships between human systems and ecological function and *** such,PES epistemologically embodies a coupled human and natural systems ***:Yet,despite this conceptual alignment,the on-the-ground implementation or evaluation of PES typically does not adopt this coupled approach and PES programs have little integration between socioeconomic,sociocultural,human demographic,and ecological *** advance the evolution of PES,we consider what and how socioeconomic and ecological factors have been incorporated into PES program implementation and *** also present a conceptual model to articulate how PES research can capture the reciprocal relationships among socioeconomics,demography,and ecology and discuss the quantitative modeling approaches that can support this conceptual development,i.e.,structural equation and agent-based modeling,and latent trajectory ***:By strengthening the conceptual framework for PES within a coupled human and natural systems approach and identifyinganalytical approaches that can be used to quantify and characterize these complex cross-disciplinary relationships,we aim to support the evolution and advancement of PES,in service of more meaningful and positive outcomes for human well-being and ecological sustainability.

Background and Aims:The prevalence of chronic liver dis-ease in adults exceeds 30%in some countries and there is significant interest in developing tests and treatments to help control disease progression and reduce healthcare *** is a rich sampling matrix that offers non-invasive so-lutions suitable for early-stage detection and disease *** previously investigated targeted analysis of a single biomarker,here we investigated a multiparametric approach to breath testing that would provide more robust and reliable results for clinical ***:To identify can-didate biomarkers we compared 46 breath samples from cir-rhosis patients and 42 from *** and analysis used Breath Biopsy OMNI™,maximizing signal and contrast to background to provide high confidence biomarker detec-tion based upon gas chromatography mass spectrometry(GC-MS).Blank samples were also analyzed to provide de-tailed information on background volatile organic compounds(VOCs)***:A set of 29 breath VOCs differed significantly between cirrhosis and controls.A classification model based on these VOCs had an area under the curve(AUC)of 0.95±0.04 in cross-validated test *** seven best performing VOCs were sufficient to maximize classifica-tion performance.A subset of 11 VOCs was correlated with blood metrics of liver function(bilirubin,albumin,prothrom-bin time)and separated patients by cirrhosis severity using principal component ***:A set of seven VOCs consisting of previously reported and novel candidates show promise as a panel for liver disease detection and mon-itoring,showing correlation to disease severity and serum biomarkers at late stage.

Objective: Patients with BRAF-mutant colorectal cancer(CRC) have a poor prognosis. Molecular status is not currently used to select which drug to use in combination with radiotherapy. Our aim was to identify drugs that radiosensitise CRC cells with known BRAF ***: We screened 298 oncological drugs with and without ionising radiation in colorectal cancer cells isogenic for BRAF. Hits from rank product analysis were validated in a 16-cell line panel of human CRC cell lines, using clonogenic survival assays and xenograft models in ***: Most consistently identified hits were drugs targeting cell growth/proliferation or DNA damage repair. The most effective class of drugs that radiosensitised wild-type and mutant cell lines was PARP inhibitors. In clonogenic survival assays, talazoparib produced a radiation enhancement ratio of 1.9 in DLD1(BRAF-wildtype) cells and 1.8 in RkO(BRAF V600 E) cells. In DLD1 xenografts, talazoparib significantly increased the inhibitory effect of radiation on tumour growth(P ≤ 0.01).Conclusions: Our method for screening large drug libraries for radiosensitisation has identified PARP inhibitors as promising radiosensitisers of colorectal cancer cells with wild-type and mutant BRAF backgrounds.
