Background Chitinase is an enzyme that hydrolyzes chitin,a major component of the exoskeleton of insects,including plant pests like *** present study aimed to investigate the expression of chemically synthesized barley ch1 and chi2 genes in cotton(Gossypium hirsutum)through Agrobacterium-mediated ***-five putative transgenic cotton plants were obtained,out of which fifteen plants successfully survived and were shifted to the *** gene-specific primers,amplification of 447 bp and 401 bp fragments confirmed the presence of the ch1 and chi2 genes in five transgenic cotton plants of the T0 *** five plants were further evalu-ated for their mRNA expression *** T0 transgenic cotton plants with the highest mRNA expression level and better yield performance in field,were selected to raise their subsequent *** The T1 cotton plants showed the highest mRNA expression levels of 3.5-fold in P10(2)for the ch1 gene and 3.7-fold in P2(1)for the chi2 *** in situ hybridization(FISH)confirmed a single copy number of ch1 and chi2(hemizygous)on chromosome ***,the efficacy of transgenes on whitefly was evaluated through an insect bioassay,where after 96 h of infestation,mortality rates of whitefly were calculated to be 78%–80%in transgenic cotton *** number of eggs on transgenic cotton plants were calculated to be 0.1%–0.12 per plant compared with the non-transgenic plants where egg number was calculated to be 0.90–1.00 per *** Based on these findings,it can be concluded that the chemically synthesized barley chitinase genes(ch1 and chi2)have the potential to be effective against insects with chitin exoskeletons,including *** transgenic cotton plants expressing these genes showed increased resistance to whiteflies,resulting in reduced egg numbers and higher mortality rates.

Wild cotton species can contribute a valuable gene pool for agronomically desirable cultivated tetraploid cultivars. In order to exploit diploid cotton a regeneration system is required to achieve transformation based goals. The present studies aimed at optimizing the conditions for regeneration of local varieties as well as wild species of cotton. Different callus induction media were tested with varying concentrations of hormones in which sucrose was used as nutritional source. Different explants (hypo-cotyls, cotyledon, root) were used to check the regeneration of both local cotton plants and wild relatives using T & G medium, BAP medium, CIM medium, EMMS medium, and cell suspension medium. Different stages of embryogenicity such as early torpedo stage, late torpedo stage, heart stage, globular stage and cotyledonary stage were observed in wild relatives of cotton. The results of this study pave the way for establishing future transformation methods.

Overexpression of the phytochrome B gene from Arabidopsis thaliana increases plant growth and yield of cotton (Gossypium hirsutum)

The phytochrome B (PHYB) gene of Arabidopsis thaliana was introduced into cotton through Agrobacterium *** and expression of PHYB gene in cotton plants were confirmed by molecular *** RNA (mRNA) expression in one of the transgenic lines,QCC11,was much higher than those of control and other transgenic *** cotton plants showed more than a two-fold increase in photosynthetic rate and more than a four-fold increase in transpiration rate and stomatal *** increase in photosynthetic rate led to a 46% increase in relative growth rate and an 18% increase in net assimilation *** recorded up to two generations,both in the greenhouse and in the field,revealed that overexpression of Arabidopsis thaliana PHYB gene in transgenic cotton plants resulted in an increase in the production of cotton by improving the cotton plant growth,with 35% more ***,the presence of the Arabidopsis thaliana PHYB gene caused pleiotropic effects like semi-dwarfism,decrease in apical dominance,and increase in boll size.

Brain Tumor Classification Based on Fine-Tuned Models and the Ensemble Method

Brain tumors are life-threatening for adults and ***,accurate and timely detection can save *** study focuses on three different types of brain tumors:Glioma,meningioma,and pituitary *** studies describe the analysis and classification of brain tumors,but few have looked at the problem of feature *** are needed to overcome the drawbacks of manual diagnosis and conventional featureengineering *** automatic diagnostic system is thus necessary to extract features and classify brain tumors *** progress continues to be made,the automatic diagnoses of brain tumors still face challenges of low accuracy and high false-positive *** model presented in this study,which offers improvements for feature extraction and classification,uses deep learning and machine learning for the assessment of brain *** learning is used for feature extraction and encompasses the application of different models such as fine-tuned Inception-v3 and fine-tuned *** classification of brain tumors is explored through deep-and machine-learning algorithms including softmax,Random Forest,Support Vector Machine,K-Nearest Neighbors,and the ensemble *** results of these approaches are compared with existing *** Inception-v3 model has a test accuracy of 94.34%and achieves the highest performance compared with other recently reported *** improvement may be sufficient to support a significant role in clinical applications for brain tumor ***,this type of approach can be used as an effective decisionsupport tool for radiologists in medical diagnostics as a second opinion based on the magnetic resonance imaging(MRI)*** may also save valuable time for radiologists who have to manually review numerous MRI images of patients.

High-dose-rate intraluminal brachytherapy during preoperative chemoradiation for locally advanced rectal cancers

AIM:To determine the feasibility and safety of high dose rate intraluminal brachytherapy(HDR-ILBT) boost during preoperative chemoradiation for rectal ***:Between 2008 and 2009,thirty-six patients with locally advanced rectal cancer(≥ T3 or N+),were treated initially with concurrent capecitabine(825 mg/m2 oral twice daily) and pelvic external beam radiotherapy(EBRT)(45 Gy in 25 fractions),then were randomized to group A;HDR-ILBT group(n = 17) to receive 5.5-7 Gy×2 to gross tumor volume(GTV) and group B;EBRT group(n = 19) to receive 5.4 Gy×3 fractions to GTV with *** patients underwent total mesorectal ***:Grade 3 acute toxicities were registered in 12 patients(70.6%) in group A and in 8(42.1%) in group *** pathologic response of T stage(ypT0) in group A was registered in 10 patients(58.8%) and in group B,3 patients(15.8%) had ypT0(P < 0.0001).Sphincter preservation was reported in 6/9 patients(66.7%) in group A and in 5/10 patients(50%) in group B(P < 0.01).Overall radiological response was 68.15% and 66.04% in Group A and B,*** a median follow up of 18 mo,late grade 1 and 2 sequelae were registered in 3 patients(17.6%) and 4 patients(21.1%) in the groups A and B,***:HDR-ILBT was found to be effective dose escalation technique in preoperative chemoradiation for rectal cancers,with higher response rates,downstaging and with manageable acute toxicities.

Highly charged^(129)Xe^(q+)(q=10-30)and^(40)Ne^(q+)(q=4-8)ion-induced secondary electron emissions on the surface of highly oriented pyrolytic graphite(HOPG)are *** total secondary electron yield is measured as a function of the potential energy of incident *** experimental data are used to separate contributions of kinetic and potential electron *** results show that about 4.5%and 13.2%of ion's potential energies are consumed in potential electron emission due to different Xe^(q+)-HOPG and Ne^(q+)-HOPG combinations.A simple formula is introduced to estimate the fraction of ion's potential energy for potential electron emission.

Objective: To evaluate normal spermatozoa morphometry and ultrastructure in estuarine crocodile (Crocodylus porosus). Methods: Four adult male crocodiles aged between 12-15 years, with an average snout to tail length of (3.15±0.01) m were selected for this study. Manipulation of the phallus digitally from the base of the penis was performed on four adult male crocodiles to facilitate the flow of semen to the sulcus. Semen was collected from all individuals for spermatozoa morphometry and ultrastructure study. Morphometry analysis was performed from eosin-nigrosin stained spermatozoa sample. Scanning electron microscopy was conducted to observe the surface ultrastructure of spermatozoa. Results: The morphology of crocodile spermatozoa was made up of acrosome, head, and tail which corresponded to (5.55±1.20) μm, (12.74±1.57) μm, and (70.67±4.40) μm, respectively. The total length of spermatozoa in estuarine crocodile was measured at (88.96±0.52) μm. The most common spermatozoa abnormalities found in the samples were cytoplasmic droplets. Conclusions: Normal morphology, morphometry of spermatozoa in estuarine crocodile which are vermiform shaped-head with a long tail along with questionably high cytoplasmic droplets count are described in this study.

In this paper, the problem of robust H∞ observer design for one-sided Lipschitz nonlinear systems in the presence of time-varying parametric uncertainties is addressed. The main idea is to minimize the L2 gain from the disturbance to the state estimation error and at the same time the admissible one-sided Lipschitz constant is to be maximized. In addition to asymptotic convergence, the designed observer also ensures robustness against disturbances,parametric uncertainties and additive one-sided Lipschitz nonlinear uncertainty. The solution is represented in terms of LMIs which enables us to carry out the design procedure using Matlab LMI toolbox making the nonlinear observer design an easy task.
