WTAP在肝母细胞瘤增殖和进展中的作用

目的:探讨RNA甲基化酶WTAP在肝母细胞瘤细胞中的生物学功能。方法:收集2016年8月至2020年6月同济大学附属第十人民医院手术治疗的肝母细胞瘤患者的肿瘤组织和正常对照组织13对,采用Western blot检测组织中WTAP的表达,在肝母细胞瘤细胞系中转染siRNA敲减WTAP,慢病毒包装WTAP过表达载体WTAP过表达,使用q PCR和Western blot检测敲减和过表达效率,采用CCK8实验检测细胞增殖活性,克隆形成实验检测细胞克隆形成能力,流式细胞术检测细胞凋亡。裸鼠皮下成瘤实验检测细胞在体内的成瘤能力。结果:WTAP在肝母细胞瘤组织中高表达(P=0.0007);Huh6细胞(P=0.0002)和HepG2细胞(P=0.0030)中的WTAP表达水平与QSG-7701细胞相比显著升高,Huh6细胞(P<0.0001)和HepG2细胞(P=0.0035)中的WTAP表达水平与HL-7702细胞相比也显著升高。在肝母细胞瘤细胞中敲减WTAP后,细胞增殖活性和克隆形成能力受到显著抑制,而过表达WTAP则显著增强了细胞增殖活性和克隆形成能力。流式细胞术检测结果显示,Huh6细胞MOCK组的凋亡细胞百分比(10.93±0.2603)%显著低于siWTAP-1组[(16.43±0.6333)%,P=0.0013]和siWTAP-2组[(20.27±0.7535)%,P=0.0003],HepG2细胞MOCK组的凋亡细胞百分比(4.733±0.1764)%显著低于siWTAP-1组[(14.33±0.2728)%,P<0.0001]和siWTAP-2组[(15.33±0.2728)%,P<0.0001]。WTAP稳定敲减细胞株形成的移植瘤的体积(701±82.31)mm^(3)显著小于对照组[(200.2±31.59)mm^(3),P=0.0001];WTAP稳定敲减细胞株形成的移植瘤的重量(0.6368±0.08391)g显著小于对照组[(0.1356±0.03329)g,P<0.0001];WTAP稳定敲减细胞株形成的移植瘤的Ki-67阳性细胞数(108±13.05)显著少于对照组[(406±14.73),P=0.0001]。结论:WTAP在肝母细胞瘤细胞中发挥着促进增殖抑制凋亡的重要生物学功能,可能是肝母细胞瘤诊疗的一个潜在靶点。

Sodium-ion batteries (SIBs) have great potential to be the next major energy storage devices due to their obvious advantages and developing advanced electrodes and electrolytes is urgently necessary to promote its future ***,hard carbon as a state-of-the-art anode of SIBs still suffers from the low initial Coulomb efficiency and unsatisfactory rate capability,which could be improved by forming desirable solid electrolyte interphases (SEI) to some ***,the chemistry and morphology of these interfacial layers are fundamental parameters affecting the overall battery operation,and optimizing the electrolyte to dictate the quality of SEI on hard carbon is a key ***,this review summarizes the recent research on SEI design by electrolyte manipulation from solvents,salts,and *** also presents some potential mechanisms of SEI formation in various electrolyte ***,the current advanced characterization techniques for electrolyte and SEI structure analyses have been comprehensively ***,current challenges and future perspectives of SEI formation on hard carbon anode for SIBs are provided from the viewpoints of its compositions,evolution processes,structures,and characterization techniques,which will promote SEI efficient manipulation and improve the performance of hard carbon,and further contribute to the development of SIBs.

Background The impact of changing land-use patterns and associated anthropogenic threats on scale-dependent habitat use of semi-aquatic mustelids in scent-marking and social behaviour can provide important insights into the habitat ecology of smooth-coated otters(Lutrogale perspicillata).Methods We sampled 180 stream segments(100–400 m) as spatial replicates of 60 1-km2 sites to record indirect evidence(i.e. spraints and mass latrine sites) and group sizes of smooth-coated otters along the Tungabhadra Otter Conservation Reserve(TOCR) during the dry season. To quantify habitat, we recorded stream characteristics, riparian vegetation, and anthropogenic disturbances at the local scale, and hydro-environmental characteristics and land uses at the landscape scale. Using Markovian-chain detection and occupancy models, we assessed the multi-scale habitat use of otters in their selection of suitable areas for scent-marking based on repeated presence-absence data on spraint/latrine locations along the TOCR. We further used linear regression techniques to explore relationships between the number of individuals in smooth-coated otter group and hydro-environmental characteristics, spraint/latrine encounter rate, anthropogenic pressure, land cover, topography, and *** At the local scale, the probability of spraint deposition and group size decreased with anthropogenic disturbance while the probability of spraint detections decreased with grass cover. At the landscape scale, the probability of otter site use for spraint deposition and group sizes increased in southeast flowing streams. Objective:To investigate the effect of Foeniculum vulgare extract against lipopolysaccharide(LPS)-induced microglial activation in vitro as well as cognitive behavioral deficits in ***:LPS-activated BV-2 cell viability was measured using MTT assay and reactive oxygen species(ROS)was studied using DCF-DA *** antioxidative enzymes and pro-inflammatory mediators were analyzed using respective ELISA kits and Western *** in vivo testing,LPS(1 mg/kg,i.p.)was given daily for five days in male Swiss albino mice to produce chronic *** and behavioral tests were performed using open-field,passive avoidance,and rotarod experiments in LPS-induced ***:Foeniculum vulgare extract(25,50 and 100μg/mL)significantly attenuated the LPS-activated increase in nitric oxide(NO),ROS,cyclooxygenase-2,inducible NO synthase,IL-6,and TNF-alpha(P<0.05).Moreover,LPS-induced oxidative stress and reduced antioxidative enzyme levels were significantly improved by Foeniculum vulgare extract(P<0.05).The extract also regulated the NF-κB/MAPK signaling in BV-2 *** an in vivo study,Foeniculum vulgare extract(50,100,and 200 mg/kg)markedly mitigated the LPS-induced cognitive and locomotor impairments in *** fingerprinting analysis showed distinctive peaks with rutin,kaempferol-3-O-glucoside,and anethole as identifiable ***:Foeniculum vulgare extract can ameliorate LPS-stimulated neuroinflammatory responses in BV-2 microglial cells and improve cognitive and locomotor performance in LPS-administered mice.
