Maize(Zea mays),a major food crop worldwide,is susceptible to infection by the saprophytic fungus Aspergillus flavus that can produce the carcinogenic metabolite aflatoxin(AF)especially under climate change induced abiotic stressors that favor mold *** studies have used“-omics”approaches to identify genetic elements with potential roles in AF resistance,but there is a lack of research identifying the involvement of small RNAs such as microRNAs(miRNAs)in *** *** this study,we compared the miRNA profiles of three maize lines(resistant TZAR102,moderately resistant MI82,and susceptible Va35)at 8 h,3 d,and 7 d after *** infection to investigate possible regulatory antifungal role of miRNAs.A total of 316 miRNAs(275 known and 41 putative novel)belonging to 115 miRNA families were identified in response to the fungal infection across all three maize ***-two unique miRNAs were significantly differentially expressed with 39 miRNAs exhibiting temporal differential regulation irrespective of the maize genotype,which targeted 544 genes(mRNAs)involved in diverse molecular *** two most notable biological processes involved in plant immunity,namely cellular responses to oxidative stress(GO:00345990)and reactive oxygen species(GO:0034614)were significantly enriched in the resistant line *** network analysis identified 34 hubs of miRNA-mRNA pairs where nine hubs had a node in the module connected to their target gene with potentially important roles in resistance/susceptible response of maize to *** miRNA hubs in resistance modules(TZAR102 and MI82)were mostly connected to transcription factors and protein ***,the module of miRNA zma-miR156b-nb-squamosa promoter binding protein(SBP),zma-miR398a-3p-SKIP5,and zma-miR394a-5p-F-box protein 6 combinations in the resistance-associated modules were considered important candidates for future functional studies.

Identification of an Mg²⁺-Independent Soluble Phosphatidate Phosphatase in Cottonseed (Gossypium hirsutum L.)

Cotton (Gossypium hirsutum L.) provides a major source of oil for food and feed industries, but little was known about the enzymes in the oil biosynthesis pathway in cottonseed. We are interested in a better understanding of enzymatic components for oil accumulation in cottonseed. The objective of this study was to identify one key enzyme in oil biosynthesis pathway: phosphatidic acid phosphatase (PAP, 3-sn-phosphatidate phosphohydrolase, EC 3.***.3.4). PAP hydrolyzes the phosphomonoester bond in phosphatidate yielding diacylglycerol and Pi. PAPs are generally categorized into Mg2+-dependent soluble PAP and Mg2+-independent membrane-associated PAP. Cottonseed from 25 - 30 days post anthesis was used for the study. The results showed that an Mg2+-independent soluble PAP activity was identified from the cottonseed. While the microsomal fraction of the extract provided only 9% of the PAP activity, 69% of the PAP activity was associated with the cytosol. The PAP activity correlated well with enzyme concentration and incubation time. The pH and temperature optima of the enzyme were pH 5 and 55℃, respectively. Under optimized assay conditions, the Vmax and Km values of cottonseed PAP for dioleoyl phosphatidic acid as the substrate were 2.8 nkat/mg of protein and 539 μM, respectively. Inclusion of the detergent Triton X-100 (0% - 0.5%) or magnesium chloride (1 mM) in the reaction mix did not alter activity to a significant degree. This is the first report of a PAP activity in the seeds of Gossipium hirsutum. This study should provide a basis for purification and characterization of this important enzyme from cottonseed in the future.

Distribution of persistent organic pollutants in aggregate fractions of a temperate forest and semi-rural soil

The fate of persistent organic pollutants (POPs) and their interactions with aggregates of forest soils are not completely understood. Our objectives here were to quantify the distribution of different POPs in water-stable aggregate fractions and to study their influence on soil organic carbon (Corg) content. Soil samples were taken from a forest-site, Gogerddan (G) and a semi-rural site, Hazelrigg (H) in Great Britain, from 0-2 and 2-5 cm and 0-4 and 8-12 cm soil depth, respectively. POPs analyzed were PAHs, PCBs, total DDT, PBDEs and HCB. The bulk soil analysis showed that the concentration of POPs was significantly higher (p ≤ 0.05) in forest site G than in semi-rural site H, particularly at the surface soil levels compared to the subsurface soil depths in both sites. Total concentrations of PCBs and PAHs of both sites were positively correlated with Corg contents. POPs concentrations and Corg, Nt contents of forest site G were significantly higher (p ≤ 0.05) in water-stable macro aggregates (>0.25, >1, >2 mm) than the micro aggregates (>0.053 mm). The POP concentrations of all aggregate fractions after normalizing to their respective Corg content were increased due higher contamination and strong sorption by Corg. These results showed a strong effect of Corg on the partitioning of organic pollutants to soil aggregate size fractions. The present study affirms the ecological significance of forest soils act as a potential sink of POPs. In summary, our results suggest that aggregate fractions may promote soil C storage and act as a potential POP sink in surface soil without increasing their concentration in the aggregate fraction of subsoil.

Recent snow cover variation in the Upper Indus Basin of Gilgit Baltistan, Hindukush Karakoram Himalaya

Mountainous basins like the Upper Indus Basin(UIB) of Gilgit Baltistan(GB) are dependent on seasonal snowmelt and glacier melt. Monitoring of the snow-covered area(SCA) is not only vital for the overall hydrology of the Indus basin but also important to the sustainable agriculture and hydropower system. The snow-covered area in the UIB of GB was investigated for changes over the last 18 years using the Moderate Resolution Imaging Spectroradiometer(MODIS) snow product. The study area was divided into five elevation zones ranging from 877-8564 meters above sea level(m ASL). In contrast to the global cryosphere related studies, SCA in the UIB is slightly increasing. Elevation based SCA analysis also indicated that SCA is slightly increasing in each elevation zone. However, a significant amount of snow is concentrated in areas above 5000 m ASL. Due to the strong correlation between SCA and precipitation, the precipitation data also follow a similar trend. Analysis of the climatic data suggests a statistically significant increase in total monthly precipitation and relative humidity, a slight decrease in mean monthly temperature and a significant upward tendency in monthly solar irradiance data. All these trends in combination with the increasing trend in global precipitation, winter westerly disturbances and orographic precipitation are the important factors behind the slightly increasing SCA in the study area. Our results though constrained by short observation period mainly contribute to the understanding of advancing snow cover and glaciers in Hindukush Karakoram.

Exploring the structural and functional effect of pRB by significant nsSNP in the coding region of RB1 gene causing retinoblastoma

In this study,we identified the most deleterious nsSNP in RB1 gene through structural and functional properties of its protein (pRB) and investigated its binding affinity with *** of 956 SNPs,we investigated 12 nsSNPs in coding region in which three of them (SNPids rs3092895,rs3092903 and rs3092905) are commonly found to be damaged by I-Mutant 2.0,SIFT and PolyPhen *** this effort,we modeled the mutant pRB proteins based on these deleterious *** a comparison of total energy,stabilizing residues and RMSD of these three mutant proteins with native pRB protein,we identified that the major mutation is from Glutamic acid to Glycine at the residue position of 746 of ***,we compared the binding efficiency of both native and mutant pRB (E746G) with *** found that mutant pRB has less binding affinity with E2F-2 as compared to native *** is due to sixteen hydrogen bonding and two salt bridges that exist between native type and E2F-2,whereas mutant type makes only thirteen hydrogen bonds and one salt bridge with *** on our investigation,we propose that the SNP with an id rs3092905 could be the most deleterious nsSNP in RB1 gene causing retinoblastoma.
