General Regulatory Patterns of Plant Mineral Nutrient Depletion as Revealed by serat Quadruple Mutants Disturbed in Cysteine Synthesis

Sulfate is an essential macronutrient for plants. Plants have developed strategies to cope with sulfate deficiency, and other nutrient ion limitations. However, the regulation of these adaptive responses and the coordinating signals that underlie them are still poorly characterized. O-acetylserine （OAS） is a marker metabolite of sulfate starvation and has been speculated to have a signaling function. OAS is synthesized by the enzyme serine acetyltransferase （SERAT）, which is encoded by five distinct genes in Arabidopsis. We investigated quadruple knockout mutants of SERAT that retained only one functional isoform. These mutants displayed symptoms of sulfate starvation. Furthermore, some of them displayed phenotypes typical of prolonged sulfate starvation, in particular, developmental programs associated with senescence or stress responses. Thus, we compared metabolite and transcriptome data from these mutants with N-, P-, K-, and S-depleted plants. This revealed many similarities with general nutrient-depletion-induced senescence （NuDIS）, in- dicating the recruitment of existing regulatory programs for nutrient-starvation responses. Several candidate genes that could be involved in these processes were identified, including transcription factors and other regulatory proteins, as well as the functional categories of their target genes. These results outline components of the regulatory network controlling plant development under sulfate stress, forming a basis for further investigations to elucidate the complete network. In turn, this will advance our broader understanding of plant responses to a range of other nutrient stresses.

Background： Bacillus spp. seem to be an alternative to antimicrobial growth promoters for improving animals＇ health and performance. However, there is little information on the effect of Bacillus spp. in combination with different dietary crude protein（CP） levels on the ileal digestibility and microbiota composition. Therefore, the objective of this study was to determine the effect of Bacillus spp. supplementation to low-（LP） and high-protein diets（HP） on ileal CP and amino acid（AA） digestibility and intestinal microbiota ***： Eight ileally cannulated pigs with an initial body weight of 28.5 kg were randomly allocated to a rowcolumn design with 8 pigs and 3 periods of 16 d each. The assay diets were based on wheat-barley-soybean meal with two protein levels： LP（14% CP, as-fed） and HP diet（18% CP, as-fed）. The LP and HP diets were supplemented with or without Bacil us spp. at a level of 0.04%（as-fed）. The apparent ileal digestibility（AID） and standardized ileal digestibility（SID） of CP and AA was determined. Bacterial community composition from ileal digesta was analyzed by Illumina amplicon sequencing and quantitative real-time PCR. Data were analyzed as a 2 × 2 factorial design using the GLIMMIX procedures of ***： The supplementation with Bacillus spp. did not affect both AID and SID of CP and AA in growing ***, there was no difference in AID of CP and AA between HP and LP diets, but SID of cystine, glutamic acid,glycine, and proline was lower（P 〈 0.05） in pigs fed the HP diets. The HP diets increased abundance of Bifidobacterium spp. and Lactobacillus spp.,（P 〈 0.05） and by amplicon sequencing the latter was identified as predominant genus in microbiota from HP with Bacillus spp., whereas dietary supplementation of Bacillus spp. increased（P 〈 0.05） abundance of Roseburia ***： The HP diet increased abundance of Lactobacillus spp. and Bifidobacterium spp.. Objective:To investigate the effect of electro-acupuncture(EA)on vasomotor symptoms in rats with acute cerebral infarction,by observing the changes in the expression of factors related to the phosphatidylinositol(PI)***:Forty-two Wistar rats were randomly divided into 3 groups by a random number table:the control group(n=6),the model group(n=18)and the EA group(n=18).The EA group was given EA treatment at Shuigou(GV 26)instantly after modeling with middle cerebral artery occlusion(MCAO)method,while the model and control groups were not given any *** degrees of neurological deficiency were evaluated using neurological severity scores(NSS)and the brain blood flow was evaluated by a laser scanning confocal *** blot analysis was conducted to detect the expression levels of G-protein subtype(Gq)and calmodulin(CaM).Competition for protein binding was conducted to detect the expression level of inositol triphosphate(IP3).Thin layer quantitative analysis was conducted to detect the expression level of diacylglycerol(DAG).The expression level of intracellular concentration of free calcium ion([Ca^(2+)]i)was detected by flow ***:The NSS of the model group was significantly higher than the control group at 3 and 6 h after MCAO(P<0.01),while the EA group was significantly lower than the model group at 6 h(P<0.01).The cerebral blood flow in the model group was significantly lower than the control group at 1,3 and 6 h after MCAO(P<0.01),while for the EA group it was remarkably higher than the model group at the same time points(P<0.01).The expressions of Gq,CaM,IP3,DAG and[Ca^(2+)]i in the model group were significantly higher than the control group(P<0.05 or P<0.01),and those in the EA group were significantly lower than the model group at the same time points(P<0.05 or P<0.01).Conclusion:EA treatment at GV 26 can effectively decrease the over-expression of related factors of PI system in rats with acute cerebral infarction,impro

A study to investigate needle insertion at Shenshu(BL23) to puncture psoas major muscle

OBJECTIVE： It is unknown whether the psoas major muscle, thought to be a key muscle for treatment of lower back pain, can be punctured at Shenshu（BL23）.METHODS： Twelve dissected specimens were used for studying the needling pathway of BL23 by perpendicularly inserting the depth-measuring blade of a vernier caliper at BL23. Dimensions of psoas muscle were measured. Correlation studies were conducted. In addition, our samples were grouped by gender and underlying medical conditions for ***： Half（50%） of the needle insertions successfully punctured psoas muscle. The mean depth of needle insertion to puncture psoas muscle（D_（min）） in the group with short-term underlying medical conditions was 38.0 mm（interquartile range 29.0–51.8 mm）, approximately 6 mm deeper than 32.0（29.3–42.5） mm in the group with long-term health problems（P = 0.041）. The cross-sectional area（CSA） of psoas muscle in the former group was on average approximately 1.5 times that of the latter group（P = 0.04）. When the data were analysed by gender, the thickness of psoas muscle in the male group was 19.0（6.5–24.0） mm compared to 19.5（5.8–34.8） mm in the female group（P = 0.02）. The age in the female group（P = 0.04） and the body length of the total group（P = 0.04） negatively correlated to D_（min）.CONCLUSION： Needle insertion at BL23 might be able to puncture psoas muscle. Differences in the CSA of psoas muscle and D_（min） were observed in groups with short-term and long-term underlying medical conditions.

The biogenic silica of sediment samples from Core CJ01-185 which is collected from the eastern India Ocean off the Sunda Strait is analyzed to evaluate the impact of the opening of the Sunda Strait on a paleoproductivity evolution. The new results indicate that the biogenic silica mass values of Core CJ01-185 show the lowest 0.86% in the last glacial period, and reach its maxima of 1.89% in the late Holocene. Furthermore, the biogenic silica mass accumulation rate(MARBSi) values also vary with much higher during the late Holocene than during the last glaciation. The input of additional terrigenous materials from the Java Sea has enhanced the paleoproductivity and increased the biogenic silica mass and MARBSi values after the opening of the Sunda Strait. It is suggested that the paleoproductivity in the study area is mainly influenced by the southeast monsoon and upwelling before the opening of the Sunda Strait. However, the paleoproductivity is dominated by the terrigenous materials input other than by the southeast monsoon or upwelling in the Holocene.

Temperature dependence of nitrogen mineralization and microbial status in O_H horizon of a temperate forest ecosystem

It was hypothesized that increasing air and/or soil temperature would increase rates of microbial processes including litter decomposition and net N mineralization, resulting in greater sequestration of carbon and nitrogen in humus, and consequently development in OH horizon （humus horizon）. To quantify the effect of temperature on biochemical processes controlling the rate of OH layer development three adjacent forest floors under beech, Norway spruce and mixed species stands were investigated at Soiling forest, Germany by an incubation experiment of OH layer for three months. Comparing the fitted curves for temperature sensitivity of OH layers in relation to net N mineralization revealed positive correlation across all sites. For the whole data set of all stands, a Q10 （temperature sensitivity index） value of 2.35-2.44 dependent on the measured units was found to be adequate for describing the temperature dependency of net N mineralization at experimental site. Species-specific differences of substrate quality did not result in changes in biochemical properties of OH horizon of the forest floors. Temperature elevation increased net N mineralization without significant changes in microbial status in the range of I to 15℃. A low Cmic /Corg （microbial carbon/organic carbon） ratio at 20℃ indicated that the resource availability for decomposers has been restricted as reflected in significant decrease of microbial biomass.

Temperature dependence of carbon mineralization and nitrous oxide emission in a temperate forest ecosystem

The measurement of CO2 and N2O effiux from forest soils is of great importance in evaluating the role of forests as sequestering agents of atmospheric CO2 and nitrogen. To quantify the effect of site on temperature dependence of net C-mineralization and N2O-N emissions, three adjacent forest floors under beech, Norway spruce and mixed species stands were investigated at Soiling forest, Germany, by an incubation experiment for three months. The investigated net C-mineralization and N2O-N emissions from all forest floors exhibited an exponential increase with respect to temperature elevation. The temperature coefficient function （Q10 value）, was fitted to flux rates to describe the temperature sensitivity of forest floors on temperature in the range of 1-20℃. Comparing the fitted curves for temperature sensitivity of the forest floors in relation to net carbon mineralization and nitrous oxide emission rates revealed a strong positive correlation across all sites. For the whole data set of all stands, a Q10 value of 1.73-2.10 for net C-mineralization and 2.81-3.58 for N2O-N emissions per measured unit was found to describe the temperature dependency of net C-mineralization and N20-N efflux at experimental site. The absence of clear differences between beech and spruce in mono and mixed species cultures on temperature dependencies of net C-mineralization and N2O-N emission rates indicated that the flux rates were not affected by species-specific differences of litter quality.
