Analysing the quality of Swiss National Forest Inventory measurements of woody species richness

Background: Under ongoing climate and land-use change, biodiversity is continuously decreasing and monitoring biodiversity is becoming increasingly important. National Forest Inventory(NFI) programmes provide valuable timeseries data on biodiversity and thus contribute to assessments of the state and trends in biodiversity, as well as ecosystem functioning. Data quality in this context is of paramount relevance, particularly for ensuring a meaningful interpretation of changes. The Swiss NFI revisits about 8%–10% of its sample plots regularly in repeat surveys to supervise the quality of ***: we analysed the relevance of observer bias with equivalence tests, examined data quality objectives defined by the Swiss NFI instructors, and calculated the pseudo-turnover(PT) of species composition, that is, the percentage of species not observed by both teams. Three attributes of woody species richness from the latest Swiss NFI cycles(3 and 4) were analysed: occurrence of small tree and shrub species(1) on the sample plot and(2) at the forest edge, and(3) main shrub and trees species in the upper ***: we found equivalent results between regular and repeat surveys for all attributes. Data quality, however,was significantly below expectations in all cases, that is, as much as 20%–30% below the expected data quality limit of 70%–80%(proportion of observations that should not deviate from a predefined threshold). PT values were about 10%–20%, and the PT of two out of three attributes decreased significantly in NFI4. This type of uncertainty –typically caused by a mixture of overlooking and misidentifying species – should be considered carefully when interpreting change figures on species richness estimates from NFI ***: our results provide important information on the data quality achieved in Swiss NFIs in terms of the reproducibility of the collected data. Emerging regenerative cell therapies for alveolar bone loss have begun to explore the use of cell laden hydrogels for minimally invasive surgery to treat small and spatially complex maxilla-oral ***,the oral cavity presents a unique and challenging environment for in vivo bone tissue engineering,exhibiting both hard and soft periodontal tissue as well as acting as key biocenosis for many distinct microbial communities that interact with both the external environment and internal body systems,which will impact on cell fate and subsequent treatment ***,we design and bioprint a facile 3D in vitro model of a human dentine interface to probe the effect of the dentine surface on human mesenchymal stem cells(hMSCs)encapsulated in a microporous hydrogel *** demonstrate that the dentine substrate induces osteogenic differentiation of encapsulated hMSCs,and that both dentine andβ-tricalcium phosphate substrates stimulate extracellular matrix production and maturation at the gel-media interface,which is distal to the gel-substrate *** findings demonstrate the potential for long-range effects on stem cells by mineralized surfaces during bone tissue engineering and provide a framework for the rapid development of 3D dentine-bone interface models.

Three-dimensional seismic face stability of shield tunnels inundrained clay

Tunnel face stability has received increasing research interest over the past few decades. However, computing time-efficient and safethree-dimensional solutions under seismic loading is still an unsolved problem, while case studies indicate that seismic loading can be onecritical destabilizing factor affecting tunnel stability. The primary objective of this work is to fill this gap in knowledge by providing compromisingand computationally efficient solutions, along with their respective lower and upper bounds, to compute face stability underseismic conditions. The analyses employ the finite element limit analysis method to evaluate the limit support pressure in undrained clay,considering horizontal pseudo-static seismic forces pointing outwards from the face. Moreover, the analyses employ both constant andlinearly increasing shear strengths with depth. The results are summarized as dimensionless stability charts and tables to facilitate theirinterpretation and future use for tunnel design. A new design equation has been developed to evaluate the stability of the tunnel faceconsidering the effect of seismic forces. Additionally, the effects of different parameters on the shape of the failure mechanism have beeninvestigated by analysing the distribution of shear dissipation.

Telomeric DNA breaks in human induced pluripotent stem cells trigger ATR-mediated arrest and telomerase-independent telomere damage repair

Although mechanisms of telomere protection are well-defined in differentiated cells,how stem cells sense and respond to telomere dysfunction,in particular telomeric double-strand breaks(DSBs),is poorly ***,we report the DNA damage signaling,cell cycle,and transcriptome changes in human induced pluripotent stem cells(iPSCs)in response to telomere-internal *** engineer human iPSCs with an inducible TRF1-FokI fusion protein to acutely induce DSBs at *** this model,we demonstrate that TRF1-FokI DSBs activate an ATR-dependent DNA damage response,which leads to p53-independent cell cycle arrest in *** CRISPR–Cas9 to cripple the catalytic domain of telomerase reverse transcriptase,we show that telomerase is largely dispensable for survival and lengthening of TRF1-FokI-cleaved telomeres,which instead are effectively repaired by robust homologous recombination(HR).In contrast to HR-based telomere maintenance in mouse embryonic stem cells,where HR causes ZSCAN4-dependent extension of telomeres beyond their initial lengths,HR-based repair of telomeric breaks is sufficient to maintain iPSC telomeres at a normal length,which is compatible with sustained survival of the cells over several days of TRF1-FokI *** findings suggest a previously unappreciated role for HR in telomere maintenance in telomerase-positive iPSCs and reveal distinct iPSC-specific responses to targeted telomeric DNA damage.

Comparing Fine-Tuning, Zero and Few-Shot Strategies with Large Language Models in Hate Speech Detection in English

Large Language Models(LLMs)are increasingly demonstrating their ability to understand natural language and solve complex tasks,especially through text *** of the relevant capabilities is contextual learning,which involves the ability to receive instructions in natural language or task demonstrations to generate expected outputs for test instances without the need for additional training or gradient *** recent years,the popularity of social networking has provided a medium through which some users can engage in offensive and harmful online *** this study,we investigate the ability of different LLMs,ranging from zero-shot and few-shot learning to *** experiments show that LLMs can identify sexist and hateful online texts using zero-shot and few-shot approaches through information ***,it is found that the encoder-decoder model called Zephyr achieves the best results with the fine-tuning approach,scoring 86.811%on the Explainable Detection of online Sexism(EDoS)test-set and 57.453%on the Multilingual Detection of Hate Speech Against Immigrants and women in Twitter(HatEval)***,it is confirmed that the evaluated models perform well in hate text detection,as they beat the best result in the HatEval task *** error analysis shows that contextual learning had difficulty distinguishing between types of hate speech and figurative ***,the fine-tuned approach tends to produce many false positives.

The objective of this study consisted of evaluating the effect of the application of chitosan(Q),1-methylcyclopropene(1-MCP)under two controlled temperatures on some physical,physiological,and chemical parameters contributing to the quality of Persian lime Citrus latifolia *** treatments were evaluated,resulting from the combination of four senescence retardant applications(Q,1-MCP,1-MCP+Q,and without application)on fruits stored at two temperatures(12/20℃.Epidermis color(luminosity,chromaticity,and hue),fruit appearance,respiration,weight loss,total juice content,total soluble solids,and titratable acidity were registered during the *** results indicate that the 1-MCP and Q combination favored fruit postharvest quality conditions such as reducing respiration rate;maintaining fruit good appearance,turgidity,and brighter fruit chromaticity;fruits treated with chitosan and stored at 12℃,favored total soluble *** the other hand,Variables such as weight loss,juice content,acidity,phenols,flavonoids,antioxidant activity,and hue values were favored by a 12℃ storing temperature regardless of the *** general,the use of modified atmospheres,senescence retardants,and refrigeration as postharvest methods,can increase Persian lime shelf life.
