Survival in changing and challenging environments requires an organism to efficiently obtain and use its resources. Due to their sessile nature, it is particularly critical for plants to dynamically optimize their meta- bolism. In plant primary metabolism, metabolic fine-tuning involves feed-back mechanisms whereby the output of a pathway controls its input to generate a precise and robust response to environmental changes. By contrast, few studies have addressed the potential for feed-back regulation of secondary metabolism. In Arabidopsis, accumulation of the defense compounds glucosinolates has previously been linked to genetic variation in the glucosinolate biosynthetic gene AOP2. AOP2 expression can increase the transcript levels of two known regulators （MYB28 and MYB29） of the pathway, suggesting thatAOP2 plays a role in positive feed-back regulation controlling glucosinolate biosynthesis. We generated mutants affecting AOP2, MYB28/29, or both. Transcriptome analysis of these mutants identified a so far unrecognized link between AOP2 and jasmonic acid （JA） signaling independent of MYB28 and MYB29. Thus, AOP2 is part of a regulatory feed-back loop linking glucosinolate biosynthesis and JA signaling and thereby allows the glucosinolate pathway to influence JA sensitivity. The discovery of this regulatory feed-back loop provides insight into how plants optimize the use of resources for defensive metabolites.

Mitochondrial GPX1 silencing triggers differential photosynthesis impairment in response to salinity in rice plants

The physiological role of plant mitochondrial glutathione peroxidases is scarcely known. This study attempted to elucidate the role of a rice mitochondrial isoform（GPX1） in photosynthesis under normal growth and salinity conditions. GPX1 knockdown rice lines（GPX1s） were tested in absence and presence of 100 mM Nacl for 6 *** reduction of GPX1 s line under non-stressful conditions, compared with non-transformed（NT） plants occurred in parallel to increased H_2O_2 and decreased GSH contents. These changes occurred concurrently with photosynthesis impairment, particularly in calvin cycle＇s reactions, since photochemical efficiency did not ***, GPX1 silencing and downstream molecular/metabolic changes modulated photosynthesis differentially. In contrast, salinity induced reduction in both phases of photosynthesis, which were more impaired in silenced *** changes were associated with root morphology alterations but not shoot growth. Both studied lines displayed increased GPX activity but H_2O_2 content did not change in response to salinity. Transformed plants exhibited lower photorespiration, water use efficiency and root growth, indicating that GPX1 could be important to salt tolerance. Growth reduction of GPX1 s line might be related to photosynthesis impairment, which in turn could have involved a cross talk mechanism between mitochondria and chloroplast originated from redox changes due to GPX1 deficiency.

The Influence of Exercise and caffeine on cognitive Function in college Students

Exercise has widely been shown to improve cognition, potentially by making individuals more receptive to sensory stimulation or inhibiting irrelevant information. caffeine, one of the world’s most widely used stimulants, seems to have similar effects. It seems that both exercise and caffeine improve cognitive function separately, but little research has been done examining their combined effects. The purpose of this study was to examine the impact of caffeine and exercise, independently and combined, on cognitive function. 20 healthy college students completed the study. These participants were low caffeine consumers. Each participant came to the lab 5 times. During the first session, they completed a graded exercise test on a cycle ergometer to determine ventilatory threshold (VT). The following four sessions were test sessions involving supplementation and exercise. During these, each participant engaged in 30 minutes of cycling (at 90% VT) or 30 minutes of quiet reading after consuming either caffeine (at 4 mg/kg body weight) or a placebo. The contingent continuous Performance Task (cPT) and Wisconsin card Sorting Task were used to measure cognitive function and were completed 5 minutes and 20 minutes after exercise or quiet reading. There were no significant differences found for any variables tested, for condition effect, time effect or condition*time interaction, except for a significant time effect on false alarms on the contingent cPT (p = 0.017). This study may have been limited by multiple variables including the population, executive function measures, caffeine dosage, or exercise prescription. These findings point to the need for future research to understand the changes in cognition from exercise and caffeine in combination. Future research may include looking at exercise at different intensities, different dosages of caffeine, or looking at the long-term cognitive effects.

Experimental hybridization and reproductive isolation between two sympatric species of tephritid fruit flies in the Anastrepha fraterculus species group

Among tephritid fruit flies,hybridization has been found to produce local adaptation and speciation,and in the case of pest species,induce behavioral and ecological alterations that can adversely impact efficient pest *** species group within Anastrepha (Diptera:Tephritidae),is a rapidly radiating aggregate,which includes cryptic species complexes,numerous sister species,and several pest species. Increased DNA binding activity of NF-κB,STAT-3,SMAD3 and AP-1 in acutely damaged liver

AIM: To investigate the role of genes and kinetics of specific transcription factors in liver regeneration, and to analyze the gene expression and the activity of some molecules crucially involved in hepatic regeneration. METHODS: USING gel-shift assay and RT-PcR, transcription factors, such as NF-κB, STAT-3, SMAD3 and AP-1, and gene expression of inducible nitric oxide synthase (iNOS), hepatocyte growth factor (HGF) and c-met were analyzed in an animal model of chemically induced hepatectomy. RESULTS: Gene expression of HGF and its receptor c-met peaked at 3 h and 24 h after acute ccl4 intoxi- cation. iNOS expression was only observed from 6 to 48 h. Transcriptional factor NF-κB had an early activation at 30 min after acute liver damage. STAT-3 peaked 3 h post- intoxication, while AP-1 displayed a peak of activation at 48 h. SMAD3 showed a high activity at all analyzed times. cONcLUSION: TNF-α and IL-6 play a central role in hepatic regeneration. These two molecules are responsible for triggering the cascade of events and switch-on of genes involved in cell proliferation, such as growth factors, kinases and cyclins which are direct participants of cell proliferation.

BAcKGROUND Para-aortic lymph nodes(PALN)are found in the aortocaval groove and they are staged as metastatic disease if involved by pancreatic ductal adenocarcinoma(PDAc).The data in the literature is conflicting with some studies having associated PALN involvement with poor prognosis,while others not sharing the same *** resection is not included in the standard lymphadenectomy during pancreatic resections as per the International Study Group for Pancreatic Surgery and there is no consensus on the management of these *** To investigate the prognostic significance of PALN metastases on the oncological outcomes after resection for *** This is a retrospective cohort study of data retrieved from a prospectively maintained database on consecutive patients undergoing pancreatectomies for PDAc where PALN was sampled between 2011 and *** comparison of the data between PALN+and PALN-subgroups,survival analysis with the Kaplan-Meier method and risk analysis with univariable and multivariable time to event cox regression analysis were performed,specifically assessing oncological outcomes such as median overall survival(OS)and disease-free survival(DFS).RESULTS 81 cases had PALN sampling and 17(21%)were *** N stage was significantly different between PALN+and PALN-patients(P=0.005),while no difference was observed in any of the other *** imaging diagnosed PALN positivity in one *** and DFS were comparable between PALN+and PALN-patients with lymph node positive disease(OS:13.2 mo vs 18.8 mo,P=0.161;DFS:13 mo vs 16.4 mo,P=0.179).No difference in OS or DFS was identified between PALN positive and negative patients when they received chemotherapy either in the neoadjuvant or in the adjuvant setting(OS:23.4 mo vs 20.6 mo,P=0.192;DFS:23.9 mo vs 20.5 mo,P=0.718).On the contrary,when patients did not receive chemotherapy,PALN disease had substantially shorter OS(5.5 mo vs 14.2 mo;P=0.015)and DFS(4.4 mo

Metallated Schiff-Base Macromolecules as Alternative Metalloprotein Electron Transfer Intermediates

In the construction of biosensors, enzymes function as mediators converting biological signals generated by specific biological processes, into electrochemical signals. The ideology of bio-sensor design is retention of electron transfer activity of the enzyme utilizing superior interfacial architecture. In this work a Schiff-base macromolecule has been synthesized by reflux of 2, 3-diaminonaphthalene and pyrrole-2-carboxaldehyde starting materials. The Schiff-base ligand was subsequently complexed with Fecl2?4H2O under reflux, to produce the Fe-Schiff-base complex. The Schiff-base ligand and Fe-Schiff-base complex were characterized using nuclear magnetic resonance (NMR) spectroscopy, Ultra Violet/Visible (UV/Vis) spectroscopy, Fourier transfer infrared resonance (FTIR) and electron energy loss spectroscopy (EELS) to confirm the structure of the synthesis products. NMR spectroscopy confirmed the imide linkage of Schiff-base formation as two symmetrical peaks at 8.1 and 7.7 ppm respectively. comparison of starting materials and product spectra by UV/Vis spectroscopy confirmed the disappearance of the diaminonaphthalene peak at 250 nm as evidence of complete conversion to product. FTIR spectroscopy of the Schiff-base ligand confirmed the formation of the imine bond at 1595 cm-1. EELS spectra comparing Fecl2?4H2O and the Fe-Schiff-base complex, showed good agreement in the energy loss profiles associated with changes to the electronic arrangement of Fe d-orbitals. EDS clearly identified a spectral band for Fe (7 - 8 eV) in the Fe-Schiff-base complex. Electrochemical evaluation of the Fe-Schiff-base complex was compared to the electrochemical signature of denatured cytochrome-c using cyclic voltammetry and square wave voltammetry. The Fe2+/Fe3+ quasi-reversible behavior for iron in the metallated complex was observed at -0.430 V vs. Ag/Agcl, which is consistent with reference values for iron in macromolecular structures.
