Advanced nanoparticles in osteoarthritis treatment

Osteoarthritis (OA) is the most prevalent degenerative joint disorder, affecting hundreds of millions of people globally. Current clinical approaches are confined to providing only symptomatic relief. research over the past two decades has established that OA is not merely a process of wear and tear of the articular cartilage but involves abnormal remodelling of all joint tissues. Although many new mechanisms of disease have been identified in the past several decades, the efficient and sustainable delivery of drugs targeting these mechanisms in joint tissues remains a major challenge. Nanoparticles recently emerged as favoured delivery vehicles in OA treatment, offering extended drug retention, enhanced drug targeting, and improved drug stability and solubility. In this review, we consider OA as a disease affecting the entire joint and initially explore the pathophysiology of OA across multiple joint tissues, including the articular cartilage, synovium, fat pad, bone, and meniscus. We then classify nanoparticles based on their composition and structure, such as lipids, polymers, inorganic materials, peptides/proteins, and extracellular vesicles. We summarise the recent advances in their use for treatment and diagnosis of OA. Finally, we discuss the current challenges and future directions in this field. In conclusion, nanoparticle-based nanosystems are promising carriers that advance OA treatment and diagnosis. © 2024, Chinese Medical Multimedia Press Co ltd. All rights reserved.

Background:Skeletal muscle energetics decline with age,and physical activity(pa)has been shown to offset these declines in older ***,many studies reporting these effects were based on self-reported pa or structured exercise ***,we examined the associations of accelerometry-measured and self-reported pa and sedentary behavior(SB)with skeletal muscle energetics and explored the extent to which pa and sedentary behavior would attenuate the associations of age with muscle ***:As part of the Study of Muscle,Mobility and Aging,enrolled older adults(n=879),810(age=76.4±5.0 years old,mean±SD;58%women)had maximal muscle oxidative capacity measured ex vivo via high-resolution re spirometry of permeabilized myofibers(maximal oxidative phosphorylation(maxOXPHOS))and in vivo by ^(31)phosphorus magnetic resonance spectroscopy(maximal adenosine triphosphate(ATP_(max))).Accelerometry-measured sedentary behavior,light activity,and moderate-to-vigorous pa(MVpa)were assessed using a wrist-worn ActiGraph GT9X over 7 ***-reported sedentary behavior,MVpa,and all pa were assessed with the Community Healthy Activities Model Program for Seniors(CHAMPS)*** regression models with progressive covariate adjustments evaluated the associations of sedentary behavior and pa with muscle energetics,as well as the attenuation of the age/muscle energetics association by MVP A and sedentary *** a sensitivity analysis,we also examined activpal-measured daily step count and time spent in sedentary behavior and their associations with muscle ***:Every 30 min/day more of ActiGraph-measured MVpa was associated with 0.65 pmol/(s×mg)higher maxOXPHOS and 0.012 mM/s higher ATP_(max)after adjusting for age,site/technician,and sex(plight activity was not associated with maxOXPHOS or ATP_(max).Meanwhile,every 30 min/day spent in ActiGraph-measured sedentary behavior was associated with 0.39 pmol/s×mg lower maxOXPHOS and0.

Accelerated intermetallic phase amorphization in a Mg-based high-entropy alloy powder

We describe a novel mechanism for the synthesis of a stable high-entropy alloy powder from an otherwise immiscible Mg-Ti rich metallic mixture by employing high-energy mechanical *** presented methodology expedites the synthesis of amorphous alloy powder by strategically injecting entropic disorder through the inclusion of multi-principal elements in the alloy *** from first principles and materials theory corroborate the results from microscopic characterizations that reveal a transition of the amorphous phase from a precursor intermetallic *** transformation,characterized by the emergence of antisite disorder,lattice expansion,and the presence of nanograin boundaries,signifies a departure from the precursor intermetallic ***,this phase transformation is accelerated by the presence of multiple principal elements that induce severe lattice distortion and a higher configurational *** atomic size mismatch of the dissimilar elements present in the alloy produces a stable amorphous phase that resists reverting to an ordered lattice even on annealing.

There are five vital signs that healthcare providers assess: temperature, pulse, respiration, blood pressure, and pain. Normal levels for the five vital signs are published by the American Heart Association, and other specialty organizations, however, the sixth vital sign (resilience) which adopts the measure of immune resilience is suggested in this paper. resilience is the ability of the immune system to respond to attacks and defend effectively against infections and inflammatory stressors, and psychological resilience is the capacity to resist, adapt, recover, thrive, and grow from a challenge or a stressor. Individuals with better optimal immune resilience had better health outcomes than those with minimal immune resilience. The purpose of this paper is to conceptualize, contextualize, and operationalize all six vital signs. We suggest measuring resilience subjectively and objectively. Subjectively, use a 5-item guided interview revised from the Connor-Davidson resilience Scale (CDrC), a scale of 10 items. The revised CDrC scale is a 5-item scale. The scale is rated on a 5-point likert scale from 0 (not true) to 4 (true all the time). The total score ranges from 0 to 20, with higher total scores indicating greater resilience. The scale demonstrated good construct validity and internal consistency (α = 0.85) during the development of the scale. The CD-rISC had a good Cronbach’s alpha level of 0.85. The revised CD-rISC can be completed in 2 - 4 minutes. To measure resilience objectively, we suggest using Immune resilience (Ir) levels, the level of resilience to preserve and/or rapidly restore immune resilience functions that promote disease resistance and control inflammation and other inflammatory stress. Ir levels are gauged with two peripheral blood metrics that quantify the balance between CD8 and CD4 T-cell levels and gene expression signatures tracking longevity-associated immunocompetence and mortality- or entropy-associated inflammation. Background： lycopene（lYC） is a natural carotenoid with powerful reactive oxygen species（rOS） scavenging activities. The aim of this study was to investigate if lycopene has the ability to reverse rOS-mediated alterations to the motility, viability and intracellular antioxidant profile of bovine spermatozoa subjected to ferrous ascorbate（Fe AA）. Spermatozoa were washed out of fresh bovine semen, suspended in 2.9 % sodium citrate and subjected to lYC treatment（0.25, 0.5, 1 or 2 mmol/l） in the presence or absence of Fe AA（150 μmol/l Fe SO4 and 750 μmol/l ascorbic acid） during a 6 h in vitro culture. Spermatozoa motion characteristics were assessed using the Sperm Vision？computer-aided sperm analysis（CASA） system. Cell viability was examined with the metabolic activity（MTT） assay,rOS generation was quantified via luminometry and the nitroblue-tetrazolium（NBT） test was applied to quantify the intracellular superoxide formation. Cell lysates were prepared at the end of the in vitro culture to investigate the intracellular activity of superoxide dismutase（SOD）, catalase（CAT）, glutathione peroxidase（GPx） as well as the concentrations of glutathione（GSH） and malondialdehyde（MDA）.results： FeA A treatment led to a reduced spermatozoa motility（P 〈 0.001）, viability（P 〈 0.001） and a decline of the antioxidant capacity of spermatozoa（P 〈 0.001） but increased the rOS generation（P 〈 0.001）, superoxide production（P 〈 0.001） and lipid peroxidation（P 〈 0.001）. lYC administration resulted in a preservation of the spermatozoa motion parameters（P 〈 0.001）, mitochondrial activity（P 〈 0.001） and antioxidant characteristics（P 〈 0.001 with respect to SOD;P 〈 0.01 in relation to CAT; P 〈 0.05 as for GPx and GSH） with a concentration range of 1 and 2 mmol/l lYC revealed to be the most ***： Our results suggest that lYC exhibits significant rOS-scavenging and antioxidant properties which may prevent spermatozoa alterations caused by oxidative stress, and preserve the functional
