Leelamine suppresses cMyc expression in prostate cancer cells in vitro and inhibits prostate carcinogenesis in vivo

Aim:Leelamine(LLM)inhibits the growth of human prostate cancer cells but the underlying mechanism is not fully *** present study was undertaken to determine the effect of LLM on cMyc,which is overexpressed in a subset of human prostate ***:The effect of LLM on cMyc expression and activity was determined by western blotting/confocal microscopy and luciferase reporter assay,respectively.A transgenic mouse model of prostate cancer(Hi-Myc)was used to determine the chemopreventive efficacy of ***:Exposure of androgen-sensitive(LNCaP)and castration-resistant(22rv1)human prostate cancer cells to LLM resulted in downregulation of protein and mrNA levels of *** of cMyc partially attenuated LLM-mediated inhibition of colony formation,cell viability,and cell migration in 22rv1 and/or PC-3 *** treatment decreased protein levels of cMyc targets(e.g.,lactate dehydrogenase),however,overexpression of cMyc did not attenuate these effects.A trend for a decrease in the expression level of cMyc protein was discernible in 22rv1 xenografts from LLM-treated mice compared with control *** treatment(10 mg/kg body weight,5 times/week)was well-tolerated by Hi-Myc transgenic *** incidence of high-grade prostatic intraepithelial neoplasia,adenocarcinoma in situ,and microinvasion were lower in LLM-treated Hi-Myc mice but the difference was not statistically ***:The present study reveals that LLM inhibits cMyc expression in human prostate cancer cells in vitro but concentrations higher than 10 mg/kg may be required to achieve chemoprevention of prostate cancer.

Dengue virus non-structural 1 protein interacts with heterogeneous nuclear r.bonucleoprotein H in human monocytic cells

Objective: To study protein-protein interaction between heterogeneous nuclear r.bonucleoprotein H(hn rNP H) and Dengue virus(DENV) proteins. Methods: DENV proteins were screened against the host hn rNP H protein, in order to identify the host-viral protein-protein interactions in DENV infected THP-1 cells by co-immunoprecipitation. The co-localization of the interacting proteins was further confirmed by immunofluorescence microscopy. results: The host protein hn rNP H was found to interact with DENV nonstructural 1 protein and help the virus to multiply in the cell. Conclusions: The non-structural 1 glycoprotein is a key modulator of host immune response and is also involved in viral replication. Therefore, disruption of this key interaction between hn rNP H and DENV nonstructural 1 could be an important therapeutic strategy for management of DENV infection.

Origin of red Color of the Lower Siwalik Palaeosols:A Micromorphological Approach

Palaeosols associated with fluvial of the Siwalik Group are and lacustrine deposits that occur as thick multiple pedocomplexes. The bright red color of the palaeosol beds has been earlier interpreted as a result of hot and arid palaeoclimate. However, as against this view, our investigations of the bright red palaeosol beds of the Lower Siwaliks suggest that the climate was cool and subhumid, instead of hot and arid during the deposition of these beds. Since cold climate is not very conducive to impart red coloration, further research is needed to explain the cause of these red beds. For this, the micromorphological study of soil thin sections was done which showed the presence of features such as dissolution and recrystallisation of quartz, feldspar and mica, compaction, slickensides, presence of calcite spars, subrounded and cracked nature of quartz grains, microfabric, complex patterns of birefringence fabrics, pigmentary ferric oxides, thick cutans and cementation by calcite. These features indicate that diagenesis took place on a large scale in these sediments. The positive Eh and neutral-alkaline pH of soils also suggest that the conditions were favor.ble for the formation of diagenetic red beds. During burial diagenesis of sediments, the hydroxides of ferromagnesian minerals got converted into ferric oxide minerals （hematite）. During deep burial diagenesis smectite was converted into illite and the preponderance of illite over smectite with increasing depth of burial also indicates the diagenesis of sediments. Thus, the red color of the Lower Siwalik palaeosols seems to be due mainly to the burial diagenesis of sediments and does not appear to be due to the then prevailing climatic condition.

Effect of abiotic factors on understory community structures in moist deciduous forests of northern India

Understory vegetation controls, in a significant way, the regeneration of overstory trees, carbon sequestration and nutrient retention in tropical forests. Development and organization of understory vegetation depend on climate, edaphic and biotic factors which are not well correlated with plant community structures. This study aimed to explore the relationships between understory vegetation and abiotic factors in natural and planted forest ecosystems. A non-metric multidimensional scaling(NMS) ordination technique was applied to represent forest understory vegetation among five forest communities, i.e., a dry miscellaneous forest(DMF), a sal mixed forest(SMF), a teak plantation(TP), a low-land miscellaneous forest(LMF) and a savanna area(SAV) of the Katerniaghat Wildlife Sanctuary, located in northern India. Microclimatic variables, such as photosynthetically active radiation(PAr), air temperature(AT), soil temperature(ST), ambient atmospheric CO 2 concentration, absolute air humidity(AH), physical and chemical soil properties as well as biological properties were measured. Understory species were assessed via 100 random quadrats(5 m × 5 m) in each of the five forests in which a total of 75 species were recorded encompassing 67 genera from 37 families, consisting of 32 shr.bs and 43 plant saplings. DMF was the most dense forest with 34,068 understory individuals per ha of different species, whereas the lowest understory population(13,900 per ha) was observed in the savanna. Ordination and correlation revealed that microclimate factors are most important in their effect compared to edaphic factors, on the development of understory vegetation in the various forest communities in the north of India.

Stimulated by thermal optimization in magnetic materials process engineering,the present investigation investigates theoretically the entropy generation in mixed convection magnetohydrodynamic(MHD)flow of an electrically-conducting nanofluid from a vertical *** mathematical model includes the effects of viscous dissipation,second order velocity slip and thermal slip,has been *** cylindrical partial differential form of the two-component non-homogenous nanofluid model has been transformed into a system of coupled ordinary differential equations by applying similarity *** effects of governing parameters with no-flux nanoparticle concentration have been examined on important quantities of ***,the dimensionless form of the entropy generation number has also been evaluated using homotopy analysis method(HAM).The present analytical results achieve good correlation with numerical results(shooting method).Entropy is found to be an increasing function of second order velocity slip,magnetic field and curvature *** is elevated with increasing curvature parameter and magnetic parameter whereas it is reduced with mixed convection *** flow is accelerated with curvature parameter.but decelerated with magnetic *** transfer rate(Nusselt number)is enhanced with greater mixed convection parameter,curvature parameter and first order velocity slip parameter.but reduced with increasing second order velocity slip *** generation is also increased with magnetic parameter,second order slip velocity parameter,curvature parameter,thermophoresis parameter.buoyancy parameter and reynolds number whereas it is suppressed with first order velocity slip parameter.brownian motion parameter and thermal slip parameter.

Studies on selected polymeric materials using the photoacoustic spectroscopic technique

Polymethylmethacrylat-graft-polybisphenol A-carbonate （PMMA-G-PC） with 50% grafting is synthesized. The graft co-polymerization of methyhnethacrylate （0.036 mol. lit-1） onto polybisphenol-A-carbonate （0.5 g） in the presence of a redox couple formed from potassium persulphate （40 mol-lit-1） and thio-urea （30 mmol. lit-1） in aqueous nitric acid （0.18 M, 100 ml） in air at （45±2）℃ for 3.0 h. Condensation of （PMMA-G-PC） with N- [p-（carboxyl phenyl amino acetic acid）] hydrazide （PCPH） affords polybisphenol-A-carbonate-graft-polymethylmethacrylate hydrazide （PCGH）. The photoacoustic （PA） spectra of （PCGH） are recorded in a wavelength range from 200 nm to 800 nm at a modulation frequency of 22 Hz, and compared with those of pure polybisphenol-A-carbonate （PC）, （PMMA-G-PC） and （PCPH）. In the present work, a non-destructive and non-contact analytical method, namely the photoacoustic technique, is successfully implemented for optical and thermal characterization of selected polymeric materials. The indigenous PA spectrometer used in the present study consists of a 300-W xenon arc lamp, a lock-in amplifier, a chopper, a （1/8）-m monochromator controlled by computer and a home-made PA cell.

bianchi Type-Ⅲ Cosmological Models with Gravitational Constant G and the Cosmological Constant A

Einstein field equations with variable gravitational and cosmological constants are considered in the presence of perfect fluid for the bianchi type-Ⅲ universe by assuming conservation law for the energy-momentum tensor. Exact solutions of the field equations are obtained by using the scalar of expansion proportional to the shear scalar θ∝σ, which leads to a relation between metric potential b = Cn, where n is a constant. The corresponding physical interpretation of the cosmological solutions are also discussed.
