Notch signaling： a novel regulating differentiation mechanism of human umbilical cord blood-derived mesenchymal stem cells into insulin-producing cells in vitro

Background Human umbilical cord blood-derived mesenchymal stem cells （UCB-MSCs） could be induced to differentiate into insulin producing cells （IPCs） in vitro, which have good application potential in the cell replacement treatment of type-1 diabetes. However, the mechanisms regulating this differentiation have remained largely unknown. Notch signaling is critical in cell differentiation. This study investigated whether Notch signaling could regulate the IPCs differentiation of human UCB-MSCs. Methods Using an interfering Notch signaling protocol in vitro, we studied the role of Notch signaling in differentiation of human UCB-MSCs into IPCs. In a control group the induction took place without interfering Notch signaling. Results Human UCB-MSCs expressed the genes of Notch receptors （Notch 1 and Notch 2） and ligands （Jagged 1 and Deltalike 1）. Human UCB-MSCs with over-expressing Notch signaling in differentiation resulted in the down-regulation of insulin gene level, proinsulin protein expression, and insulin-positive cells percentage compared with the control group. These results showed that over-expressing Notch signaling inhibited IPCs differentiation. Conversely, when Notch signaling was attenuated by receptor inhibitor, the induced cells increased on average by 3.06-fold （n=-4, P 〈0.001） in insulin gene level, 2.60-fold （n=-3, P 〈0.02） in proinsulin protein expression, and 1.62-fold （n=-6, P 〈0.001） in the rate of IPCs compared with the control group. Notch signaling inhibition significantly promoted IPCs differentiation with about 40% of human UCB-MSCs that converted to IPCs, but these IPCs were not responsive to glucose challenge very well both in vitro and in vivo. Hence, further research has to be carried out in the future. Conclusions Notch signaling may be an important mechanism regulating IPCs differentiation of human LICB-MSCs in vitro and Notch signaling inhibition may be an efficient way to increase the number of IPCs, which may resolve the shortage of

As one of the payloads for the Advanced Space-based Solar Observatory(ASO-S)mission,the Lyman-alpha(Lyα)Solar Telescope(LST)is aimed at imaging the Sun and the inner corona up to 2.5 R⊙(mean solar radius)in both the Lyα(121.6 nm)and visible wavebands with high temporo-spatial resolution,mainly targeting solar flares,coronal mass ejections(CMEs)and filaments/*** observations allow us to trace solar eruptive phenomena from the disk center to the inner corona,to study the relationships between eruptive prominences/filaments,solar flares and CMEs,to explore the dynamical processes and evolution of solar eruptions,to diagnose solar winds,and to derive physical parameters of the solar *** is actually an instrument suite,which consists of a Solar Disk Imager(SDI),a Solar Corona Imager(SCI),a White-light Solar Telescope(WST)and two Guide Telescopes(GTs).This is the first paper in a series of LST-related *** this paper,we introduce the scientific objectives,present an overview of the LST payload and describe the planned *** detailed design and data along with potential diagnostics are described in the second(Paper II)and third(Paper III)papers,respectively,appearing in this issue.

The newly launched fengyun-3D(FY-3D)satellite carried a wide-field auroral imager(WAI)that was developed by Changchun Institute of Optics,Fine Mechanics and Physics,Chinese Academy of Sciences(CIOMP),which will provide a large field of view(FOV),high spatial resolution,and broadband ultraviolet images of the aurora and the ionosphere by imaging the N2 LBH bands of *** WAI consists of two identical cameras,each with an FOV of 68°in the along-track direction and 10°in the cross-track *** two cameras are tilted relative to each other to cover a fan-shaped field of size 130°×10°.Each camera consists of an unobstructed four-mirror anastigmatic optical system,a BaF2 filter,and a photon-counting imaging *** spatial resolution of WAI is ~10 km at the nadir point at a reference height of 110 km above the Earth’s *** sensitivity is>0.01 counts s^(−1) Rayleigh^(−1) pixel^(−1)(140-180 nm)for both cameras,which is sufficient for mapping the boundaries and the fine structures of the auroral oval during storms/*** on the tests and calibrations that were conducted prior to launch,the data processing algorithm includes photon signal decoding,geometric distortion correction,photometric correction,flatfield correction,line-of-sight projection and correction,and normalization between the two *** processed images are compared with DMSP SSUSI *** agreement between the images that were captured by two instruments demonstrates that the WAI and the data processing algorithm operate normally and can provide high-quality scientific data for future studies on auroral dynamics.

Sky location of massive black hole binaries in the foreground of Galactic white dwarf binaries

For space-based gravitational wave （GW） detection,the main noise source for massive black hole binaries （MBHBs） is attributed to approximately 10～7double white dwarf binaries in the *** a GW source,the amplitude of the detector response,recorded by a space-based gravitational wave detector,exhibits a modulation effect with a year period when observing the source from various orbital *** the adverse conditions mentioned above,where there is a strong foreground noise and annual modulation in the signals,we employed the wavelet transform and the strong-amplitude relevant orbital position search methods,which allows the weak MBHB sources to achieve higher locating *** detail,for two MBHB sources of lower intensity,the precision of luminosity distance,represented by the ratio ?DL/DLat the 95%confidence level,is enhanced by factors of ～*** angular resolutions,denoted by??s,are enhanced by a factor of ～*** improvements increase the number of detectable GW sources,facilitate multi-messenger follow-up observations and provide constraints on the cosmological constant.

The solar X-ray and Extreme Ultraviolet Imager(X-EUVI),developed by the Changchun Institute of Optics,Fine Mechanics and Physics,Chinese Academy of Sciences(CIOMP),is the first space-based solar X-ray and Extreme ultraviolet(EUV)imager of China loaded on the fengyun-3E(FY-3E)satellite supported by the China Meteorological Administration(CMA)for solar *** started work on July 11,2021,X-EUVI has obtained many solar *** instrument employs an innovative dual-band design to monitor a much larger temperature range on the Sun,which covers 0.6-8.0 nm in the X-ray region with six channels and 19.5 nm in the EUV region.X-EUVI has a field of view of 42′,an angular resolution of 2.5″per pixel in the EUV band and an angular resolution of 4.1″per pixel in the X-ray *** instrument also includes an X-ray and EUV irradiance sensor(X-EUVS)with the same bands as its imaging optics,which measures the solar irradiance and regularly calibrates the solar *** radiometric calibration of X-EUVS on the ground has been completed,with a calibration accuracy of 12%.X-EUVI is loaded on the FY-3E satellite and rotates relative to the Sun at a uniform ***-field calibration is conducted by utilizing successive rotation solar *** agreement between preliminarily processed X-EUVI images and SDO/AIA and Hinode/XRT images indicates that X-EUVI and the data processing algorithm operate properly and that the data from X-EUVI can be applied to the space weather forecast system of CMA and scientific investigations on solar activity.

石墨烯独特的结构和性能使其在纳米电子、半导体器件等领域成为研究的热点,但其零带隙的特性严重限制了其应用。采用化学气化沉积法制备了多层石墨烯,并使用溴蒸汽对制备的多层石墨烯进行掺杂,分析研究了溴蒸汽化学掺杂对石墨烯带隙的影响。为了对比溴蒸汽掺杂对石墨烯带隙的影响,使用633nm He-Ne光分别测量了石墨烯掺杂前和掺杂后的拉曼光谱,根据拉曼光谱计算了石墨烯费米能级移动与掺杂溴蒸汽之间的关系。实验结果表明:溴蒸汽掺杂对石墨烯拉曼光谱G带产生影响;随着掺杂溴蒸汽体积的增加,拉曼光谱G带向高频移动并逐渐趋于稳定;G带和2D带强度比也迅速增加,并最终趋于稳定。费米能级的移动与G峰位置成线性关系,利用G峰峰值位置与费米能级实验关系式计算了溴掺杂后石墨烯的费米能级,分析了化学掺杂对石墨烯带隙的影响。

As one of the three payloads for the Advanced Space-based Solar Observatory(ASO-S)mission,the Lyman-alpha(Lyα)Solar Telescope(LST)is composed of three instruments:a Solar Corona Imager(SCI),a LyαSolar Disk Imager(SDI)and a full-disk White-light Solar Telescope(WST).When working in-orbit,LST will simultaneously perform high-resolution imaging observations of all regions from the solar disk to the inner corona up to 2.5 R⊙(R⊙stands for the mean solar radius)with a spatial resolution of 4.8′′and 1.2′′for coronal and disk observations,respectively,and a temporal resolution of 30–120 s and 1–120 s for coronal and disk observations,*** maximum exposure time can be up to20 s due to precise pointing and image stabilization *** the three telescopes of LST,SCI is a dual-waveband coronagraph simultaneously and independently observing the inner corona in the HI Lyα(121.6±10 nm)line and white light(WL)(700±40 nm)wavebands by using a narrowband Lyαbeam splitter and has a field of view(FOV)from 1.1 to 2.5 R⊙.The stray-light suppression level can attain<10^-6 B⊙(B⊙is the mean brightness of the solar disk)at 1.1 R⊙and≤5×10^-8 B⊙at 2.5 R⊙.SDI and WST are solar disk imagers working in the Lyαline and 360.0 nm wavebands,respectively,which adopt an off-axis two-mirror reflective structure with an FOV up to 1.2 R⊙,covering the inner coronal edge area and relating to coronal *** present the up-to-date design for the LST payload.

Background:Insomnia is a prevalent clinical condition and Shangxia Liangji formula(SXLJF)is a well-established method of ***,the specific mechanism of action of SXLJF remains ***:The mouse model of insomnia was established by intraperitoneal injection of ***-two mice were randomly divided into a negative control group,model group,SXLJF group(18.72 g/kg/day),and positive control group(diazepam,2 mg/kg)and treated with the corresponding drugs for 7 consecutive *** open field test and pentobarbital-induced sleeping test were ***-MS-based untargeted metabolomics and network pharmacology were applied to explore the potential targets of SXLJF for treating ***,key targets were validated using ***:Behavioral tests demonstrated that SXLJF reduced the total distance,average velocity,central distance,and sleep latency,and prolonged sleep *** and network pharmacology revealed potential targets,signaling pathways,metabolic pathways,and metabolites associated with the anti-insomnia effects of ***,tyrosine hydroxylase(TH)and tyrosine metabolism emerged as crucial metabolic pathways and targets,***-qPCR results supported the role of TH in the mechanism of SXLJF in treating ***:In conclusion,TH and tyrosine metabolism may represent significant targets and pathways for SXLJF in treating insomnia.
