Fluorescence imaging has facilitated fluorescent probes to analyze the subcellular localization and dynamics of biological targets. In this paper, we reported a fluorogenic probe for bacteria imaging. The probe was an imidazolium-derived pyrene compound, which self-assembled to form nano-particles and the pyrene fluorescence was quenched by the aggregation effects. When the self-assembly nanoparticles interacted with anionic bacteria surfaces, synergistic effects of electrostatic interaction and hydrophobic force caused competing binding between bacteria surfaces and imidazoliums. This binding resulted in the disassembly of the aggregates to give fluorescence turn-on signal. Meanwhile, the probe bound bacteria surfaces and displayed both pyrene-excimer and pyrene-monomer fluorescence, which gave ratiometric signal. Then, fluorescent labeling by the probe enabled the two-photo ratiometric imaging of bacteria.

The need for temporal resolution and long-term stability in super-resolution fluorescence imaging has motivated research to improve the photostability of fluorescent *** to the inevitable photobleaching of fluorophores,it is difficult to obtain long-term super-resolution imaging regardless of the self-healing strategy of introducing peroxide scavengers or the strategy of fluorophore structure modification to suppress TICT *** buffered fluorogenic probe uses the intact probes in the buffer pool to continuously replace the photobleached ones in the target,which greatly improves the photostability and enables stable dynamic super-resolution imaging for a long *** the buffering capacity comes at the expense of reducing the number of fluorescent probes in targets,resulting in low staining fluorescence *** this paper,we selected BODIPY 493,a lipid droplet probe with high fluorescence brightness,to explore the dynamic process of lipid droplet staining of this probe in *** found that BODIPY 493 only needs very low laser power for lipid droplet imaging due to the high molecular accumulation in lipid droplets and the high brightness,and the spatiotemporal resolution is greatly *** importantly,we found that BODIPY 493 also has a certain buffering capacity,which enables BODIPY 493 to be used for super-resolution imaging of lipid droplet *** work reminds researchers to coordinate the buffering capacity and brightness of fluorogenic probes.

Cascading reactions in fluorophores accompanied by the replacement of different fluorescence wavelengths can be used to develop luminescent materials and reactive fluorescent *** on multiple signal channels,the selectivity of probes can be improved and the range of response to guest molecule recognition can be *** regulating the position,number,and activity of active sites in fluorophores,fluorescent probes that successively react with thiol and amino groups in cysteine(Cys),homocysteine(Hcy)have been developed,which can only react with the thiol group of GSH,In this paper,we report the first probe capable of cascading nucleophilic substitution reaction with the thiol group and amino group of GSH at a single reaction site,and showed the dual-color recognition of GSH,which improved the selectivity of GSH also was an extension of GSH *** probe Rho-DEA was based on a TICS fluorophore,and the intramolecular cascade nucleophilic substitution reaction occurs with Cys/*** thiol substitution of the first step reaction with Cys/Hcy was quenched due to intersystem crossing to triplet state,so GSH can be selectively recognized from the fluorescence ***-DEA has the ability of mitochondrial localization,and finally realized in situ dual-color fluorescence recognition of GSH in mitochondria.

White light emitting systems of pure organic materials have attracted extensive research interest due to their better compatibility and functional *** reported organic white light materials are mainly based on the multi-channel emission regulation of the compound itself or the mixing of multicolor luminescence materials,but studies on the dependence between multicolor luminescence and the external environment are lacking,which limits the application of these materials in areas such as identification and *** paper reports that the 4-or 3?hydroxyl-substituted naphthalimides NapH1 and NapH2 form intermolecular hydrogen bonds with adjacent molecules in the environment,and undergo excited-state intermolecular proton transfer under irradiation,resulting in blue-yellow or blue-red dual fluorescence emission,*** the two compounds have different two-color luminescence channels and the two-color intensity ratio is affected by the environment,and the intermolecular hydrogen bond is determined by the hydrogen bond receptor,polarity,and temperature in the environment,the full spectrum from blue to red light and white light emission can be obtained by adjusting the mixing ratio of the two dyes and the solvent polarity and the ambient *** environmentally sensitive white emission is used to detect the alkalinity of different papers,and the dyed paper can be used as a test strip for acid-base vapor detection.

With the development of single-molecule detection and super-resolution fluorescence imaging,rhodamine dyes gain new *** the modification of the N-substituents a nd the replacement of the oxygen atom in xanthene,the wavelength and brightness can be effectively ***,the spectra of rhodamine,especially due to the balance between ring-closed non-fluorescent lactone and ring-opened fluorescent zwitterion/cation,are sensitive to interference from various environmental facto rs,In this way,the spectral data of various rhodamines repo rted by different re search groups under different test conditions lacked comparability,sometimes even lacked *** order to meet the requirements for the accuracy and uniformity of spectral data in the research of single molecule imaging and dye structure-fluorescence relationship study,we have tested the spectra of fifteen rhodamine dyes that cover the visible and near-infrared regions under exactly the same *** studying the dependence of the spectra on dye concentrations,it was confirmed that 1 μmol/L was ideal fo r detection less from the interference of dye molecule *** provide compre hensive and reliable spectral data of these fifteen dyes,which are expected to be used as references for future *** the direct comparison of different rhodamine spectra would help to understand the structure-fluorescence relationship of rhodamines.

The transmembrane protein HER2 is overexpressed in approximately 30% of breast cancer ***2-positive breast cancers tend to spread more aggressively, which result in increased mortality inwomen. Nowadays, the real-time monitoring of HER2 status is important in clinical diagnosis andtreatment for HER2-positive patients. Although IHC and FISH assay are standard methods to evaluate thetissue HER2 status, both approaches which required high quality tissue samples and are not suitable formonitoring the status of HER2 in real time. Since extracellular domain （ECD） of the HER2 receptor can beshed into the circulation, the serum test of HER2 ECD has been developed as an additional approach toprobe HER2 overexpression. The serum test will be able to monitor the dynamic changes of HER2 *** this paper, we detected serum HER2 ECD using CyS-labeled HB5 aptamer as a result of its specificbinding ability to HER2 ECD. This aptamer-based fluorescent probe is easily synthesized and modifiedand as sensitive as anti-HER2 antibodies. We believe that Cy5-HB5 may have application potentials inserum HER2 test for clinical utility of breast cancer, such as recurrence and metastases.

Understanding relationships between molecular structures and fluorescent properties is critical to enable rational deployment of fluorophores. 1-Acetyl pyrene is an important pyrene-derivative, used extensively as an environment-sensitive probe. In the past, the fluorescence of 1-acetylpyrene was considered to be polarity-sensitive with a large positive solvatochromism, and its monomer emissions were believed in the range of 410-470 nm. In this paper, our experimental and theoretical studies showed that the monomer fluorescence of 1-acetylpyrene is centered at ～390 nm,which is similar to that of pyrene dyes and not polarity-sensitive. Previously observed "monomer emission" has been re-assigned to that of dimers, which represent the dominant existence form of 1-acetylpyrene in the solution phase,as a result of strong intermolecular π-π stacking interactions.
