More than 50%of marketed drugs and 90%drug candidates are believed to be poorly water-soluble, and these figures keep growing due to input of new drug entities into the pool, thanks to the fast development in drug discovery. It has long been recognized that poor solubility poses problems of retarded dissolution rate and thereby poor bioavailability.

文章对近十年内发生的30起典型高校实验室安全事故进行了深入分析,研究发现,高校实验室爆炸事故占比较为突出,其次是火灾事故。此外,结合典型事故致因分析,文章基于“2-4”事故致因模型,从基层面、管理层面和外部层面三个维度出发提出了一个全面构建高校实验室安全管理体系的思路。这一体系的构建不仅为高校实验室的安全管理提供了科学参考,也为高校的科研和人才培养工作提供了坚实的保障。

目的探索1H-MRS活体检测结肠癌耐药性的可能性。方法 5只耐药(耐5-FU)与5只不耐药人类结肠癌SW480荷瘤裸鼠于肿瘤直径大于1.5cm时行1H-MRS检查,随后处死荷瘤鼠,切下肿瘤检测其PKC、P-gp、MRP1蛋白表达(Western Blot),观察肿瘤凋亡(TUNNEL法)。分析结肠癌耐药性与1H-MRS检测代谢物、肿瘤耐药基因表达、肿瘤凋亡的相关性。结果不耐药组人类结肠癌SW480裸鼠肿瘤的cho、lac、Lip及m I峰/cr面积比较耐药组人类结肠癌SW480/5-FU裸鼠增加,差别有统计学差异(P0.05)。肿瘤代谢物cho、Lac、Lip、m I/cr与肿瘤耐药性呈正相关性;cho面积与肿瘤PKC表达光密度呈正相关,与肿瘤Pgp表达光密度呈正相关。结论肿瘤代谢物cho、lac、Lip的改变有可能有助于利用1H-MRS活体检测结肠癌耐药性改变。

This paper conducts laboratory tests to investigate detailedly the soil deformation law around the pipeline and its penetration depth under self-gravity. The seabed model is prepared by consolidating saturated soil using vacuum pressure technology, and the pipeline models are specifically designed to possess different radii. Based on the experimental results and digital images, the soil deformation process is analyzed and summarized, a kinematic admissible velocity field is given and an upper bound solution of pipeline penetration depth and soil reaction force is derived and proposed in this paper. In order to verify the accuracy of the upper bound solution deduced in this paper,a comparison is made among some published results and the solution suggested in this paper, the comparison results confirm that the upper bound solution and the soil failure mode are reasonable. Finally two empirical formulas are given in this paper to estimate the soil reaction force of seabed and the penetration depth of pipeline. The empirical formulas are in agreement with the upper bound solution derived in this paper, and the conclusion of this paper could provide some theoretical reference for the further study of the interaction between the pipeline and the soil.

Estuarine environments are influenced by both river flows and oceanic tidal movement of water, sediment, and nutrients, often forming ecosystems that are rich in resources and biodiversity. The Yellow River once carried the world’s largest sediment load, but artificial structures have transformed its hydrodynamic processes. An annual Water-Sediment Regulation Scheme(WSRS) was introduced to flush accumulated sediment from the Xiaolangdi Reservoir, which provides flood control and water ***, the effect of the WSRS process on the runoff and sediment conditions, nutrients, and ecological environment of the Yellow River Estuary and adjacent ocean are not well understood. In the current study, a coupled hydrodynamic-nutrient-plankton ecosystem dynamics model(FVCOM-FABM-NPZD)was constructed to simulate changes in the ecological parameters and in the vertical response in the Yellow River Estuary before, during, and after the WSRS, This model also was used to quantify the effects of changes in the incoming material flux on the hydrodynamic and ecological environment of the estuary, using the Yellow River Xiaolangdi Reservoir water transfer and sand transfer as an example. The study found that the WSRS changed the spatial and temporal distribution of temperature, salinity,sediment, inorganic nitrogen, and phytoplankton in the Yellow River Estuary and adjacent waters. It also had a significant effect on the temperature, salinity, and ecology of the adjacent near-shore marine environment. The spatial and temporal responses of zooplankton and phytoplankton to the WSRS differed. Zooplankton showed a lag in response to the WSRS and were mainly influenced by temperature and phytoplankton. The phytoplankton concentration was positively influenced by inorganic nitrogen and negatively influenced by salinity and sediment. 目的探讨PRKCQ-AS1在气道平滑肌细胞(airway smooth muscle cells,ASMCs)的作用。方法首先通过转录组测序技术(RNA-seq)检测哮喘儿童肺泡灌洗液中的差异表达基因(differentially expressed genes,DEGs)。随后,分别构建PRKCQ-AS1慢病毒敲低和过表达载体并转染ASMCs。qRT-PCR方法检测了ASMCs中PRKCQ-AS1的表达;用CCK-8和流式细胞术分别检测ASMCs的增殖和凋亡;酶联免疫吸附测定(enzyme-linked immunosorbent assay,ELISA)检测ASMCs培养上清液中TNF-α和IL-1β的含量;lncRNA芯片检测PRKCQ-AS1敲低后的DEGs并进行KEGG通路富集;Western blot检测p-Akt和Akt的水平。结果RNA-seq结果显示,PRKCQ-AS1是上调最明显的DEGs之一。PRKCQ-AS1过表达促进了ASMCs的增殖和迁移并抑制了其凋亡,同时,PRKCQ-AS1过表达使上清液中炎性因子TNF-α和IL-1β的水平升高,而PRKCQ-AS1敲低则呈现相反的结果。对PRKCQ-AS1敲低后得到的606个DEGs进行KEGG信号通路富集分析,结果显示PI3K-Akt信号被明显富集。Western blot结果表明,PRKCQ-AS1敲低抑制了Akt的活化;相反,PRKCQ-AS1过表达则促进了Akt的水平。结论PRKCQ-AS1可能通过增强Akt信号促进ASMCs炎症反应,进而加剧了ASMCs的增殖和迁移,最终导致哮喘的发展。

bstract A chemometric study using pattern recognition technology was carried out to characterize the geographic origins of the brown planthopper, Nilaparvata lugens. The concentrations of 23 trace elements （Mn, Mo, Cd, Ce, V, Th, Cs, Be, T1, Fe, Nd, Pr, Se, Tm, Lu, Eu, Ho, Br, Dy, Gd, U, Sm and Er） in 53 samples from seven regions in southern China were determined using inductively coupled plasma mass spectrometry. The data obtained were successively evaluated using a multivariate statistical approach, namely, linear discriminant analysis, which allowed classification and discrimination of the N. lugens samples from Fuqing, Shaoguan, Hepu, Yongfu, Hengnan, Wan-an and Yongkang with high accuracy and a clear separation among the seven regions. The results show that pattern identification on the basis of trace elements in the bodies ofN. lugens is feasible for determining the geographic origins of individuals.
