Endosulfan-mediated Biochemical Changes in the Freshwater Fish Clarias batrachus

Objective Endosulfan is an extremely toxic organochlorine pesticide to aquatic organisms which might be hampering fish health through impairment of metabolism sometimes leading to death. So an experimental protocol was designed to look at endosulfan effects on a number of selected biochemical endpoints as well as to develop a mechanistic understanding of biochemical effects of endosulfan in freshwater fish. Methods The adult freshwater catfish Clarias batrachus were collected and acclimatized to laboratory condition for two weeks prior to experimentation. The toxicity bioassay test of commercial grade endosulfan (35% EC) was conducted for 21 days to determine its initial lethal concentration. The fish were exposed to sublethal concentration of endosulfan (0.06 mg/L) for 21 days. Pesticide - withdrawal experiments were also performed to study recovery. Protein synthesis inhibitors were injected to know the possible mechanism of recovery. The specimens of C. batrachus were sacrificed and brain, liver and caudal white skeletal muscle were removed. Tissues were homogenized and fractions were obtained by differential centrifugation. The activities of citrate synthase (CS), glucose 6-phosphate dehydrogenase (G6-PDH) and lactate dehydrogease (LDH) were assayed spectrophotometrically. Similarly, DNA, RNA and protein content were measured as per standard procedure. Results The exposure of sublethal concentration of endosulfan decreased the activity of citrate synthase (CS) and glucose 6-phosphate dehydrogenase (G6-PDH) in the brain, liver and skeletal muscle of the freshwater catfish, C. batrachus. The brain lactate dehydrogenase (LDH) activity was also reduced in response to endosulfan toxicity. The maximum reduction in activities of these enzyme was 34%-43%. Withdrawal of endosulfan restored the enzyme activity to control level in all the three tissues. Vertical Decomposition Approach to Solve Single Stage Capacitated Warehouse Location Problem (SSCWLP)

Single Stage Capacitated Warehouse Location Problem (SSCWLP) has been attempted by few researchers in the past. These are Geoffrion and Graves [1], Sharma [2], Sharma [3] and Sharma and Berry [4]. In this paper we give a “vertical decomposition” approach to solve SSCWLP that uses Lagrangian relaxation. This way SSCWLP is broken into two versions of capacitated plant location problem (the CPLP_L and CPLP_R) by relaxing the flow balance constraints. For CPLP_R, we use well known Lagrangian relaxations given in literature (Christofides and Beasley [5] and Nauss [6]);and adopt them suitably for solving CPLP_L. We show theoretically in this paper that SSCWLP can be more efficiently solved by techniques of vertical decomposition developed in this paper than the method available in literature (Sharma and Berry [4]). Encouraging computational study is reported in this paper.

Medication adherence and quality of life among type-2 diabetes mellitus patients in India

Predictors of cerebrospinal fluid GeneXpert MTB/RIF positivity in patients with tuberculous meningitis:A prospective cohort study

Characterization of winged bean (<i>Psophocarpus tetragonolobus</i>(L.) DC.) based on molecular, chemical and physiological parameters

Winged bean (Psophocarpus tetragonolobus (L.) DC.) is a potential legume crop of the tropics with high protein and oil content in the seeds. Analysis of the mutual genotypic relationships among twenty four genotypes of P. tetragonolobus through Mantel test found a significant correlation (r = 0.839) between similarity matrices of the results obtained from the use of the RAPD and ISSR molecular markers. The UPGMA tree based on Jaccard’s similarity coefficient generated from their cumulative data showed two distinct clusters and seven sub-clusters among these accessions. Quantification of total polyphenols, flavonoids and tannin revealed the highest percentage of occurrence of kaempferol (1.07-790.5 μg/g) and the lowest percentage of gallic acid (0.09-3.49 μg/g) in the seeds. Phytochemical analysis of the winged bean genotypes revealed that, some of the exotic lines are distinct. Analysis of photosynthesis rate, photosynthetic yield and stomatal conductance data also showed two clusters and was in congruence with the phytochemical affinities of the genotypes. The overall high level of polymorphism and varied range of genetic distances across the genotypes revealed a wide range of genetic base of P. tetragonolobus. The present investigation therefore, has provided significant insights for further improvement of winged bean germplasm for its qualitative and quantitative traits.
