Quantitation of tadalafil in human plasma using a sensitive and rapid LC-MS/MS method for a bioequivalence study

A highly sensitive liquid chromatography-tandem mass spectrometry（LC-MS/MS） method was developed for the determination of tadalafil（TAD） in human plasma. TAD and its deuterated internal standard（IS）, tadalafil-d3, were extracted from 200 mL plasma using Phenomenex Strata-X-C 33 m extraction cartridges. Chromatographic analysis was carried out on Synergi Hydro-RP C18（100 mm × 4.6 mm, 4 mm）column with a mobile phase consisting of methanol and 10 m M ammonium formate, p H 4.0（90：10, v/v）,delivered at a flow rate of 0.9 m L/min. Quantitation of the protonated analyte was done on a triple quadrupole mass spectrometer using multiple reaction monitoring via electrospray ionization. The precursor to product ions transitions monitored for TAD and TAD-d3 were m/z 390.3-268.2 and m/z393.1-271.2, respectively. The calibration curve was linear over the concentration range of 0.50–500 ng/m L with correlation coefficient, r2 Z 0.9994. Acceptable intra-batch and inter-batch precision（r3.7%） and accuracy（97.8% to 104.1%） were obtained at five concentration levels. The recovery of TAD from spiked plasma was highly precise and quantitative（98.95% to 100.61%）. Further, the effect of endogenous matrix components was minimal. TAD was found to be stable under different storage conditions in human plasma and also in whole blood samples. The validated method was successfully used to determine TAD plasma concentration in a bioequivalence study with 20 mg TAD tablets in 24 healthy volunteers. Method performance was evaluated by reanalyzing 115 study samples.

The study of viruses and their genetics has been an opportunity as well as a challenge for the scientific *** recent ongoing SARSCov2(Severe Acute Respiratory Syndrome)pandemic proved the unpreparedness for these *** only the countermeasures for the effect caused by virus need to be tackled but the mutation taking place in the very genome of the virus is needed to be kept in check *** major way to find out more information about such pathogens is by extracting the genetic data of such *** genetic data of viruses have been cultured and stored as well as isolated in form of their genome sequences,there is still limited methods on what new viruses can be generated in future due to *** research proposes a deep learning model to predict the genome sequences of the SARS-Cov2 virus using only the previous viruses of the coronaviridae family with the help of RNN-LSTM(Recurrent Neural Network-Long ShortTerm Memory)and RNN-GRU(Gated Recurrent Unit)so that in the future,several counter measures can be taken by predicting possible changes in the genome with the help of existing mutations in the *** the process of testing the model,the F1-recall came out to be more than *** mutation detection’s accuracy of both the models come out about 98.5%which shows the capability of the recurrent neural network to predict future changes in the genome of virus.

Lithium/Sodium-ion batteries(LIB/SIB)have attracted enormous attention as a promising electrochemical energy storage system due to their high energy density and long cycle *** of the major hurdles is the initial irreversible capacity loss during the first few cycles owing to forming the solid electrolyte interphase layer(SEI).This process consumes a profusion of lithium/sodium,which reduces the overall energy density and cycle ***,a suitable approach to compensate for the irreversible capacity loss must be developed to improve the energy density and cycle ***-lithiation/sodiation is a widely accepted process to compensate for the irreversible capacity loss during the initial *** strategies such as physical,chemical,and electrochemical pre-lithiation/sodiation have been explored;however,these approaches add an extra step to the current manufacturing *** to these strategies,pre-lithiation/sodiation additives have attracted enormous attention due to their easy adaptability and compatibility with the current battery manufacturing *** this review,we consolidate recent developments and emphasize the importance of using pre-lithiation/sodiation additives(anode and cathode)to overcome the irreversible capacity loss during the initial cycles in lithium/sodium-ion *** review also addresses the technical and scientific challenges of using pre-lithiation/sodiation additives and offers the insights to boost the energy density and cycle life with their possible commercial *** most important prerequisites for designing effective pre-lithiation/sodiation additives have been explored and the future directions have been discussed.

Treatment of pediatric femoral shaft fractures by stainless steel and titanium elastic nail system： A randomized comparative trial

Purpose： Literature suggests that the lower modulus of elasticity of titanium makes it ideal for use in children compared with stainless steel. Better fracture stability was observed in association with tita- nium nails on torsional and axial compression testing. However, stainless steel nails are stiffer than ti- tanium counterparts, which may provide a rigid construct when fixing paediatric femoral shaft fractures. Complications have been observed more frequently by various researchers when titanium nails are used for fracture fixation in patients with increasing age or weight. The concept of this study was to compare the functional outcome after internal fixation with titanium elastic nail system and stainless steel elastic nail system in paediatric femoral shaft fractures. Methods： The study was conducted on 34 patients admitted in the department of orthopaedics, LLRM Medical College ＆ SVBP Hospital, Meerut, India from January 2013 to August 2014. We included patients aged 5-12 years with fracture of the femoral shaft, excluding compound fractures, pathological fractures and other lower limb fractures. Patients were treated by titanium （n = 17） or stainless steel （n - 17） elastic nail system and followed up for one year. The clinical parameters like range of motion at hip and knee joints, time to full weight bearing on the operated limb and radiological parameters like time to union were compared between two groups. A special note was made of intra- and post-operative complications. Functional outcomes were analysed according to Flynn criteria. Results： Based on the Flynn criteria, 59% of patients had excellent results, 41% had satisfactory results, and no one showed poor results. There was no clinically significant difference between the two groups with respect to time to union and full weight bearing. Sensitive and rapid determination of amantadine without derivatization in human plasma by LC–MS/MS for a bioequivalence study

A highly sensitive, rapid and rugged liquid chromatography-tandem mass spectrometw （LC-ESI-MS/MS） method was developed for reliable estimation of amantadine （AMD）, an antiviral drug in human plasma. The analyte and internal standard （IS）, amantadine-d6 （AMD-d6）, were extracted from 200μL plasma by solid phase extraction on Phenomenex Strata-X-C 33 ~t cartridges. Chromatography was performed on Synergi^TM Hydro-RP C18 （150 mm×4.6 mm, 4 μm） analytical column using a mixture of acetonitrile and 10 mM ammonium formate, pH 3.0 （80：20, v/v） as the mobile phase. Detection and quantitation was done by multiple reaction monitoring in the positive ionization mode for AMD （m/z 152.1→ 135.1 ） and IS （m/z 158.0 → 141.1） on a triple quadrupole mass spectrometer. The assay was linear in the concentration range of 0.50-500 ng/mL with correlation coefficient （r^2）〉 0.9969. The limit of detection of the method was 0.18 ng/mL The intra-batch and inter-batch precisions were 〈 5.42% and the accuracy varied from 98.47% to 105.72%. The extraction recovery of amantadine was precise and quantitative in the range of 97.89%-100.28%. IS-normalized matrix factors for amantadine varied from 0.981 to 1.012. The stability of AMD in whole blood and plasma was evaluated under different conditions. The developed method was successfully applied for a bioequivalence study with 100 mg of AMD in 32 healthy volunteers. The reproducibility of the assay was determined by reanalysis of 134 subject samples.

In the present work,ferrite(Fe)doped TiO_(2)thin films with different volume percentage(vol%)were synthesized using a spray pyrolysis *** effect of Fe doping on structural properties such as crystallite size,texture coefficient,microstrain,dislocation densities *** evaluated from the X ray diffratometry(XRD)*** data revealed a polycrystalline anatase TiO_(2)phase for sample synthesized up to 2 vol%and mixed anatase and rutile crystalline phase for sample synthesized at 4 vol%Fe doped TiO_(2).The crystalline size was observed to decrease with increase in Fe dopant vol%and also other structural parameters changes with Fe dopant *** the present work,electrical resistance was observed to decrease with a rise in Fe dopant vol%and temperature of the *** properties like temperature coefficient of resistance and activation energy also showed strong correlation with Fe dopant vol%.Humidity sensing properties of the synthesized sample altered with a change in Fe dopant vol%.In the present paper,maximum sensitivity of about 88.7%for the sample synthesized with 2 vol%Fe doped TiO_(2)and also the lowest response and recovery time of about 52 and 3 s were reported for the same sample.

The current work describes the synthesis of a new bio-waste derived cellulosic-carbon supportedpalladium nanoparticles enriched magnetic nanocatalyst(Pd/Fe_(3)O_(4)@C)using a simple multi-step process under aerobic *** mild reaction conditions,the Pd/Fe_(3)O_(4)@C magnetic nanocatalyst demonstrated excellent catalytic activity in the Hiyama cross-coupling reaction for a variety of ***,the Pd/Fe_(3)O_(4)@C magnetic nanocatalyst exhibited excellent catalytic activity up to five recycles without significant catalytic activity loss in the Hiyama cross-coupling ***,we explored the use of Pd/Fe_(3)O_(4)@C magnetic nanocatalyst as an electrocatalyst for hydrogen evolution ***,the Pd/Fe_(3)O_(4)@C magnetic nanocatalyst exhibited better electrochemical activity compared to bare carbon and magnetite(Fe_(3)O_(4)nanoparticles)with an overpotential of 293 mV at a current density of 10 mA·cm^(–2).
