The widespread adoption of bead-based multiplexed bioassays requires the ability to easily synthesize encoded microspheres and conjugate analytes of interest to their ***,we present a simple method(mRBLEs 2.0)for the efficient high-throughput generation of microspheres with ratiometric barcode lanthanide encoding(mRBLEs)that bear functional groups for downstream surface *** production in mRBLEs 2.0 relies on the manual mixing of lanthanide/polymer mixtures(each of which comprises a unique spectral code)followed by droplet generation using single-layer,parallel flow-focusing devices and the off-chip batch polymerization of droplets into *** streamline downstream analyte coupling,mRBLEs 2.0 crosslinks copolymers bearing functional groups on the bead surface during bead *** the mRBLEs 2.0 pipeline,we generate monodisperse mRBLEs containing 48 distinct well-resolved spectral codes with high throughput(>150,000/min and can be boosted to 450,000/min).We further demonstrate the efficient conjugation of oligonucleotides and entire proteins to carboxyl mRBLEs and of biotin to amino ***,we show that mRBLEs can also be magnetized via the simultaneous incorporation of magnetic nanoparticles with only a minor decrease in the potential code *** the advantages of dramatically simplified device fabrication,elimination of the need for custom-made equipment,and the ability to produce spectrally and magnetically encoded beads with direct surface functionalization with high throughput,mRBLEs 2.0 can be directly applied by many labs towards a wide variety of downstream assays,from basic biology to diagnostics and other translational research.

Homogentisic acid γ-lactone suppresses the virulence factors of Pseudomonas aeruginosa by quenching its quorum sensing signal molecules

Bacterial quorum sensing（QS） molecules are involved in the coordination of certain behaviors such as biofilm formation, virulence and antibiotic resistance. QS molecules（autoinducers） and their corresponding receptors have been recognized as important therapeutic targets for drug-resistant infections and biofilm-associated infections（BAI）. This study assessed the multiple biological effects of homogentisic acid y-lactone（HgAL）, a furanone derivative. The anti-QS and anti-biofilm effects of HgAL against PAO1 strain were evaluated using CLSm, SEm, HPLC and other biochemical methods The results showed that HgAL could effectively inhibit the production of pyocyanin and extracellular matrix, as well as reduce the adherence ability and biofilm formation of Pseudomonas aeruginosa. Inhibition of virulence is attributed to the suppressive effect of HgAL on biosynthesis of 3-oxo-C12-HSLand C4-HSL（two kinds of QS signaling molecules in P. aeruginosa）. Our results support HgAL as a potential agent for prevention of BAI in the healthcare settings.

The dynamics of a single strain HIV model is studied. The basic reproduction number R0 used as a bifurcation parameter shows that the system undergoes transcritical and saddle-node bifurcations. The usual threshold unit value of R0 does not completely determine the eradication of the disease in an HIV infected person. In particular, a sub-threshold value Rc is established which determines the system＇s number of endemic states： multiple if Rc 〈 Ro 〈 1, only one if Rc=Ro = 1, and none if R0 〈 Rc 〈 1.

Non-steroidal anti-inflammatory drugs and statins in relation to colorectal cancer risk

AIm： To investigate the association between individual or combined use of non-steroidal anti-inflammatory drugs （NSAIDs） or statins and colorectal cancer ***： In a population-based case-control study in women, we examined the association between NSAIDs and statin use and the risk of colorectal cancers. We further investigated whether the use of statins modifies the protective effect of NSAIDs. Female cases （n = 669）of colorectal cancer aged 50-74 years were identified from a storewide registry in Wisconsin during 1999-2001. Community control women （n = 1375） were randomly selected from lists of licensed drivers and medicare beneficiaries. medication use and risk factor information were gathered during a structured telephone interview. A multivariable logistic regression model was used to calculate odds ratio （OR） and 95% confidence interval （CI）.RESULTS： Overall, NSAIDs users had a 30% reduction in risk of colorectal cancer （95% CI： 0.56-0.88）. Statin use was not associated with colorectal cancer risk （OR = 1.17, 95% CI： 0.74-1.85）, regardless of structural type （lipophilic or hydrophilic）, duration of use, or recency. There was no evidence of an interaction between NSAIDs and statins and colorectal cancer risk （P-interaction = 0.28）.CONCLUSION： Although our results confirm the inverse association between NSAIDs use and colorectal cancer risk, they do not support a risk reduction in statin users, or an interaction effect of combined NSAIDs and statin use.

Emotionless Holism:Factor and Rasch Analysis of the Chinese Integrative medicine Attitude Questionnaire

T Objective： To examine the Eastem-Westem difference in the interpretation of Integrative medicine Attitude Questionnaire （ImAQ） by assessing the psychometric properties of a revised Chinese medicine （Cm）- specific version of ImAQ （Cm-ImAQ）. methods： Factor and Rasch analysis were performed with data collected from a mail survey of 165 Hong Kong Western medical doctors （WmD） randomly sampled from the official registry. The structural validity, unidimensionality, item fit, and differential item functioning （DIF） of the Hong Kong Cm-ImAQ were evaluated. Results： Confirmatory factor analysis （CFA） demonstrated that the original ImAQ factor structure was not concordant with our data on Chinese WmD, and subsequent explanatory factor analysis （EFA） validated a new three-factor model for Cm-ImAQ：（1） attitude towards tonification , （2） attitude towards the effectiveness of Cm, and （3） attitude towards Cm knowledge. The original ImAQ factor on holism and doctor-patient relationship disappeared. Rasch analysis confirmed the unidimensionality of tonification and the effectiveness domains, but further refinement of the knowledge domain is needed. Coaclusions： Cultural adaptation of the ImAQ has demonstrated differences between Eastern and Westem doctors trained in allopathic medicine in their interpretations of hoiism in healthcare. For Chinese WmD, the emphasis of holistic care is placed on tonifying＇ the body rather than on nurturing the mind and spidt. Confucian and Taoist conceptualizations of mental health as well as the persistent stigma towards mental illness within modern Chinese culture may explain why Chinese WmD do not regard mental health promotion as part of routine healthcare.

Current understanding and management of aggressive posterior retinopathy of prematurity

Aggressive posterior retinopathy of prematurity(ROP), previously referred to as "Rush disease", is a rapidly progressive form of ROP. This form of ROP typically presents in very low birth weight babies of early gestational age. Historically, anatomical and functional outcomes have been poor with standard treatment. This review is designed to discuss current knowledge and treatment regarding this aggressive form of ROP. Recommendations regarding management of these difficult cases are detailed.

Background:The prevalence of mRSA(methicillin-resistant Staphylococcus aureus)colonized wounds in home care residents is expected to grow continuously as a result of the substantial proportion of older people requiring institutionalized care due to chronic disease and declining functional status,which contribute to more frequent skin breakdown and wound *** tree oil has been claimed to have anti-bacterial,analgesic and anti-inflammatory effects that have been suggested in many in-vitro studies to have good efficacy against *** aims of this study were to evaluate the effectiveness of 10%topical tea tree preparation to eradicatemRSA and to ascertain its influence on wound healing formRSA-colonized ***:It was a randomized controlled trial,single-blind *** with stage II or above mRSA-colonized wounds and who had given their informed consent formed the *** determined sample sizewasbasedonthe effect size of our previouspilot study,whichwas *** outcome measurements were taken for the mRSA bacterial count and wound healing condition at baseline and at 1-week intervals during the 4-week dressing intervention ***:Thirty-two participants were recruited from two non-government nursing homes,16 in the control group and 16 in the tea tree oil *** control group residents received routine saline gauze dressing,while the tea tree oil group residents received the 10%topical tea tree preparation *** the tea tree oil group,all chronic wounds that had previously been delayed in healing were healed within 28 days without adverse *** was also completely eradicated in 14(87.5%)out of 16 wounds in the group receiving the 10%topical tea tree preparation.

Survival after inflammatory bowel disease-associated colorectal cancer in the Colon Cancer Family Registry

AIm: To investigate the survival of individuals with colorectal cancer (CRC) with inflammatory bowel disease (IBD-associated CRC) compared to that of individuals without IBD diagnosed with CRC. mETHODS: Epidemiologic, clinical, and follow-up data were obtained from the Colon Cancer Family Registry (Colon CFR). IBD-associated cases were identified from self-report of physician diagnosis. For a subset of participants, medical records were examined to confirm self-report of IBD. Cox proportional hazards regression was applied to estimate adjusted hazard ratios (aHR) and 95%CI of mortality, comparing IBD-associated to non-IBD-associated CRC, adjusted for age at CRC diagnosis, sex, Colon CFR phase, and number of prior endoscopies. Following imputation to complete CRC stage information, adjustment for CRC stage was examined. RESULTS: A total of 7202 CRC cases, including 250 cases of IBD-associated CRC, were analyzed. Over a twelve year follow-up period following CRC diagnosis, 2013 and 74 deaths occurred among non-IBD associated CRC and IBD-associated CRC patients, respectively. The difference in survival between IBD-associated and non-IBD CRC cases was not statistically significant (aHR = 1.08; 95%CI: 0.85-1.36). However, the assumption of proportional hazards necessary for valid inference from Cox regression was not met over the entire follow-up period, and we therefore limited analyses to within five years after CRC diagnosis when the assumption of proportional hazards was met. Over this period, there was evidence of worse prognosis for IBD-associated CRC (aHR = 1.36; 95%CI: 1.05-1.76). Results were similar when adjusted for CRC stage, or restricted to IBD confirmed in medical records. CONCLUSION: These results support the hypothesis that IBD-associated CRC has a worse prognosis than non-IBD-associated CRC.
