Screening and evaluation of human single-chain fragment variable antibody against hepatitis B virus surface antigen

BACKGROUND: Phage display technology has become a vital tool in studies aimed at identifying molecules binding to a specific target. It enables the rapid generation and selection of high affinity, fully human antibody product candidates to essentially any disease target appropriate for antibody therapy. In this study, we prepared the recombinant single-chain fragment variable ( ScFv) antibody to hepatitis B virus surface antigen (HBsAg) by the phage display technology for obtaining a virus-targeting mediator. METHODS: mRNA was isolated from B-lymphocytes from a healthy volunteer and converted into cDNA. The fragment variables of heavy and light chain were amplified separately and assembled into ScFv DNA with a specially constructed DNA linker by polymerase chain reaction. The ScFv DNA was ligated into the phagmid vector pCANT-AB5E and the ligated sample was transformed into competent E. coli TG1. The transformed cells were infected with M13K07 helper phage to form a human recombinant phage antibody library. The volume and recombinant rate of the library were evaluated by bacterial colony count and restriction analysis. After two rounds of panning with HBsAg. the phage clones displaying ScFv of the antibody were selected by enzyme-linked immunosorbant assay ( ELISA) from the enriched phage clones. The antigen binding affinity of the positive clone was detected by competition ELISA. HB2151 E. coli was transfected with the positive phage clone demonstrated by competition ELISA for production of a soluble form of the anti-HBsAg ScFv. ELISA assay was used to detect the antigen binding affinity of the soluble anti-HBsAg ScFv. Finally, the relative molecular mass of soluble anti-HBsAg ScFv was measured by SDS-PAGE. RESULTS: The variable heavy ( VH ) and variable light (VL) and ScFv DNAs were about 340bp, 320bp and 750bp, respectively. Design of Bandgap Reference in Switching Power Supply

A bandgap voltage reference is designed to meet the requirements of low power loss,low temperature coefficient and high power source rejection ratio(PSRR) in the intergrated circuit. Based on the analysis of conventional bandgap reference circuit,and combined with the integral performance of IC,the specific design index of the bandgap reference is put forward. In the meantime,the circuit and the layout are designed with Chartered 0.35 μm dual gate CMOS process. The simulation result shows that the coefficient is less than 30ppm/℃ with the temperature from -50 ℃ to 150 ℃. The bandgap reference has the characteristics of low power and high PSRR.

利用对流参数化模型对8颗s+r星的研究

目前观测到的s+r星数目正逐渐增多,为了进一步确定s+r星的形成机制以及它们与s-星和r-星的关系,采用对流模型的参数化方法拟合了新观测到的8颗s+r星的观测数据,拟合结果与观测符合得较好,进一步验证了原有模型的有效性和正确性.

Time-dependent color-tunable room temperature phosphorescence from unusual conformational transitions in phenothiazine polymers under UV irradiation

Achieving color-tunable room-temperature phosphorescence(RTP),especially including blue RTP from a single-component polymer still faces a formidable ***,we wisely choose conformation-dependent phenothiazine with trifluoromethyl substituent as the side group of the phosphor monomer(Cz PT) and then copolymerize it with N-isopropylacrylamide(NIPAM) through photopolymerization to obtain polymers PPCz ***-dependent color-tunable phosphorescence from unusual quasi-equatorial(eq) to quasi-axial(ax) conformers are obtained,and the RTP color changes from orange(550 nm) to blue(470 nm) with phosphorescence lifetime up to 0.96 *** theoretical calculations confirm that the quasi-axial conformer is the preferred structure that facilitates the formation of intramolecular hydrogen bonds on the trifluoromethyl *** EPR spectra illustrate that the persistent UV irradiation generates radical cations to induce the conformational transitions first,followed by photopolymerization immobilizing the ax conformation in PPCz *** of data encryption and anti-counterfeiting are fabricated to show prompt and delayed multicolor *** work affords a simple and feasible avenue for two-dimensional color tunable room temperature phosphorescence from a single-component polymer.

Objective:To investigate the protective effects of stir-fried Semen Armeniacae Amarum(SAA)against aristolochic acidⅠ(AAⅠ)-induced nephrotoxicity and DNA adducts and elucidate the underlying mechanism involved for ensuring the safe use of Asari Radix et ***:In vitro,HEK293T cells overexpressing Flag-tagged multidrug resistance-associated protein 3(MRP3)were constructed by Lentiviral transduction,and inhibitory effect of top 10 common pairs of medicinal herbs with Asari Radix et Rhizoma in clinic on MRP3 activity was verified using a self-constructed fluorescence screening *** m RNA,protein expressions,and enzyme activity levels of NAD(P)H quinone dehydrogenase 1(NQO1)and cytochrome P4501A2(CYP1A2)were measured in differentiated Hepa RG *** toxicity after inhibition of AAⅠmetabolite transport was detected using cell counting kit-8 *** vivo,C57BL/6 mice were randomly divided into 5 groups according to a random number table,including:control(1%sodium bicarbonate),AAⅠ(10 mg/kg),stir-fried SAA(1.75 g/kg)and AAⅠ+stir-fried SAA(1.75 and 8.75 g/kg)groups,6 mice in each *** 7 days of continuous gavage administration,liver and kidney damages were assessed,and the protein expressions and enzyme activity of liver metabolic enzymes NQO1 and CYP1A2 were determined ***:In vivo,combination of 1.75 g/kg SAA and 10 mg/kg AAⅠsuppressed AAⅠ-induced nephrotoxicity and reduced d A-ALI formation by 26.7%,and these detoxification effects in a dose-dependent manner(P<0.01).Mechanistically,SAA inhibited MRP3 transport in vitro,downregulated NQO1 expression in vivo,increased CYP1A2 expression and enzymatic activity in vitro and in vivo,respectively(P<0.05 or P<0.01).Notably,SAA also reduced AAⅠ-induced hepatotoxicity throughout the detoxification process,as indicated by a 41.3%reduction in the number of liver adducts(P<0.01).Conclusions:Stirfried SAA is a novel drug candidate for the suppression of AAⅠ-induced liver and kidney dam

Thermal Analysis of High Power LED on Heat Sink

Thermal management of LED junction temperature is one of the fundamental technologies for LED lamp to ensure basic specifications in many aspects. Analysed is the high power LED's distribution on heat sink. Using mathematical statistical methods, a formula is conlcuded to calculate the size of heat sink under LED safe working temperature, which provides a method to researchers and LED lamp manufacturers.
