InterStoreDB:A Generic Integration Resource for Genetic and Genomic Data

Associating phenotypic traits and quantitative trait loci （QTL） to causative regions of the underlying genome is a key goal in agricultural research. InterStoreDB is a suite of integrated databases designed to assist in this process. The individual databases are species independent and generic in design, providing access to curated datasets relating to plant populations, phenotypic traits, genetic maps, marker loci and QTL, with links to functional gene annotation and genomic sequence data. Each component database provides access to associated metadata, including data provenance and parameters used in analyses, thus providing users with information to evaluate the relative worth of any associations identified. The databases include CropStoreDB, for management of population, genetic map, QTL and trait measurement data, SeqStoreDB for sequence-related data and AlignStoreDB, which stores sequence alignment information, and allows navigation between genetic and genomic datasets. Genetic maps are visualized and compared using the CMAP tool, and functional annotation from sequenced genomes is provided via an EnsEMBL-based genome browser. This framework facilitates navigation of the multiple biological domains involved in genetics and genomics research in a transparent manner within a single portal. Thermal characterization of bio-based phase changing materials in decorative wood-based panels for thermal energy storage

Decorative wood panels containing pouches of bio-based phase changing materials(PCMs) were prepared. Three different PCM mixtures were used: a blend of capric and lauric acids as well as two commercial products, Puretemp?20 and Puretemp?23(Puretemp). The panels consist of engraved Medium Density Fiberboard(MDF) filled with a plastic pouch filled with PCM. High density fiberboard(HDF) was used on top of the panels to enclose the PCM pouches. PCM mixtures were first tested by differential scanning calorimetry(DSC). Phase change temperature and total heat storage of the panels were measured for both fusion and solidification with a Dynamic Heat-Flow Meter Apparatus(DHFMA). DSC and DHFMA results were compared, allowing a better understanding of results gathered from these two techniques. DSC calibration has been revealed important when assessing PCMs. The panels present a phase change temperature and a latent heat storage suitable for buildings applications. The panel made with Puretemp?23 presented the highest energy, with 57.1 j g^(-1). Thermal cycling was conducted on the panels to investigate thermal reliability, which revealed small modifications of thermal properties for two products. For all cases, latent heat was found stable. Hygro-mechanical behavior of the panels was also evaluated as these where designed to be esthetic decorative panels. This study exposes the potential of a new type of wood-based panels loaded with PCM for thermal energy storage and brings overall knowledge about PCM products thermal characterization.

The AFB1 auxin receptor controls the cytoplasmic auxin response pathway in Arabidopsis thaliana

The phytohormone auxin triggers root growth inhibition within seconds via a *** members of the TIR1/AFB auxin receptor family,AFB1 has a primary role in this rapidresponse. However, the unique features that confer this specific function have not been *** we show that the N-terminal region of AFB1, including the F-box domain and residues thatcontribute to auxin binding,is essential and sufficient for its specific role in the rapid response. Substitutionof the N-terminal region of AFB1 with that of TIR1 disrupts its distinct cytoplasm-enriched localizationand activity in rapid root growth inhibition by auxin. Importantly, the N-terminal region of AFB1 isindispensable for auxin-triggered calcium influx, which is a prerequisite for rapid root growth ***, AFB1 negatively regulates lateral root formation and transcription of auxin-induced genes,suggesting that it plays an inhibitory role in canonical auxin signaling. These results suggest that AFB1may buffer the transcriptional auxin response, whereas it regulates rapid changes in cell growth thatcontributeto rootgravitropism.

Multiple sclerosis（MS） is characterized by chronic inflammation in conjunction with neurodegeneration within the central nervous system. Most individuals with MS begin with a relapsing remitting course that later transitions to secondary progressive MS. Currently available disease-modifying therapies（DMTs） for relapsing MS have been demonstrated to reduce disease activity, however most patients require a change in therapy over the course of their disease. Treatment goals include the prevention of relapses and disability accumulation and to achieve this objective requires careful planning. Sequencing of DMTs for individual patients should be designed in such a way to maximize disease control and minimize risk based on the mechanism of action, pharmacokinetic and pharmacodynamic properties of each therapy. This includes the DMT patients are being switched from to those they are being switched to. The reversibility of immune system effects should be a key consideration for DMT sequence selection. This feature varies across DMTs and should factor more prominently in decision making as newer treatments become available for the prevention of disability accumulation in patients with progressive MS. In this short review, we discuss the landscape of existing therapies with an eye to the future when planning for optimal DMT sequencing. While no cure exists for MS, efforts are being directed toward research in neuroregeneration with the hope for positive outcomes.

Chemical characterization of fine and ultrafine PM, direct and indirect genotoxicity of PM and their organic extracts on pulmonary cells

Particulate matter in ambient air constitutes a complex mixture of fine and ultrafine particles composed of various chemical compounds including metals, ions, and organics. A multidisciplinary approach was developed by studying physico-chemical characteristics and mechanisms involved in the toxicity of particulate atmospheric pollution. PM0.3-2.s and PM2.5 including ultrafine particles were sampled in Dunkerque, a French industrialized seaside city. PM samples were characterized from a chemical and toxicological point of view. Physico-chemical characterization evidenced that PM2.5 comes from several sources： natural ones, such as soil resuspension and marine sea-salt emissions, as well as anthropogenic ones, such as shipping traffic, road traffic, and industrial activities. Human BEAS-2B tung cells were exposed to PM0.3-2.5, or to the Extractable Organic Matter （EOM） of PM0.3-2.5 and PM2.s. These exposures induced several mechanisms of action implied in the genotoxicity, such as oxidative DNA adducts and DNA Damage Response. The toxicity of PM-EOM was higher for the sample including the ultrafine fraction （PM2.5） containing also higher concentrations of polycyclic aromatic hydrocarbons. These results evidenced the major role of organic compounds in the toxicity of PM.

The role of externally-modulated electrostatic interactions in amplifying charge transport across lysine-doped peptide junctions

Many evolved biomolecular functions such as ion pumping or redox catalysis rely on controlled charge transport through the polypeptide matrix, which can be regulated by shifts in molecular protonation states and dependent supramolecular packing modes in response to environmental cues. However, the exact roles of such dynamic, non-covalent interactions in peptide charge transport have remained elusive. To tackle this challenge, here we report the modulation of charge transport in a series of lysine(Lys)-substituted hepta-glycine(Gly) peptide self-assembled monolayers(SAMs) on template-striped gold(Au^(TS)) bottom electrodes with eutectic gallium-indium(EGaIn) liquid metal top electrodes. We demonstrate systematic modulation of hydrogen bonding and more general electrostatic interactions by shifting the position of the charged Lys-residue and creating different protonation patterns by changing the environmental pH in the Au^(TS)/peptide//Ga Ox/EGa In junctions. The effective modulation is evidenced by current density-voltage(j-V) measurements combined with SAM characterization using ultraviolet photoelectron spectroscopy(UPS) and angle-resolved X-ray photoelectron spectroscopy(ARXPS), polarization modulation-infrared reflection-absorption spectroscopy(PM-IRRAS), and molecular dynamics(MD) simulations. Decreasing the hydrogen bonding inside the peptide SAMs and increasing the electrostatic interactions by environmental counterions amplifies the charge transport differently with Lys-position, which means that the sensitive electrical response of peptide SAMs can be tuned by the peptide sequence. Our results provide insights into the relationship between molecular design and in situ modulation of charge transport properties for the development of bionanoelectronics.

Have some landscapes in the eastern Canadian boreal forest moved beyond their natural range of variability?

Background: In the contxt of ecosystem management, the present study aims to compare the natural and the present-day forested landscapes of a large territory in Quebec(Canada). Using contemporary and long-term fire cycles, each natural forst landscape is defined according to the variability of its structure and composition, and compared to the present-day landscape. This analysis was conducted to address the question of whether human activities have moved these ecosystems outside the range of natural landscape ***: The study encompassed a forested area of 175 000 km2 divided into 14 landscapes. Using a framework that integrates fire cycles, age structure and forest dynamics, we characterized the forest composition and age structures that resulted from three historical fire cycles(110,140, and 180 years) representative of the boreal forest of eastern Canada. The modeled natural landscapes were compared with present-day landscapes in regard to the proportion of old-growth forests(landscape level) and the proportion of late-successional forest stands(landscape level and potential vegetation type).Results: Four landscapes(39%) remain within their natural range of variability. In contrast, nine landscapes(54%)show a large gap between natural and present-day landscapes. These nine are located in the southern portion of the study area, and are mainly associated with Abies-Betula vegetation where human activities have contributed to a strong increase in the proportion of Populus tremuloides stands(early-successional stages) and a decrease of oldgrowth forest stands(more than 100 years old).
