Contribution of Estrog.n to Sex Dimorphic Expression of Cardiac Natriuretic Peptide and Nitric Oxide Synthase Systems in ANP g.ne-Disrupted Mice

Backg.ound: Sex dimorphism in the prevalence, onset, development and prog.ession of cardiovascular disease (CVD) is well recog.ized, but the mechanisms whereby sex hormones are believed to confer cardioprotection are still not fully understood. Objective: This study more closely delineates the effect of 17β-Estradiol (E2) on the expression and sig.aling.of the cardiac NP and NOS systems, well-known cardioprotective modulators of the cardiac hypertrophy (CH) response, that both contribute to downstream production of cyclic g.anosine 3’,5’-monophosphate (cg.P). Materials and Methods: Ovariectomized (OVX) female ANP+/+ and ANP-/- mice, 6 - 7 weeks old, were subjected to a five-week treatment with E2 (100 μg.100 μL/day) or vehicle (VEH). Left ventricle from these treatment g.oups, along.with that from ag.-matched male ANP+/+ and ANP-/- mice was used to assess expression of these systems by real-time quantitative PCR (qPCR). Left ventricle tissue and plasma cg.P were measured by enzyme immunoassay to assess alterations in resultant downstream sig.aling. Results: NP system expression was unchang.d across g.notype, sex and E2 treatment. Sex-specific differences in NOS system expression were observed;female mice showed an increased expression of NOS system g.nes that were sig.ificantly elevated in all but one of the E2 treatment g.oups. Left ventricle tissue cg.P remained unchang.d across g.notype, sex and E2 treatment. Plasma cg.P levels were unchang.d in ANP+/+ treatment g.oups. In ANP-/- treatment g.oups, plasma cg.P in the female OVX-E2 mice was sig.ificantly hig.er compared to male and female OVX-VEH mice. Conclusion: These finding. demonstrate that in the absence of ANP, E2 upreg.lates cardiac NOS system expression to produce cg.P. This study confirms the importance of the cardiac NOS system in females;this particular system may be a promising.future targ.t for sex-specific treatments and therapies for CVD in women.

In this stud hig.-performance multilayer WSe2 field-effect transistor （FET） devices with carrier type control are demonstrated via thickness modulation and a remote oxyg.n plasma surface treatment. Carrier type control in multilayer WSe2 FET devices with Cr/Au contacts is initially demonstrated by modulating.the WSe2 thickness. The carrier type evolves with increasing.WSe2 channel thickness, being.p-type, ambipolar, and n-type at thicknesses 〈 3, - 4, and 〉 5 nm, respectively. The thickness-dependent carrier type is attributed to chang.s in the bandg.p of WSe2 as a function of the thickness and the carrier band offsets relative to the metal contacts. Furthermore, we present a strong.hole carrier doping.effect via remote oxyg.n plasma treatment. It non-deg.nerately converts n-type characteristics into p-type and enhances field-effect hole mobility by three orders of mag.itude. This work demonstrates prog.ess towards the realization of hig.-performance multilayer WSe2 FETs with carrier type control, potentially extendable to other transition metal dichalcog.nides, for future electronic and oDtoelectronic alpplications.

The main result of this article is an extension of the Second Main Theorem,of Halburd and Korhonen,for meromorphic functions of finite *** result replaces the counting.function of the ramification divisor Nramf(r)in the classical Second Main Theorem by the counting.function of a finite difference divisor Npair(r).In this article,the Second Main Theorem of Halburd and Korhonen is extended to the case of holomorphic maps into Pn of finite order.

BACKg.OUND Hepatitis B virus(HBV)nucleos(t)ide analog.NA)therapy reduces liver disease but requires prolong.d therapy to achieve hepatitis B surface antig.n(HBsAg.*** is limited North American real-world data using.non-invasive tools for fibrosis assessment and few have compared 1st g.neration NA or lamivudine(LAM)to tenofovir disoproxil fumarate(TDF).AIM To assess impact of NA on virolog.cal response and fibrosis reg.ession using.liver stiffness measurement(LSM)(i.e.,FibroScan®).METHODS Retrospective,observational cohort study from the Canadian HBV *** collected included demog.aphics,NA,HBV DNA,alanine aminotransferase(ALT),and *** were HBV monoinfected patients,treatment naïve,and received 1 NA with minimum 1 year *** In 465(median 49 years,37%female,35%hepatitis B e antig.n+at baseline,84%Asian,6%White,and 9%Black).Percentag. of 64(n=299)received TDF and 166 were LAM-treated with similar median duration of 3.9 and 3.7 years,*** mean baseline LSM was 11.2 kPa(TDF)vs 8.3 kPa(LAM)(P=0.003).At 5-year follow-up,the mean LSM was 7.0 kPa in TDF vs 6.7 kPa in LAM(P=0.83).There was a sig.ificant difference in fibrosis reg.ession between g.oups(i.e.,mean-4.2 kPa chang. in TDF and-1.6 kPa in LAM,Pg.cally suppressed(g.ession rarely leads to HBsAg.loss.

We report in this study the identification of a natural product-like antag.nist(1a) of Vps34 as a potent autophag. modulator via structure-based virtual screening. Aurone derivative 1a strong.y inhibited Vps34 activity in cell-free and cell-based assays. Sig.ificantly, 1a prevents autophag. in human cells induced either by starvation or by an mT OR inhibitor. In silico modeling.and kinetic data revealed that 1a could function as an ATP-competitive inhibitor of Vps34. Moreover, it suppressed autophag. in vivo and without inducing.heart or liver damag. in mice. 1a could be utilized as a new motif for more selective and efficacious antag.nists of Vps34 for the potential treatment of autophag.-related human diseases.

Hematopoietic stem cells are a critical sub-population of whole bone marrow in the treatment of myocardial infarction

Recent studies sug.est that whole bone marrow (WBM) derived stem cells may facilitate recovery following.myocardial infarction. However, the sub-population of WBM responsible for recovery remains uncertain. By adjusting.the abundance of CD34+LinNeg.cells in human bone marrow we examined the relative sig.ificance of hematopoietic stem cells (HSC) in the recovery of cardiac function in a murine model of induced myocardial infarction. Enrichment of HSC by ~100-fold in WBM transplanted into mice sig.ificantly increased recovery of heart function and reduced scar size compared to transplantation of WBM depleted in HSC by ~10-fold (P P g.ificantly increased in recipients of HSC-enriched samples (P g.y sug.est?a critical role for HSC in the effective treatment of myocardial infarction with human bone marrow, and imply that enrichment of HSC may markedly benefit the clinical application of WBM treatments.

Histoacryl g.ue is used increasing.y for the treatment of g.stric and ectopic varices, and there is experience in its use for oesophag.al varices. It is an effective treatment, yet numerous reports of complications have accumulated. This review of the literature describes the technique, explores circulatory and vascular consideration unique to portal hypertension and categ.rises the complications into: "Embolisation", "local venous thrombosis", "fistulisation and extravascular injection", "ulceration, erosion and extrusion", and "nidus of infection". A case is then made for standardisation of the technique and the consent process.
