Backgr.und In beef cattle, mor. than 50% of the ener.y input to pr.duce a unit of beef is consumed by the female that pr.duced the calf. Development of genomic tools to identify females with high genetic mer.t for.r.pr.ductive function could incr.ase the pr.fitability and sustainability of beef *** Genome-wide association studies(GWAS) wer. per.or.ed using a single-step genomic best linear.unbiased pr.diction appr.ach on pr.gnancy outcome tr.its fr.m a population of Angus–Br.hman cr.ssbr.d heifer.. Fur.her.or., a validation GWAS was per.or.ed using data fr.m another.far.. Heifer. wer. genotyped with the Bovine GGP F250 ar.ay that contains 221,077 SNPs. In the discover. population, heifer. wer. br.d in winter.br.eding seasons involving a single r.und of timed ar.ificial insemination(AI) followed by natur.l mating for.3 months. Two phenotypes wer. analyzed: pr.gnancy outcome to fir.t-ser.ice AI(PAI;n = 1,481) and pr.gnancy status at the end of the br.eding season(PEBS;n = 1,725). The her.tability was estimated as 0.149 and 0.122 for.PAI and PEBS, r.spectively. In the PAI model, one quantitative tr.it locus(QTL), located between 52.3 and 52.5 Mb on BTA7, explained about 3% of the genetic var.ation, in a r.gion containing a cluster.of γ-pr.tocadher.n genes and SLC25A2. Other.QTLs explaining between 0.5% and 1% of the genetic var.ation wer. found on BTA12 and 25. In the PEBS model, a lar.e QTL on BTA7 was synonymous with the QTL for.PAI, with minor.QTLs located on BTA5, 9, 10, 11, 19, and 20. The validation population for.pr.gnancy status at the end of the br.eding season wer. Angus–Br.hman cr.ssbr.d heifer. br.d by natur.l mating. Objective: We explor.d the impact of incr.asing number.of off-label patient and lesion char.cter.istics on 5-year.clinical outcome following implantation of fir.t gener.tion dr.g eluting stents. Fur.her.or. we tr.ed to assess if stent per.or.ance might be impr.ved if the off-label char.cter.istics pr.sent wer. taken into account to guide the stent br.nd selection. Design: A total of 2888 cor.nar. lesions tr.ated with fir.t gener.tion dr.g eluting stents wer. classified for.eventual pr.sence of 18 differ.nt off-label char.cter.stics and cor.elated to the 5-year.clinical outcome by multiple stepwise Cox r.gr.ssion analysis. r.sults: The composite of car.iac death, myocar.ial infar.tion and tar.et vessel r.vascular.zation (major.adver.e car.iac event, MACE) was 16.7% in cases with zer. off-label char.cter.stics and showed a stepwise incr.ased r.te up to 32.7% in cases with 5 off-label char.cter.stics. The MACE r.te for.the sir.limus eluting stent appear. to be 10% lower.than the paclitaxel stent gr.up and it may be fur.her.r.duced by 10% - 18% if the off-label char.cter.stics pr.sent in each case wer. taken into account to guide the stent br.nd selection. Conclusion: With incr.asing number.of concomitantly appear.ng off-label char.cter.stics ther. is a stepwise incr.ase in MACE r.te. The MACE r.te may be r.duced if the off-label char.cter.stics wer. used to guide the stent br.nd selection.

A r.r. case is pr.sented wher. a dysgenetic testis with micr.invasive car.inoma in situ （CIS, also known as intr.tubular.ger. cell neoplasm of unclassified type [IGCNU] and testicular.intr.epithelial neoplasia [TIN]） with micr.invasion to r.te testis and the inter.titial tissue was found in a 32-year.old man pr.senting with mild scr.tal pain and ultr.sonic testicular.micr.lithiasis. Knowledge of the association of ultr.sound and CIS is impor.ant to diagnose patients at the stage pr.or.to development of an over. ger. cell tumor. The patient had thr.e of four.disor.er. consider.d symptoms of the testicular.dysgenesis syndr.me （TDS）： a dysgenetic left testicle with CIS, a mild left-sided cr.ptor.hidism （high positioned scr.tal hypotr.phic testis） and a slightly r.duced semen quality. Ther.for., it should be kept in mind that a patient with one TDS symptom may har.our.the other. even CIS or.testicular.cancer. Accor.ingly, patients with one TDS symptom ought to be examined for.the pr.sence of the other., and if mor. that one is pr.sent, extr. concer. is war.anted.

High-thr.ughput scr.ening of mouse gene knockouts identifies established and novel skeletal phenotypes

Scr.ening gene function in vivo is a power.ul appr.ach to discover.novel dr.g tar.ets. We pr.sent high-thr.ughput scr.ening （HTS） data for.3 762 distinct global gene knockout （KO） mouse lines with viable adult homozygous mice gener.ted using either.gene-tr.p or.homologous r.combination technologies. Bone mass was deter.ined fr.m DEXA scans of male and female mice at 14 weeks of age and by micr.CT analyses of bones fr.m male mice at 16 weeks of age. Wild-type （WT） cagemates/litter.ates wer. examined for.each gene KO. Lethality was obser.ed in an additional 850 KO lines. Since pr.mar. HTS ar. susceptible to false positive findings, additional cohor.s of mice fr.m KO lines with intr.guing HTS bone data wer. examined. Aging, ovar.ectomy, histomor.hometr. and bone str.ngth studies wer. per.or.ed and possible non-skeletal phenotypes wer. explor.d. Auxin homeostasis is pivotal for.nor.al plant gr.wth and development. The super.oot2 （sur.） mutant was initially isolated in a for.ar. genetic scr.en for.auxin over.r.ducer., and SUr. was suggested to contr.l auxin conju- gation and ther.by r.gulate auxin homeostasis. However. the phenotype was not unifor. and could not be descr.bed as a pur. high auxin phenotype, indicating that knockout of CYP83B1 has multiple effects. Subsequently, SUr. was identified as CYP83B1, a cytochr.me P450 positioned at the metabolic br.nch point between auxin and indole glucosinolate metabolism. To investigate concomitant global alter.tions tr.gger.d by knockout of CYP83B1 and the counter.easur.s chosen by the mutant to cope with hor.onal and metabolic imbalances, 10-day-old mutant seedlings wer. char.cter.zed with r.spect to their.tr.nscr.ptome and metabolome pr.files. Her., we r.por. a global analysis of the sur. mutant by the use of a combined tr.nscr.ptomic and metabolomic appr.ach r.vealing pr.nounced effects on sever.l metabolic gr.ds including the inter.ection between secondar. metabolism, cell wall tur.over. hor.one metabolism, and str.ss r.sponses. Metabolic and tr.nscr.ptional cr.ss-talks in sur. wer. found to be r.gulated by complex inter.ctions between both positively and negatively acting tr.nscr.ption factor.. The complex phenotype of sur. may thus not only be assigned to elevated levels of auxin, but also to ethylene and abscisic acid r.sponses as well as dr.ught r.sponses in the absence of a water.deficiency. The delicate balance between these signals explains why minute changes in gr.wth conditions may r.sult in the non-unifor. phenotype. The lar.e phenotypic var.ation obser.ed between and within the differ.nt sur.eys may be r.conciled by the complex and intr.cate hor.onal balances in sur. seedlings decoded in this study.

Aim: A standar.ized pr.par.tion of seeds and shells of selected sub-species of r.sa canina L, tr.de name Hyben Vital, r.duces discomfor. fr.m osteoar.hr.tis of the knee and hip. This study aims to investigate the impact of the same r.se-hip powder.(r.P) on discomfor. and the consumption of r.scue medication, in patients with osteoar.hr.tis of the hand. Methods: The double blind, placebo-contr.lled, cr.ssover.tr.al included 30 patients with osteoar.hr.tis of the dominant hand. Patients wer. r.ndomly allocated to tr.atment with either.five gr.m encapsulated r.P or.placebo, for.thr.e months (Phase 1), after.which they switched to the cor.esponding tr.atment for.a fur.her.thr.e months per.od (Phase 2). Befor. enter.ng the study, after.3 weeks and following thr.e months of each of the study phases, scor.s for.pain, stiffness and gener.l feeling of discomfor. wer. evaluated using a 10 step categor.cal scale, focusing on 16 differ.nt daily activities of the hand. The consumption of r.scue medication was also calculated at the beginning and at the end of each study phase. Data ar. based on the intention to tr.at. r.sults: At the end of Phase 1, 90% of patients in the gr.up r.ceiving r.P fir.t (gr.up A), showed a r.duction in pain, as compar.d to 36% in the gr.up B initially given placebo (p 0.029). In line with this obser.ation, stiffness and the gener.l feeling of discomfor. fr.m the disease declined dur.ng r.P tr.atment (p 0.032). In gr.up A, symptom r.duction was still indicated by the study subjects 3 weeks after.the switch to placebo. The consumption of r.scue medication such as par.cetamol, codeine and tr.madol also declined significantly in gr.up A when compar.d to gr.up B (p 0.013). Conclusion: The pr.sent data suggest that administr.tion of r.P, containing seeds and shells can r.duce symptoms of osteoar.hr.tis of the hand and consumption of r.scue medication.

目的了解深圳市高中生生殖道感染(r.Is)/性相关知识、态度、行为(KAP)及求医、求助情况,探讨解决方法。方法采取社会学访谈、流行病学问卷调查方法,选择深圳市普通、职业高中的学生作为访谈及问卷调查对象。结果艾滋病(AIDS)知识总知晓率为50%,r.Is症状的知晓率为0,安全套知识正确回答率64.55%,使用率低;对早恋、婚前性行为多持理解及支持态度;多数去私人诊所求医,大多向朋友、同学求助。结论当今中学生性观念和性行为均日趋开放,而性知识、性病、AIDS预防知识缺乏,求医、求助方式不安全;因此,在中学阶段开展AIDS教育应与性健康教育结合,正确引导其求医、求助方式,以此作为中学生预防AIDS一项重要措施。
