Background:Uveal melanoma(UM)is the most common primary eye tumor in adults,and the most frequent site of malignant transformation of the melanocytes after the *** spreads to the liver in half of the cases,and the death rate following the report of metastasis is 92%at 2 *** tropism of UM cannot simply be explained by the blood circulatory system organization,and illustrates the“seed and soil”hypothesis that describes an interaction between tumor cells(seed)and a specific microenvironment(soil).We decided to focus our study on the synergic interaction between UM cells and hepatic stellate cells(HSC),whose role has been previously described in the metastatic progression of colon and pancreatic ***,HSC have been found surrounding UM liver metastasis,and the UM secretome contains activating cytokines of hepatic stromal *** hypothesis is that HSC provide a specific microenvironment in the liver enhancing the growth of UM cells and increasing their therapeutic *** an in vitro 3D model and an original xenograft mouse model,we aim to decipher the mechanisms of UM metastatic progression,in order to elaborate new therapeutic ***:First,using an agar coating,spheroids were generated with UM cells and were allowed to grow for 72 *** tumor spheroids were then embedded in Matrigel and the HSC conditioned medium was used to evaluate the impact of the HSC secretome on UM ***,an original in vivo xenograft mouse model was generated,in which metastatic UM cells were injected alone or with human HSC in the spleen of immunodeficient *** model allows us to evaluate in 3-6 weeks the metastatic potential of each cell population,and thus to determine the cooperation between HSC and UM cells in the ***:The HSC conditioned medium increased the invasion of UM spheroids compared to non-conditioned medium in our in vitro *** addition,UM cells inoculated in the mouse spleen alone or with human HSC we

爱德士牛怀孕检测ELISA试剂盒,以牛血清或血浆中存在的早期妊娠相关糖蛋白(PAGs)作为判断怀孕的标志。实验中总共使用78 3份血清样本和1 001份血浆样本。用于评估特异性的样本为经超声波或直肠检查确认的已配空怀牛以及产后60天以上和未配种的牛只。结果表明,授精后28天已确认怀孕牛的621份血浆样品和552血清样品敏感性均为99.5%。231份血清样品和380份血浆样品的特异性分别为93.1%和94.7%。产犊后,爱德士ELISA方法检测发现PAG水平下降(图5),产后60天检测血清(n=65)和血浆(n=90),PAGs含量值降到基准线以下,特异性为100%。

The FAIR principles have been widely cited,endorsed and adopted by a broad range of stakeholders since their publication in *** intention,the 15 FAIR guiding principles do not dictate specific technological implementations,but provide guidance for improving Findability,Accessibility,Interoperability and Reusability of digital *** has likely contributed to the broad adoption of the FAIR principles,because individual stakeholder communities can implement their own FAIR ***,it has also resulted in inconsistent interpretations that carry the risk of leading to incompatible ***,while the FAIR principles are formulated on a high level and may be interpreted and implemented in different ways,for true interoperability we need to support convergence in implementation choices that are widely accessible and(re)-*** introduce the concept of FAIR implementation considerations to assist accelerated global participation and convergence towards accessible,robust,widespread and consistent FAIR *** self-identified stakeholder community may either choose to reuse solutions from existing implementations,or when they spot a gap,accept the challenge to create the needed solution,which,ideally,can be used again by other communities in the ***,we provide interpretations and implementation considerations(choices and challenges)for each FAIR principle.

Fungi are highly important biotic components of terrestrial ecosystems,but we still have a very limited understanding about their diversity and *** data article releases a global soil fungal dataset of the Global Soil Mycobiome consortium(GSMc)to boost further research in fungal diversity,biogeography and *** dataset comprises 722,682 fungal operational taxonomic units(OTUs)derived from PacBio sequencing of full-length ITS and 18S-V9 variable regions from 3200 plots in 108 countries on all *** plots are supplied with geographical and edaphic *** OTUs are taxonomically and functionally assigned to guilds and other functional *** entire dataset has been corrected by excluding chimeras,index-switch artefacts and potential *** dataset is more inclusive in terms of geographical breadth and phylogenetic diversity of fungi than previously published *** GSMc dataset is available over the PlutoF repository.

本文主要介绍了2001年9月11日世界贸易中心(WTC)被攻击之后在各学科领域的环境特征研究结果。文中提交的信息被世界贸易中心应急小组在2001年9月18日(热点信息)和2001年9月27日(制图和组成成分结果研究)使用。实验数据由喷气推进实验室/美国航空航天局(JPL/nASA)于2001年9月16、18、22、23日在世界贸易中心(WTC)通过机载可见光/红外成像光谱仪(AVIRIS)的高光谱遥感仪器获得。2001年9月17日和18日晚上,由两人组成的美国地质勘探局小组在世界贸易中心方圆1km之内收集了超过35个位置点的粉尘和空降碎片样品,其中为了避免受9月14日的暴雨影响选择了两个室内样本收集点,另外还收集到了两个有钢梁涂层的世贸残骸样品。美国地质勘探局地面工作人员在白天进行了实地反射光谱测量,用于现场校准AVIRIS遥感数据。AVIRIS数据的辐射定标和校正工作在喷气推进实验室/美国航空航天局(JPL/nASA)完成;表面反射率校准、光谱测图和解译工作在位于丹佛市的美国地质调查局成像光谱实验室完成;灰尘/碎屑和钢梁涂层样品的各种矿物学和化学参数的分析工作,在位于科罗拉多州丹佛市的美国地质调查局实验室完成,分析中使用了反射光谱法(RS)、扫描电子显微镜检查法(SEM)以及X射线衍射法(XRD)等。通过分析研究得出以下几个重要结论:(1)世贸中心大楼倒塌释放的粉尘主要有:玻璃纤维、石膏、混凝土、纸张以及其他建筑施工中常用的混合材料。(2)在实验室用RS、SEM和XRD方法分析发现,在三分之二以上的灰尘和空降碎片样本中只检测到微量的温石棉(小于总重量的1%),而到目前为止在所有粉尘样品中还没有检测到角闪石类石棉矿物。(3)对钢梁涂层样品分析发现,在世贸中心碎片中检测到的温石棉(蛇纹石矿物)占涂层材料总体积的比例高达20%,同样在这些钢梁涂层样品中也没有检测到角闪石类石棉矿物。(4)在地面仪器低百分比检测的限制下,AVIRIS矿物图主要是用来检测可能存在的石棉状矿物,图中并没有显示温石棉或闪石类石棉的大范围分布。从图中的孤立像素点或像素群(像素大小约2m×2m)可以看出,世贸中心周围地区的石棉状矿物含量变化范围大,从百分之几到百分之几十都有可能。由于AVIRIS数据中的某些光谱吸收强度和温石棉钢梁涂层的光谱相似,因此吸收特征图只显示出了蛇纹石矿物的存在,而不能说明蛇纹石是石棉状的,因为非石棉状蛇纹石矿物在建筑材料中也是非常常见的。AVIRIS矿物图可以显示含石棉量较高的区域和小范围非石棉状矿物的区域,但需要通过现场采样和实验分析来验证石棉的存在。另外,AVIRIS矿物图只能检测地表矿物,而不能检测地下掩埋的矿物。(5)AVIRIS矿物图中角闪石类矿物是几个孤立像素点,而不是像温石棉那样的像素群。少数几个世贸中心地区的角闪石像素点和整个城市的角闪石像素统计噪声水平相似。AVIRIS吸收特征图只显示了角闪石矿物的存在,而这些在非石棉状建筑材料中也是非常常见的,所以需要进行现场采样来确认石棉的存在。野外样品的实验室分析结果与矿物图上的蛇纹石温石棉和角闪石矿物是一致的。(6)实验分析和AVIRIS矿物图结果表明,无论是小尺度的个别样品,还是世贸中心一定方向和距离范围的大空间尺度,粉尘的成分都是多变的。通过对同一样品多次重复进行矿物学和化学物质分析发现,多变性可能是因为粉尘是由不同材料的不均匀混合造成的,这种空间差异是在几十米的大尺度到厘米甚至更小尺度范围下观测到的。AVIRIS矿物图还显示了位于东南偏南的2号大楼倒塌中心的物质铁含量较高,温石棉的分布主要(但并不唯一)呈向西和北方向不连续的放射状,也可能分布在超过世贸大厦遗址四分之三公里的东方向。(7)迄今为止,在粉尘和空降样品中只检测到微量的温石棉,但在有钢梁涂层的世贸中心碎片中温石棉含量达到总体积的20%之多,而且AVIRIS矿物图中温石棉可能存在的区域也表明在局部点可能发现石棉。(8)2001年9月16日收集到的AVIRIS数据显示,世贸中心大楼倒塌区域有若干热点存在,在核心区就有超过36个不同大小和温度的热点。分析数据发现,这些热点的温度都高于800°F,有些甚至超过了1300°F。截止到9月23日,从飞机上观察到的火灾点大部分已经消除或者减弱。(9)我们通过实验发现的灰尘样品有微量石棉的结论与美国环保署(***)进行的其他研究结果一致。我们的研究结果也进一步说明:①高含量石棉可能存在于钢梁涂层和AVIRIS矿物图中显示的局部区域;②粉尘和钢梁涂层中的石棉只含有温石棉,而不含有角闪石类石棉。根据毒理学专家、流行病学专家和矿物学专家近20年分析研究的主要成果表明,闪石类石棉矿物比温石棉更易致癌(Ross,1999;Skinner等,1988;Van Oss等,19

Biliary complications in liver transplantation: Impact of anastomotic technique and ischemic time on short- and long-term outcome

AIM: To elucidate the impact of various donor recipient and transplant factors on the development of biliary complications after liver ***: We retrospectively reviewed 200 patients of our newly established liver transplantation(LT) program, who received full size liver graft. Biliary reconstruction was performed by side-to-side(SS), end-to-end(EE) anastomosis or hepeaticojejunostomy(HJ). Biliary complications(BC), anastomotic stenosis, bile leak, papillary stenosis, biliary drain complication, ischemic type biliary lesion(ITBL) were evaluated by studying patient records, corresponding radiologic imaging and reports of interventional procedures [e.g., endoscopic retrograde cholangiopancreatography(ERCP)]. Laboratory results included alanine aminotransferase(ALT), gammaglutamyltransferase and direct/indirect bilirubin with focus on the first and fifth postoperative day, six weeks after LT. The routinely employed external bile drain was examined by a routine cholangiography on the fifth postoperative day and six weeks after transplantation as a standard procedure, but also whenever clinically indicated. If necessary, interventional(e.g., ERCP) or surgical therapy was performed. In case of biliary complication, patients were selected, assigned to different complication-groups and subsequently reviewed in detail. To evaluate the patients outcome, we focussed on appearance of postoperative/post-interventional cholangitis, need for rehospitalisation, retransplantation, ITBL or death caused by ***: A total of 200 patients [age: 56(19-72), alcoholic cirrhosis: n = 64(32%), hepatocellular carcinoma: n = 40(20%), acute liver failure: n = 23(11.5%), cryptogenic cirrhosis: n = 22(11%), hepatitis B virus /hepatitis C virus cirrhosis: n = 13(6.5%), primary sclerosing cholangitis: n = 13(6.5%), others: n = 25(12.5%) were included.
