Gelatin based adhesives have been used in the last decades in different biomedical applications due to the excellent biocompatibility,easy processability,transparency,non-toxicity,and reasonable mechanical properties to mimic the extracellular matrix(ECM).Gelatin adhesives can be easily tuned to gain different viscoelastic and mechanical properties that facilitate its ocular *** herein grafted glycidyl methacrylate on the gelatin backbone with a simple chemical modification of the precursor,utilizing epoxide ring-opening reactions and visible *** chemical modification allows the obtaining of an elastic protein-based hydrogel(GELGYM)with excellent biomimetic properties,approaching those of the native *** can be modulated to be stretched up to 4 times its initial length and withstand high tensile stresses up to 1.95 MPa with compressive strains as high as 80%compared to Gelatin-methacryloyl(GeIMA),the most studied derivative of gelatin used as a *** is also highly biocompatible and supports cellular adhesion,proliferation,and migration in both 2 and 3-dimensional *** characteristics along with its super adhesion to biological tissues such as cornea,aorta,heart,muscle,kidney,liver,and spleen suggest widespread applications of this hydrogel in many biomedical areas such as transplantation,tissue adhesive,wound dressing,bioprinting,and drug and cell delivery.

Many syndromic forms of autism spectrum disorder(ASD) are caused by mutations in proteins such as PTEN, TSC1, TSC2, and FMRP, which lie within the m TOR signaling pathway. While recent work has primarily focused on the dysregulation of protein translation via the deletion of these genes, few studies have examined how disruption of autophagy, another major downstream branch of m TOR signaling,contributes to neuronal dysfunction and ASD pathogenesis. To this end, we and others have previously examined mice deficient for autophagy in forebrain excitatory neurons via the deletion of Atg7 by Ca MKII-cre and observed ASD-like behavioural abnormalities in addition to neuronal dysfunctions from the molecular to the network level. As ASD is believed to be caused by an imbalance between excitatory and inhibitory signals in the brain, we have further extended our analysis to examine disruption of autophagy in forebrain GABAergic interneurons. Similar to disrupted autophagy in other neuronal and non-neuronal cell types, Atg7 deletion by Dlx5-cre results in time-dependent ubiquitin+ and p62+ aggregate formation in affected neurons. Notably, Dlx5-cre Atg7 c KO mice exhibit an overlapping set of ASD-like behavioural abnormalities previously observed in Ca MKII-cre Atg7 c KO animals. Furthermore, we have detected a disruption of the excitatory-inhibitory balance in both Ca MKII-cre and Dlx5-cre Atg7 c KO mice. Using primary cultured neurons, a specific reduction of surface GABAA receptors was observed in Atg7 c KO neurons. Unbiased mass spectrometric and subsequent antibody-based analyses demonstrated that members of the GABARAP(GABA receptor-associated protein) protein family accumulate and mislocalize to p62+ aggregates formed in affected neurons of both Ca MKII-cre and Dlx5-cre Atg7 c KO brains. Why People Live in Flood-Prone Areas in Akuressa, Sri Lanka

To investigate why people live in areas at high risk of floods, a qualitative case study was carried out in the areas around Akuressa, in southwest Sri Lanka. Data collection consisted mainly of semistructured interviews with local residents and government officials. The purpose was to study why people live in areas at high risk of floods, by looking beyond the purely physical aspects of living with hazards and exploring the underlying social factors. Four main factors were identified: an overall good living situation; a sense of place; difficulties relocating; and being well-adapted to the situation. The analysis also examined whether government officials shared the views of local residents. The findings highlighted both areas of consensus and discrepancies related to risk awareness, and the efficiency of risk reduction measures that had been implemented by the government. The case study identified and explored underlying social factors, such as risk normalization, risk trade-offs, and push-and-pull processes, which seem to influence the decision to live in a high-risk area.

Characterizing gastrointestinal stromal tumors and evaluating neoadjuvant imatinib by sequencing of endoscopic ultrasound-biopsies

AIM To evaluate endoscopic ultrasound(EUS)-guided biopsies for the pretreatment characterization of gastrointestinal stromal tumors(GIST) to personalize the management of *** All patients with lesions suspected to be GIST who were referred for EUS-sampling at a tertiary Swedish center were eligible for inclusion 2006-2015. During the observational study phase(2006-2011), routine fine-needle-aspiration(EUS-FNA) was *** 2012-2015, we converted to an interventional, randomized protocol with dual sampling EUS-FNA and fine-needle-biopsy-sampling(EUS-FNB) for all lesions. c-KIT-and DOG-1-immunostaining was attempted in all samples and a manual count of the Ki-67-index was performed. FNB-sampled tissue and the resected specimens were subjected to Sanger sequencing of the KIT and platelet-derived growth factor alpha(PDGFRA) genes. RESULTS In all, 64 unique patients with GIST were included, and of these, 38 were subjected to pretreatment dual sampling. EUS-FNB had a higher diagnostic sensitivity when compared head-to-head with EUS-FNA(98% vs 58%, P < 0.001) and was more adequate for Ki-67-indexing(Ki-67EUS)(92% vs 40%, P < 0.001). Sequencing of EUS-biopsies was successful in 43/44(98%) patients, and the mutation profiles(KIT-mutation 73%, PDGFRA-mutation 18%, wild-type 7%) were fully congruent with those detected in the corresponding resected specimens. In imatinib-na?ve patients, the Ki-67_(EUS) was comparable with the Ki-67-index in the corresponding surgical specimens(Ki-67_(SURG))(2.7% vs 2.9%, P = 0.68). In patients treated with neoadjuvant imatinib who also carried mutations indicating sensitivity, the Ki-67 EUS was higher than the Ki-67_(SURG)(2.5% vs 0.2%, P = 0.005), with a significant reduction in the Ki-67-index of-91.5%(95%CI:-82.4 to-96.0, P = 0.005). Natural Regeneration on Planted Clearcuts—The Easy Way to Mixed Forest?

Establishing mixed stands by stimulating natural in combination with planting could potentially ensure adequate stocking and meet shifting demands for forest products. Two long-term experiments were separately established in former Norway spruce (Picea abies L. Karst) stands in southern Sweden to be able to evaluate mixed forest establishment and management. The treatment plots were treated with different intensity of soil scarification (disc trenching, soil inversion and control) to test effects of soil disturbance on regeneration success. Five years later, both experiments were used for a first evaluation, where seedlings of all naturally regenerated tree species were counted together with estimates of the cover and height of other vegetation. This was made in sample plots in symmetric grids. Testing the effects of scarification and distance to seed source on natural regeneration of silver birch (Betula pendula Roth.) and downy birch (B. pubescens Ehrh.) showed that both scarification type and degree of soil disturbance significantly affected seedling counts but only if seed sources were close. In conclusion, conventional management practices for regenerating coniferous forest may be suitable for establishing mixed forest stands with combinations of planted and naturally regenerated seedlings.

Background: The apolipoprotein E (APOE, gene;apoE, protein) ε4 allele is the most commonly identified genetic risk factor for typical late-onset sporadic Alzheimer’s disease (AD). Each APOE ε4 allele roughly triples the relative risk for AD compared to that of the reference allele, APOE ε3. Methods: We have employed hyperspectral fluorescence imaging with an amyloid-specific, conformation-sensing probe, p-FTAA, to elucidate protein aggregate structure and morphology in fresh frozen prefrontal cortex samples from human postmortem AD brain tissue samples from patients homozygous for either APOE ε3 or APOE ε4. Results: As expected APOE ε4/ε4 tissues had a significantly larger load of CAA than APOE ε3/ε3. APOE isoform-dependent morphological differences in amyloid plaques were also observed. Amyloid plaques in APOE ε3/ε3 tissue had small spherical cores and large coronas while amyloid plaques in APOE ε4/ε4 tissues had large irregular and multi-lobulated plaques with relatively smaller coronas. Despite the different morphologies of their cores, the p-FTAA stained APOE ε3/ε3 amyloid plaque cores had spectral properties identical to those of APOE ε4/ε4 plaque cores. Conclusions: These data support the hypothesis that one mechanism by which the APOE ε4 allele affects AD is by modulating the macrostructure of pathological protein deposits in the brain. APOE ε4 is associated with a higher density of amyloid plaques (as compared to APOE ε3). We speculate that multilobulated APOE ε4-associated plaques arise from multiple initiation foci that coalesce as the plaques grow.

The Natural Forest Protection Program in China： A Contingent Valuation Study in Heilongjiang Province

In 1998, the Chinese Government implemented the NFPP （Natural Forest Protection Program）, which included logging restrictions, protected areas, replanting, and a range of other policies aimed at safeguarding the state of the country＇s forests and reducing the risk of erosion and flooding. A second phase of this program is currently being discussed. In this paper, contingent valuation is used to estimate the WTP （willingness to pay） for maintaining the program among the inhabitants in Heilongjiang Province in northern China. The results show that, even with fairly conservative assumptions, the aggregated WTP for maintaining the program for another five years is some 3.24 billion yuan per year. This can be compared with the current cost of the Program in the province, which is some 1.57 billion yuan per year.

Techniques for microspore culture of Brassica napus have been improved rapidly, and embryogenesis has been achieved in a wide range of genotypes. Plants regenerated from microspore-derived embryoids can be haploid, diploid or polyploid. From rapeseed microspore culture it is reported that 70%-90% of regenerated plants are haploid. The usual methods of chromosome doubling involve soaking roots (most common) or whole plants in a colchicine solution, or culturing plantlets in colchicine-containing medium in the greenhouse. Other alternatives are injecting colchicine into the secondary buds or applying colchicine-soaked cotton plugs to axillary buds.
