Effect of pre-ischemia on hypoxia-inducible factor expression in the brain tissue of rats with cerebral ischemia/reperfusion injury

Hypoxia-inducible factor 1 （HIF-1） can lead to the adaptative reaction of body for hypoxia and ischemia. HIF-1 plays an important role in the response of ischemia-hypoxia. At present, there has been no overall report on the significance for the expression of HIF-1 following experimental cerebral ischemia. OBJECTIVE： To observe the expression of HIF-1 after middle cerebral artery occlusion （MCAO） by immunohistochemical method. DESIGN： Completely randomly grouped controlled animal experiment. SETTING： Second Hospital, Xi＇an Jiaotong University. MATERIALS： Thirty-six Sprague-Dawley healthy male rats, with body mass of 250 - 330 g, were used in this study. Thirty-six rats were randomized into 3 groups： preischemia group, sham-operation group and control group, with 12 rats in each. METHODS： This study was carried out in the clinical laboratory, People＇s Hospital of Ningjin County of Shandong Province from March 2006 to January 2007. Rats in the pre-ischemia group were created into preischemia models by two embolisms twice. Three days after ischemic preconditioning, middle cerebral artery （MCA） was occluded for 2 hours with the same method. After being perfused for 22 hours, the rats were euthanized. In the sham-operation group, rats were not given the treatment of preischemia. In the first operation, only common carotid artery （CCA） and its crotch were exposed in the first operation, and MCA was not blocked by inserting embolism. At postoperative 3 days, rats were euthanized after being subjected to MCAO for 2 hours and reperfusion 22 hours by the same procedure as that in the preischemia group. As for each rat in the control group, only CCA and its crotch were exposed, and no any other treatment was carried out on them. MAIN OUTCOME MEASURES： Brain tissue of each rat was performed immunohistochemical staining at reperfusion 22 hours after preischemia, HIF-1 expression and brain infarct volume were detected. As a class of functional crystalline porous materials, metal-organic frameworks(MOFs) gained rapid development in the past three decades and a large number of MOFs with ordered structures, high surface areas, and functionalized channels have been investigated. MOFs and MOF-derived/composite materials show great potential in many application fields. In this review, we discussed the main applications of MOFs and MOF-derived/composite materials in small molecule storage, separation, luminescence, sensing, multitype catalysis, and energy storage. In addition, challenges and problems in the future research of MOFsrelated fields are also discussed.

Systematic research on the performance of self-designed microwave plasma reactor for CVD high quality diamond

Optical grade diamond, in particular, high-end products, are mainly used for aerospace and defense purposes. How to manufacture them with higher quality and lower cost was critical technology. Systematic research on the performance of Self-designed plasma reactor with higher power for synthesizing diamond film was carried out. Microwave input power can reach up to 10 kW, the plasma reactor has good adaptability for the deviation of microwave frequency f at 2.45 GHz ± 20 MHz. The secondary plasma could be eliminated by adjusting the working gas pressure and the cylinder adjustment structure,it is helpful to improve the deposition rate with less energy loss. The concentrated and steady plasma could be obtained device with higher gas pressure and relative lower microwave input power. Gas supply modes and inlet gas flow rate were optimized, which would be beneficial to synthesize the film with good quality.

AIM: To further investigate the role of human B7 homolog 1 (B7-H1) in the mechanism of persistent hepatitis B virus (HBV) infection. METHODS: Peripheral and intra-hepatic B7-H1 expression were compared by flow cytometry and immunochemical staining between two 2 distinct groups, one being chronic HBV tolerance patients (CHB-T) and the other being acute hepatitis B patients (AHB). B7-H1 mRNA expression level was also compared by real time polymerase chain reaction between CHB-T and AHB patients. The location of intra-hepatic B7-H1 and CD40 expression were analyzed by immunofluorescence. The levels of B7-H1 and CD40 expression on cultured myeloid dendritic cells (mDCs) with or without hepatitis B surface antigen (HBsAg) treatment were analyzed dynamically by flow cytometry. Intracellular interferon-γ (IFN-γ) staining and the stimulatory capacity of mDC of cultured mDC with or without HBsAg treatment were also compared by flow cytometry. RESULTS: Peripheral B7-H1 expression on mDCs was increased significantly in AHB compared to CHB-T patients (P sent despite a persistently elevated serum HBsAg load. In contrast, there were indeed increased B7-H1-positive cells in situ in the liver tissue from AHB. In vitro analysis showed the parallel upregulation of B7-H1 and CD40 on CD11c+ mDCs after the onset of stimulation. Addition of recombinant hepatitis B surface antigen (rHBsAg) significantly decreased CD40 expression (P < 0.05 at 16 h, 20 h and 24 h time points). B7-H1 expression was also inhibited by rHBsAg, and the inhibition rate of CD40 was greater than that of B7-H1. This preferential inhibition of CD40 expression on mDCs by rHBsAg resulted in the dysfunction of mDCs and T cells in the mixed leucocyte reaction (MLR) system. Retroperitoneal schwannoma mimicking metastatic seminoma: case report and literature review

If a testicular cancer pat ient has a mass in the retroperitoneum, a metastasis is often the first suspicion, probably leading to improper diagnosis and overtreatment. Here we report a case of retroperitoneal schwannoma mimicking metastatic seminoma. A 29-year-old man, who had a history of seminoma, presented with a single retroperitoneal mass suspected to be a metastasis. Because the patient refused radiotherapy, 3 cycles of cisplatin, etoposide, and bleomycin were offered. Post-chemotherapy computed tomography scan revealed persistence of the retroperitoneal mass, with no change in tumor size or characteristics. Subsequently, retroperitoneal lymph node dissection was performed. The dissected tissue contained negative lymph nodes but a single mass in the attached fat. Pathology revealed retroperitoneal schwannoma, which was confirmed by immunohistochemistry. Thus, clinicians should be aware of retroperitoneal schwannoma and its distinction from metastatic seminoma to avoid misdiagnosis and ensure proper treatment.

Autophagy plays a pivotal role in diverse biological processes,including the maintenance and differentiation of neural stem cells(NSCs).Interestingly,while complete deletion of Fip200 severely impairs NSC maintenance and differentiation,inhibiting canonical autophagy via deletion of core genes,such as Atg5,Atg16l1,and Atg7,or blockade of canonical interactions between FIP200 and ATG13(designated as FIP200-4A mutant or FIP200 KI)does not produce comparable detrimental *** highlights the likely critical involvement of the non-canonical functions of FIP200,the mechanisms of which have remained ***,utilizing genetic mouse models,we demonstrated that FIP200 mediates non-canonical autophagic degradation of p62/sequestome1,primarily via TAX1BP1 in *** deletion of Tax1bp1 in fip200hGFAP conditional knock-in(cKI)mice led to NSC deficiency,resembling the fip200hGFAP conditional knockout(cKO)mouse ***,reintroducing wild-type TAX1BP1 not only restored the maintenance of NSCs derived from tax1bp1-knockout fip200hGFAP cKI mice but also led to a marked reduction in p62 aggregate ***,a TAX1BP1 mutant incapable of binding to FIP200 or NBR1/p62 failed to achieve this ***,conditional deletion of Tax1bp1 in fip200hGFAP cKO mice exacerbated NSC deficiency and p62 aggregate accumulation compared to fip200hGFAP cKO ***,these findings illustrate the essential role of the FIP200-TAX1BP1 axis in mediating the non-canonical autophagic degradation of p62 aggregates towards NSC maintenance and function,presenting novel therapeutic targets for neurodegenerative diseases.

Impact of oral anti-hepatitis B therapy on the survival of patients with hepatocellular carcinoma initially treated with chemoembolization

Introduction:Most hepatocellular carcinomas(HCC) develop in a background of underlying liver disease including chronic hepatitis ***,the effect of antiviral therapy on the long-term outcome of patients with hepatitis B virus(HBV)-related HCC treated with chemoembolization is *** study aimed to evaluate the survival benefits of anti-HBV therapy after chemoembolization for patients with HBV-related ***:A total of 224 HCC patients who successfully underwent chemoembolization were identified,and their survival and other relevant clinical data were ***-Meier and Cox regression analyses were performed to validate possible effects of antiviral treatment on overall survival(OS).Results:The median survival time(MST) was 15.9(95%confidence interval[CI],9.5-27.7) months in the antiviral group and 9.6(95%CI,7.8-13.7) months in the non-antiviral group(log-rank test,P = 0.044).Cox multivariate analysis revealed that antiviral treatment was a prognostic factor for OS(P = 0.008).Additionally,a further analysis was based on the stratification of the TNM tumor *** the subgroup of early stages,MST was significantly longer in the antiviral-treatment group than in the non-antiviral group(61.8 months[95%CI,34.8 months to beyond the follow-up period]versus 26.2[95%CI,14.5-37.7]months,P= 0.012).Multivariate analysis identified antiviral treatment as a prognostic factor for OS in the early-stage subgroup(P = 0.006).However,in the subgroup of advanced stages,MST of the antiviral-treated group was comparable to that of the non-antiviral group(8.4[95%CI,5.2-13.5]months versus 7.4[95%CI,5.9-9.3]months,P = 0.219).Multivariate analysis did not indicate that antiviral treatment was a significant prognostic factor in this ***:Antiviral treatment is associated with prolonged OS time after chemoembolization for HCC,especially in patients with early-stage tumors.

The near-Earth(within~100 pc)supernova explosions in the past several million years can cause the global deposition of radioactive elements(e.g.,60Fe)on *** remnants of such supernovae are too old to be easily *** is therefore of great interest to search for million-year-old near-Earth neutron stars or black holes,the products of ***,neutron stars and black holes are challenging to find even in our Solar neighbourhood if they are not radio pulsars or X-ray/γ-ray *** we report the discovery of one of the nearest(127.7±0.3 pc)neutron star candidates in a detached single-lined spectroscopic binary LAMOST J235456.73+335625.9(hereafter J2354).Utilizing the time-resolved ground-based spectroscopy and space photometry,we find that J2354 hosts an unseen compact object with M_(inv)being 1.4-1.6 M_(⊙).The follow-up Swift ultraviolet(UV)and X-ray observations suggest that the UV and X-ray emission is produced by the visible star rather than the compact ***,J2354 probably harbours a neutron star rather than a hot ultramassive white ***-hour exceptionally sensitive radio follow-up observations with Five-hundred-meter Aperture Spherical radio Telescope fail to reveal any pulsating radio signals at the 6σflux upper limit of 12.5μ***,the neutron star candidate in J2354 can only be revealed via our time-resolved ***,the distance between J2354 and our Earth can be as close as~50 pc around 2.5 million years(Myrs)ago,as revealed by the Gaia *** discovery demonstrates a promising way to unveil the hidden near-Earth neutron stars in binaries by exploring the optical time domain,thereby facilitating understanding of the metal-enrichment history in our Solar neighbourhood.
