release Profile of Lansoprazole Encapsulated in Sodium Alginate Beads

Symptomatic gastroesophageal reflux disease (GErD) affects 11% of the population worldwide. Lansoprazole was the second commercially available proton pump inhibitor, recommended for healing and symptomatic relief of reflux esophagitis. This work presents the encapsulation of lansoprazole in sodium alginate beads, generating a controlled release system of the encapsulated drug. The quantification of lansoprazole contents from the encapsulated raw material was carried out, using the methodology prescribed in the Brazilian Pharmacopoeia 5th edition, with adjustments, the encapsulation efficiency and the release profile. Experimental tests were performed using scanning UV-Vis spectrophotometry. Encapsulation efficiency was 100%, obtained from the measurement of the absorbance of the filtrate. release profile shows the concentration of lansoprazole in the external environment was 4 μg/mL (16%) immediately after 10 min. Concentration of lansoprazole released by the beads follows a linear dynamic as a function of time, with a maximum release equivalent to 25 μg/mL after 210 min (100%). The rate of release is influenced by pH, being higher by 7.4. Beads subjected to heat treatment have a higher release rate at a temperature of 30 °C when compared to a temperature of 60 °C. Increase in temperature decreases the diameter of the beads. The presence of lansoprazole affects the release curve. The absorption rate of the microbeads with lansoprazole is 0.83, and the absorption rate of the microbeads without lansoprazole is 0.58. Sodium alginate beads have a good controlled release effect on lansoprazole.

The effect of angiotensin II (ANG II) or arginine vasopressin (AVP) alone or plus atrial natriuretic peptide (ANP) on H+-ATPase subcellular vesicle trafficking was investigated in mDCK cells following intracellular pH (pHi) acidification by exposure to20 mmNH4Cl for 2 min in a Na+-free solution containing Schering 28080, conditions under which H+-AT-Pase is the only cell mechanism for pHi recovery. Using the acridine orange fluorescent probe (5mm) and confocal microscopy, the vesicle movement was quantified by determining, for each experimental group, the mean slope of the line indicating the changes in apical/basolateral fluorescence density ratio over time during the first 5.30 min of the pHi recovery period. Under the control conditions, the mean slope was 0.079 ± 0.0033 min-1 (14) and it increased significantly with ANG II [10-12 and 10-7 m, respectively to 0.322 ± 0.038 min-1 (13) and 0.578 ± 0.061 min-1 (12)] or AVP [10-12 and 10-6 m, respectively to 0.301 ± 0.018 min-1 (12) and 0.687 ± 0.049 min-1 (11)]. However, in presence of ANP (10-6 m, decreases cytosolic free calcium), dimethyl-BAPTA/Am (5 × 10-5 m, chelates intracellular calcium) or colchicine (10-5 m, 2-h preincubation;inhibits microtubule-dependent vesicular trafficking) alone or plus ANG II or AVP the mean slopes were similar to the control values, indicating that such agents blocked the stimulatory effect of ANG II or AVP on vesicle trafficking. The results suggest that the pathway responsible for the increase in cytosolic free calcium and the microtu-bule-dependent vesicular trafficking are involved in this hormonal stimulating effect. Whether cytosolic free calcium reduction represents an important direct mechanism for ANP impairs the dose-dependent stimulatory effect of ANG II or AVP on H+-ATPase subcellular vesicle trafficking, or is a side effect of other signaling pathways which will require additional studies.

Context: Advanced heart failure (AHF) poses a global challenge, where heart transplantation is a treatment option but limited by donor scarcity. Proposal: This study aims to enhance the performance of ventricular assist devices (VADs) in the face of adverse events (AEs) using a resilience-based approach. The objective is to develop a method for integrating resilience attributes into VAD control systems, employing dynamic risk analysis and control strategies. results: The outcomes include a resilient control architecture enabling anticipatory, regenerative, and degenerative actions in response to AEs. A method of applied resilience (mAr) based on dynamic risk management and resilience attribute analysis was proposed. Conclusion: Dynamic integration between medical and technical teams allows continuous adaptation of control systems to meet patient needs over time, improving reliability, safety, and effectiveness of VADs, with potential positive impact on the health of heart failure patients.

The Gurupi Belt,in north-northeastern Brazil,is a mobile belt developed in the south-southwestern margin of the Sao Luis cratonic fragment and crops out as a tectonic and erosional window within the Phanerozoic ***,petrographic,geochemical,geochronological,and Nd isotopic information(new and published)constrain the timing and types of magmatic associations present in the belt and the tectonic settings in which they *** rhyacian was the main period of magmatic activity,which can be grouped into two main stages.(1)~2185-2130 ma:pre-collisional,juvenile,calc-alkaline magnesian and calcic ferroan granitoid suites,and minor calc-alkaline and tholeiitic mafic plutonism(now amphibolites),formed in intra-oceanic to transitional/continental arcs;and intra-or back-arc volcano-sedimentary basin.(2)~2125-2070 ma:syn-(two-mica granites)to late-collisional(potassic to shoshonitic granites and quartz-syenite)plutonic suites produced after crustal thickening and melting,with localized migmatization,that intruded during the compressive D1 deformational phase and concomitantly with greenschist to amphibolite *** is a zonation of the rhyacian episodes,with intra-oceanic stages occurring to the northeast,and the continental arc and collisional phases occurring to the southwest,indicating the presence of an active continental margin to the southwest,and subduction from NE to SW(present-day configuration).This magmatic framework is a continuation to the south of what is described for the Sao Luis cratonic fragment to the north,and the orogenic scenario is identical to what is observed for the same period in the West African Craton(Eburnean/Birrimian orogen),which additionally supports previous geological *** the Neoproterozoic,a few magmatic occurrences are *** extensional event allowed the intrusion of an anorogenic,nepheline syenite at ca.730 ma,which was followed by the intrusion of a crustal,calc-alkaline microtonalite,of uncertain

The study area is the Urucu Oil Province, municipality of Coari, State of Amazonas, Brazil. This research represents a contribution to the hydrogeological knowledge in the northern region of Brazil, particularly in the central part of the Amazon rainforest, where researches on isotopic are still incipient. The primary goal was to determine, by stable isotopes 18O and 2H measurements, interrelationships between surface water and groundwater, in order to understand the origin and mechanisms of groundwater recharge and discharge. For this, samples of rainwater, superficial water and groundwater were collected between June 2008 and may 2009 for stable isotopic analyzes. This understanding is important in cases of eventual contaminations of the area, which could degrade the water resources. The results show that the superficial waters are typically light waters and have meteoric origin, and the groundwater recharge is by direct rainfall infiltration with primary evaporation before reaching the groundwater table in the Icá-Solimoes Aquifer System. The isotopic signatures similarities between groundwater and superficial waters indicate both waters’ contributions in the streams and, therefore, in the Urucu river.

AIm To evaluate intra- and interobserver agreement in imaging features in inflammatory bowel disease and comparison with fecal calprotectin(FC) *** Our institutional computed tomography enterography(CTE) database was retrospectively queried to identify patients who underwent CTE from January 2014 to June 2015. Patient inclusion criteria were confirmed inflammatory bowel disease(IBD) and FC collected mo after CTE without any change in clinical treatment or surgical treatment during this interval. The exclusion criterion was poor image quality. Two blinded abdominal radiologists, with 12 and 3 years of experience analyzed the CTE regarding localization(small bowel, colonic, both, or no disease detected);type of IBD(inflammatory, stenosing, fistulizing, > 1 pattern, or normal); and signs of active disease(present or absent). In 42 of 44 patients evaluated, routine CTE reports were made by one of the readers who reevaluated the CTEs ≥ 6 mo later, to determine the intraobserver agreement. FC was considered a sign of disease activity when it was higher than 250 μg/*** Forty-four patients with IBD(38 with Crohn's disease and 6 with ulcerative colitis) were included. There was a moderate interobserver agreement regarding localization of IBD(k = 0.540), type of disease(k = 0.410) and the presence of active signs in CTE(k = 0.419). There was almost perfect intraobserver agreement regarding localization, type and signs of active disease in IBD. The k values were 0.902, 0.937 and 0.830, respectively. After a consensus between both radiologists regarding inflammatory activity in CTE, we found that 24(85.7%) of 28 patients who were classified with active disease had elevated FC, and six(37.5%) of 16 patients without inflammatory activity in CTE had elevated FC(P = 0.003). The correlation between elevated FC and the presence of active disease in CTE was significant(k = 0.495, P = 0.001).CONCLUSION We found almost perfect intraobserver and moderate interobserve

AIm To establish a permanent piwi like rNA-mediated genesilencing 1(PIWIL1) gene knockout in AGP01 gastric cancer cell line using CrISPr-Cas9 system and analyze phenotypic modifications as well as gene expression *** CrISPr-Cas9 system used was purchased from Dharmacon GE Life Sciences(Lafayette, CO, United States) and permanent knockout was performed according to manufacturer's recommendations. Woundhealing assay was performed to investigate the effect of PIWIL1 knockout on migration capability of cells and Boyden chamber invasion assay was performed to investigate the effect on invasion capability. For the gene expression analysis, a one-color microarray-based gene expression analysis kit(Agilent Technologies, Santa Clara, CA, United States) was used according to the protocol provided by the manufacturer. rESULTS PIWIL1 gene knockout caused a significant decrease in AGP01 migration capacity as well as a significant decrease in cell invasiveness. moreover, functional analysis based on grouping of all differentially expressed m rNAs identified a total of 35 genes(5 up-regulated and 30 down-regulated) encoding proteins involved in cellular invasion and migration. According to current literature, 9 of these 35 genes(DOCK2, ZNF503, PDE4 D, ABL1, ABL2, LPAr1, SmAD2, WASF3 and DACH1) are possibly related to the mechanisms used by PIWIL1 to promote carcinogenic effects related to migration and invasion, since their functions are consistent with the changes observed(being up-or down-regulated after knockout). CONCLUSION Taken together, these data reinforce the idea that PIWIL1 plays a crucial role in the signaling pathway of gastric cancer, regulating several genes involved in migration and invasion processes; therefore, its use as a therapeutic target may generate promising results in the treatment of gastric cancer.

m>Backgroundm>: The cognitive capacity is one of the determinants of quality of life in old age. In EEG, gamma rhythm represented electrical correlates of execution of cognitive activities. m>Objectivem>: To compare the cortical areas activated during executive activities through the gamma rhythm (25 - 45 Hz) and the performance in the Frontal Assessment Battery (FAB) among older people who exercised intellectual and mechanical work during their lifetime. m>methodsm>: We compared the electroencephalographic record of the gamma rhythm potential (25 - 45 Hz) in the cortical quadrants during the execution of the FAB among 13 elderly people who exercised intellectual activity and 12 elderly people who performed mechanical work. m>resultsm>: In the FAB, the intellectual group obtained a better performance with a mean score of 16.2/18, while the mechanical group obtained an average score of 14.9/18. Only a significant difference between groups was observed in the similarity test, with intellectual group leadership. There was no significant difference between groups in the overall analysis of gamma rhythm potential in the brain quadrants during the tests. m>Conclusionm>: Elderly people who performed intellectual activity performed better than the elderly who exercised mechanical work in the execution of the Frontal Evaluation Battery (FAB). There was a greater quantification of gamma rhythm in the group of elderly individuals who exercised intellectual activity, compared to those who exercised mechanical activity.
