Accuracy of two optical chlorophyll meters in predicting chemical composition and in vitro ruminal organic matter degradability of Brachiaria hybrid,Megathyrsus maximus, and Paspalum atratum

The objective of this study was to determine the accuracy and reliability of 2 optical chlorophyll meters:FieldScout CM 1,000 NDvI and Yara N-Tester, in predicting neutral detergent fibre(NDF), acid detergent fibre(ADF), acid detergent lignin(ADL), acid detergent insoluble nitrogen(ADIN) and in vitro ruminal organic matter degradability(IvOMD) of 3 tropical grasses. Optical chlorophyll measurements were taken at 3 stages(4, 8 and 12 weeks) of regrowth in Brachiaria hybrid, and Megathyrsus maximus and at 6 and 12 weeks of regrowth in Paspalum atratum(cv. Ubon). Optical chlorophyll measurements showed the highest correlation(r = 0.57 to 0.85) with NDF concentration. The FieldScout CM 1,000 NDvI was better than the Yara N-Tester in predicting NDF(R^2 = 0.70) and ADF(R^2 = 0.79) concentrations in Brachiaria hybrid and NDF(R^2 = 0.79) in M. maximus. Similarly, FieldScout CM 1,000 NDvI produced better estimates of 24 h IvOMD(IvOMD_(24 h)) in Brachiaria hybrid(R^2 = 0.81) and IvOMD_(48 h) in Brachiaria hybrid(R^2 = 0.65) and M. maximus(R^2 = 0.75). However, these prediction models had relatively low concordance correlation coefficients, i.e., CCC >0.90, but random errors were the main source of bias. It was,therefore, concluded that both optical chlorophyll meters were poor and unreliable predictors of ADIN and ADL concentrations. Overall, the FieldScout CM 1,000 NDvI shows potential to produce useful estimates of IvOMD_(24h) and ADF in Brachiaria hybrid and IvOMD_(48 h) and NDF concentrations in M. maximus.

The translocon on the outer membrane of mitochondria （TOM） facilitates the import of nuclear-encoded proteins. The principal machinery of mitochondrial protein transport seems conserved in eukaryotes; however, divergence in the composition and structure of TOM components has been observed between mammals, yeast, and plants. TOM9, the plant homolog of yeast Tom22, is significantly smaller due to a truncation in the cytosolic receptor domain, and its precise function is not understood. Here we provide evidence showing that TOM9.2 from Arabidopsis thaliana is involved in the formation of mature TOM com- plex, most likely by influencing the assembly of the pore-forming subunit TOM40. Dexamethasone- induced RNAi gene silencing of TOM9.2 results in a severe reduction in the mature TOM complex, and the assembly of newly imported TOM40 into the complex is impaired. Nevertheless, mutant plants are fully viable and no obvious downstream effects of the loss of TOM complex, i.e., on mitochondrial import ca- pacity, were observed. Furthermore, we found that TOM9.2 can bind calmodulin （CAM） in vitro and that CaM impairs the assembly of TOM complex in the isolated wild-type mitochondria, suggesting a regula- tory role of TOM9.2 and a possible integration of TOM assembly into the cellular calcium signaling network.

Injury to the central nervous system (CNS) is common, and though it has been well studied, many aspects of traumatic brain injury (TBI) and stroke are poorly understood. TBI and stroke are two pathologic events that can cause severe, immediate impact to the neurostructure and function of the CNS, which has been recognized recently to be exacerbated by the body's own immune response. Although the brain damage induced by the initial trauma is most likely unsalvageable, the secondary immunologic deterioration of neural tissue gives ample opportunity for therapeutic strategists seeking to mitigate TBI's secondary detrimental effects. The purpose of this paper is to highlight the cell death mechanisms associated with CNS injury with special emphasis on inflammation. The authors discuss sources of inflammation, and introduce the role of the spleen in the systemic response to inflammation after CNS injury.

SOCS3 dictates the transition of divergent time-phased events in granulocyte TNF-a signaling

Tumor-necrosis factor-a （TNF-a）-driven nuclear factor-KB （NF-KB） activation and apoptosis are opposing pathways; the growing recognition of these conflicting roles of TNF-a is perplexing. Here, we show that inflammation and apoptosis are time-phased events following TNF-a signaling and that emergence of suppressor of cytokine signaling 3 （SOCS3） expression limits the ongoing NF-KB activation and promotes apoptosis; further, we suggest an altered view of how inflammatory diseases are initiated and sustained. In vitro, TNF-a （50 ng/ml） induced granulocyte SOCS3 protein, inhibited nuclear accumulation of the p65NF-KB subunit and enhanced apoptosis, as shown by DNA laddering, annexin v positivity, and overexpression of caspase-3 and Bax in the late phase, whereas the early phase was marked by NF-KB activation. Conversely, SOCS3 knockdown by small interfering RNA （siRNA） inhibited granulocyte apoptosis and enhanced nuclear accumulation of p65 and 5＇ lipooxygenase expression in the late phase of TNF-a signaling. As apoptosis is associated with SOCS3 abundance, we suggest that these divergent TNF-a-driven events are time-phased, interconnected, opposing control mechanisms and one of the central features through which the immune system resolves pulmonary inflammation. Dysregulation may initiate mucosal inflammation, thus changing the landscape of asthma theraov.

European Association for the Study of the Liver and French hepatitis C recent guidelines: The paradigm shift

The latest Association Fran?aise pour l'Etude du Foie-French Association for Study of the Liver(AFEF) and European Association for the Study of the Liver(EASL) recommendations announce a change of paradigm, for the management of patients infected with hepatitis C virus(HCv). The AFEF recommendations focus on the elimination of HCv infection on a national level by preventing reinfection, in less than ten years. This goal involves the facilitation of patients' management in a simplified pathway by increasing screening procedures and access to pangenotypic treatments mainly in the "reservoir" population of people who inject drugs and migrants. Even in the complex pathway of patients with previous comorbidities, AFEF takes the option of a therapeutic simplification. The EASL guidelines position themselves on the state of the art with a precise description of all therapeutic options available, without separating simplified and complex pathways even if they take into account the epidemiological evolution of difficult-to-treat populations.
