Currently,1% of the United States population holds a diagnosis for celiac disease(CD),however,a more recently recognized and possibly related condition,"non-celiac gluten sensitivity"(NCGS)has been suggested to affect up to 6%of the United States *** reliable clinical tests for CD exist,diagnosing individuals affected by NCGS is still complicated by the lack of reliable biomarkers and reliance upon a broad set of intestinal and extra intestinal symptoms possibly provoked by *** has been proposed to exhibit an innate immune response activated by gluten and several other wheat *** present,an enormous food industry has developed to supply gluten-free products(GFP)with GFP sales in 2014 approaching$1 billion,with estimations projecting sales to reach$2 billion in the year *** enormous demand for GFP also reflects a popular misconception among consumers that gluten avoidance is part of a healthy lifestyle *** of NCGS and other gluten related disorders(e.g.,irritable bowel syndrome)call for a review of current distinctive diagnostic criteria that distinguish each,and identification of biomarkers selective or specific for *** aim of this paper is to review our current understanding of NCGS,highlighting the remaining challenges and questions which may improve its diagnosis and treatment.

Protein microarrays based on fluorescence detection have been widely utilized for high-throughput functional proteomic analysis. However, a drawback of such assays has been low sensitivity and narrow dynamic range, limiting their capabilities, especially for detecting low abundance biological molecules such as cytokines in human samples. Here, we present fluorescence-enhancing microarrays on plasmonic gold films for multiplexed cytokine detection with up to three orders of magnitude higher sensitivity than on conventional nitrocellulose and glass substrates. Cytokine detection on the gold plasmonic substrate is about one to two orders of magnitude more sensitive than enzyme-linked immunosorbent assay （ELISA） and can be multiplexed. A panel of six cytokines （Vascular endothelial growth factor （VEGF）, Interleukin 1β （IL-1β）, Interleukin 4 （IL-4）, Interleukin 6 （IL-6）, Interferon γ （IFN-γ）, and Tumor necrosis factor （TNF）） were detected in the culture media of cancer cells. This work establishes a new method of high throughput multiplexed cytokine detection with higher sensitivity and dynamic range than ELISA.

Longitudinal monitoring for the emergence of epidermal growth factor C797S resistance mutations in non-small cell lung cancer using blood-based droplet digital PCR

Validation of assays for the C797S mutation as a biomarker for osimertinib resistance is promising in guiding treatment decision-making for multidrug resistant non-small cell lung cancer. A newly developed droplet digital PCR (ddPCR) assay was used to retrospectively evaluate the emergence of the C797S mutation in six remnant plasma samples in this case report. It was found that the detected emergence of C797S clearly correlated with clinical signs of treatment resistance. Had these data been available to aid treatment selection in real time, there would have been hope for recaptured disease response and control instead of treatment cessation. The results of this study show that highly sensitive ddPCR methods can be used for the monitoring of emergent epidermal growth factor somatic variant mutations in circulation.

AIM: To investigate whether regional geography influences ethnic and gender trends for the development of gastric cancer(GC).METHODS: This retrospective analysis of the INVISION patient database at Louisiana State University Health Sciences Center-Shreveport(LSUHSC-S), a southern United States regional hospital, was performed from 2005-2011. Using the international statistical classification of diseases 9(ICD-9), inpatient, day surgery outpatient, and emergency outpatient diagnosis codes entered into medical records were used to identify GC patients. For each study year, the patients were evaluated for age, ethnicity, and gender, and each patient was counted only once throughout the study. Subsequent patient encounters were counted as visits and separated by inpatient and clinic visits. Complex or severe disease may require more frequent and intensive clinical management; therefore, we evaluated annual clinic visits as "surrogate markers" of disease severity. Finally, we studied the primary diagnosis for Helicobacter pylori(H. pylori) infection(ICD-9 code 41.86) as an additional factor that might increase the risk of ***: A total of 285 patients were diagnosed with GC at LSUHSC-S between 2005 and 2011. African Americans(181 patients, 89 males and 92 females, 63.5% of total patients) had significantly higher frequencies of GC diagnosis compared with non-Hispanic whites(104 patients, 54 males and 50 females, 36.5% of total patients), at a ratio of 1.74(P = 0.002). Within each ethnic group, men and women were diagnosed at approximately equal annual rates. Our findings differed significantly from United States national trends, which found that African American females and white females had lower risks for GC than their corresponding male counterparts. The United States national trend between 2005 and 2011 showed that African Americans males had a higher incidence of GC, with an annual mean(per 100000) of 16.31 ± 0.76 compared with white males(9 ± 0.1, P < 0.001),

Chronic inflammation results from excessive pro-inflammatory signaling and the failure to resolve the inflammatory *** mediators orchestrate both the initiation and resolution of *** from pro-inflammatory to pro-resolving lipid mediator biosynthesis is considered as efficient strategy to relieve chronic inflammation,though drug candidates exhibiting such features are *** from a library of Vietnamese medical plant extracts,we identified isomers of the biflavanoid 8-methylsocotrin-4’-ol from Dracaena cambodiana,which limit inflammation by targeting 5-lipoxygenase and switching the lipid mediator profile from leukotrienes to specialized pro-resolving mediators(SPM).Elucidation of the absolute configurations of 8-methylsocotrin-4’-ol revealed the 2 S,γSisomer being most active,and molecular docking studies suggest that the compound binds to an allosteric site between the 5-lipoxygenase *** identified additional subordinate targets within lipid mediator biosynthesis,including microsomal prostaglandin E2 *** production is efficiently suppressed in activated human neutrophils,macrophages,and blood,while the induction of SPM biosynthesis is restricted to M2 *** shift from leukotrienes to SPM was also evident in mouse peritonitis in vivo and accompanied by a substantial decrease in immune cell *** summary,we disclose a promising drug candidate that combines potent 5-lipoxygenase inhibition with the favorable reprogramming of lipid mediator profiles.

A two-dimensional discrete element model （DEM） simulation of cohesive polygonal particles has been developed to assess the benefit of point source vibration to induce flow in wedge-shaped hoppers. The particle-particle interaction model used is based on a multi-contact principle. The first part of the study investigated particle discharge under gravity without vibration to determine the critical orifice size （Bc） to just sustain flow as a function of particle shape. It is shown that polygonal-shaped particles need a larger orifice than circular particles. It is also shown that Bc decreases as the number of particle vertices increases. Addition of circular particles promotes flow of polygons in a linear manner. The second part of the study showed that vibration could enhance flow, effectively reducing Bc. The model demonstrated the importance of vibrator location （height）, consistent with previous continuum model results, and vibration amplitude in enhancing flow.

In this note, we provide a consistant thin layer theory for power law and Bingham incompressible fluids flowing down an inclined plane under the effect of gravity. The derivation of such equations is based on formal asymptotic expansions of solutions of Cauchy momentum equations in the shallow water scaling and in the neighbourhood of steady solutions so that we can close the average equations on the fluid height h and the total discharge rate q.

Sea wave energy generators or converters(WECs)have the potential to become a viable technology for clean,renewable energy *** the WEC technologies,the oscillating water columns(OWCs)are the most common WEC devices *** have been studied and developed over many ***-chamber oscillating water columns(MC-OWC)have the potential to have a higher energy conversion when extracting energy in mixed sea states than single-chamber *** the work reported in this paper,physical experiments are carried under regular wave conditions to test the wave power extraction of a fixed MC-OWC small-scale *** Power Take-Off(PTO)of the device is simulated using orifice *** flow characteristics through these orifices are pre-calibrated such that the extracted power can be obtained only using the pressure *** condition effects on the damping of the PTO of the device power extraction are *** test results illustrate that the PTO system damping is critical and affects device performance.
