Helicobacter pylori HopE and HopV porins present scarce expression among clinical isolates

AIM:To evaluate how widely Helicobacter pylori (H. pylori ) HopE and HopV porins are expressed among Chilean isolates and how seroprevalent they are among infected patients in ***: H. pylori hopE and hopV genes derived from strain CHCTX-1 were cloned by polymerase chain reaction (PCr), sequenced and expressed in Escherichia coli AD494 (DE3). Gel-purified porins were used to prepare polyclonal antibodies. The presence of both genes was tested by PCr in a collection of H. pylori clinical isolates and their expression was detected in lysates by immunoblotting. Immune responses against HopE, HopV and other H. pylori antigens in sera from infected and non-infected patients were tested by Western blotting using these sera as f irst antibody on recombinant H. pylori ***: PCr and Western blotting assays revealed that 60 and 82 out of 130 Chilean isolates carried hopE and hopV genes, respectively, but only 16 and 9, respectively, expressed these porins. IgG serum immunoreactivity evaluation of 69 H. pylori-infected patients revealed that HopE and HopV were infrequently recognized (8.7% and 10.1% respectively) compared to H. pylori VacA (68.1%) and CagA (59.5%) antigens. Similar values were detected for IgA serum immunoreactivity against HopE (11.6%) and HopV (10.5%) although lower values for VacA (42%) and CagA (17.4%) were obtained when compared to the IgG ***: A scarce expression of HopE and HopV among Chilean isolates was found, in agreement with the infrequent seroconversion against these antigens when tested in infected Chilean patients.

Utilization of Pyrrolic Compounds as Indicators of Secondary Migration for Woodford Oils in the Anadarko Basin, Oklahoma, USA

Migration pathways and distances of the oils in reservoir are thought to affect the distribution of pyrrolic compounds such as carbazole, its alkyl derivatives(alkylated carbazoles) and benzocarbazoles, although other factors, including maturity and depositional environments may also affect the distribution of these organic nitrogen compounds. In this study, 14 oil samples produced from conventional reservoirs in pauls Valley, south Oklahoma were investigated using organic geochemical techniques. The sterane and hopane fingerprints suggest that most of the oils were sourced from the Devonian Woodford shale. Maturity parameters consistently indicate that the maturity level of the studied samples are all of similar maturity(rc = ～0.7%), suggesting the distribution of the organic nitrogen organic compounds is possibly reflecting variations in relative migration distances. The distribution of alkylcarbazoles revealed a preferential enrichment during migration, with the nitrogen-shielded alkylcarbazole tending to be enriched relative to the nitrogen-semi-shielded alkylcarbozoles particularly in oils produced close to the Arbuckle uplift to the east. Correspondingly, another family of pyrrolic compounds, benzocarbazoles, whose distributions also indicated that the pauls Valley Woodford oils came from deeper part of the Anadarko Basin as the benzo-[a]/([a]+[c])-carbazole ratios decrease eastwards. In more specific migration systems, although the pyrrolic compound indicators are potentially disturbed by the structural complexes, the general migration directions suggest that the studied oils in pauls Valley Hunton uplift were sourced from the deep basin area, and migrate upwards in porous sediments due to the buoyancy. Plant-available water capacity of soils at a regional scale:Analysis of fixed and dynamic field capacities

Estimation of the plant-available water capacity(PAWC)of soils at a regional scale helps in adopting better land use planning,developing suitable irrigation schedules for crops,and optimizing the use of scarce water *** the current study,72 soil profiles were sampled from the Barossa region of South Australia to estimate pedo-transfer functions deduced from easily estimated soil *** functions were then used to estimate the fixed(10 and 33 kPa)and dynamic pressure head(h_(fc))water contents at field capacity(FC)for minimum drainage flux(0.01 and 0.001 cm d^(-1)),which serves as the upper boundary for plant-available water in *** estimated residual water content was corrected for subsoil constraints,especially the exchangeable sodium percentage(ESP).The results showed that the mean values of h_(fc)in sand-dominated light and medium textured soils(i.e.,sand,loamy sand,sandy loam,and loam)varied in a narrow range(15.8-18.2 kPa),whereas those in the clay-dominated heavy textured soils(i.e.,clay loam)showed a wide range(11.3-49.3 kPa).There were large differences in PAWC for dynamic FC(PAWC_(fc))and fixed FC at 10 kPa(PAWC10),33 kPa(PAWC33),and a mix of 10and 33 kPa(PAWC_(10,33))pressure heads depending on soil ***,the difference between PAWC at 10 kPa and h_(fc)(ΔPAWC_(10))was positive,whereas that between 33 kPa and h_(fc)(ΔPAWC_(33))was negative across all ***,the estimation of PAWC assuming a fixed FC at 10 and 33 kPa pressures(i.e.,PAWC_(10,33))for sandy,clay,and silty soils reduced the difference between fixed and dynamic pressure PAWCs toross the *** estimation of PAWC was improved by incorporating the impact of subsoil constraints,such as high ESP,which was more pronounced for clay and silty *** findings demonstrate the inherent inconsistencies between static pressure and flux-based dynamic FC estimations in *** heterogeneity,intra-texture variability,subsoil constraints,and swell

Hypoxic and temporal variation in the endocrine disrupting toxicity of perfluorobutanesulfonate in marine medaka(Oryzias melastigma)

Perfluorobutanesulfonate(PFBS)is an emerging pollutant capable of potently disrupting the sex and thyroid endocrine systems of ***,the hypoxic and temporal variation in PFBS endocrine disrupting toxicity remain largely *** the present study,adult marine medaka were exposed to environmentally realistic concentrations of PFBS(0 and 10μg/L)under normoxia or hypoxia conditions for 7 days,aiming to explore the interactive behavior between PFBS and *** addition,PFBS singular exposure was extended till 21days under normoxia to elucidate the time-course progression in PFBS *** results showed that hypoxia inhibited the growth and caused the suspension of egg spawn regardless of PFBS *** regard to the sex endocrine system,7-day PFBS exposure led to an acute stimulation of transcriptional profiles in females,which,subsequently,recovered after the 21-day *** potency of hypoxia to disturb the sex hormones was much stronger than PFBS.A remarkable increase in estradiol concentration was noted in medaka blood after hypoxia *** in sex endocrinology of coexposed fish were largely determined by hypoxia,which drove the formation of an estrogenic *** further enhanced the endocrine disrupting effects of ***,the hepatic synthesis of vitellogenin and choriogenin,two commonly used sensitive biomarkers of estrogenic activity,failed to initiate in response to the estrogen *** to sex endocrine system,disturbances in thyroidal axis by PFBS or hypoxia were relatively ***,the present findings will advance our toxicological understanding about PFBS pollutant under the interference of hypoxia.

Lactobacillus acidophilus and Bifidobacterium breve alleviate neonatal maternal separation induced visceral hypersensitivity and non-spatial memory impairment in rats by the NGF/TrkA pathway

Irritable bowel syndrome-diarrhea （IBS-D） is a prevalent functional bowel disease severely impairs the quality of life of patients. Visceral hypersensitivity （VH） is the main symptom of IBS-D. Probiotics have been found to effectively alleviate VH. In this study， neonatal maternal separation （NMS） was used to induce VH in 72 rats and investigated the effects of daily gavage of 3*109CFU of Lactobacillus and Bifidobacterium on alleviating the symptoms of IBS-D and behavioral interventions， as well as exploring potential mechanisms. The results demonstrated that Lactobacillus acidophilus and Bifidobacterium breve could alleviate the symptoms of IBS-D such as VH and increased frequency of bowel movements， as well as improving non-spatial memory in NMS rats. These effects were achieved by modulating the composition of gut microbiota and metabolites （including short-chain fatty acids， deoxycholic acid and adenosine）， and by regulating the expression of genes and proteins associated with the NGF/TrkA signaling pathway， tryptophan metabolism， and inflammatory factors in the hippocampus. This finding suggests that Lactobacillus acidophilus and Bifidobacterium breve have promising potential for the alleviation of IBS.
