Production and Partial Characterization of an Extracellular Phytase Produced by Muscodor sp. under Submerged Fermentation

In most of the raw materials of plant origin used in animal feed, a portion of the phosphorus is stored as phytic acid or phytate. Phytate is the main storage form of phosphorus in vegetables but is not readily assimilated into food at low concentrations of the enzyme phytase. In addition to making phosphorous unavailable, phytate binds divalent cations such as calcium, copper, magnesium, iron, manganese and zinc, preventing the absorption of these nutrients in the gut of the animal. Phytase promotes the hydrolysis of the phytate phosphorus-releasing molecule, thereby increasing its bioavailability in feed. Phytase is distributed in plant and animal tissues and it is synthesized by some species of bacteria and fungi. The addition of this enzyme in the diet of animals is essential to promote greater uptake of phosphorus and also contributes to a decrease in the levels of phosphorus excreted by animals, thus reducing the pollution caused by excess phosphorus in the environment. This work aimed to select a fungus that stands out in the production of phytase among 100 isolates from Brazilian caves belonging to the genera Aspergillus, Penicillium and Cladosporium and 13 endophytic fungi of the aerial part of the coffee plant. For selection, the fungi were cultured in medium containing phytic acid as a sole source of phosphorus. After seven days at 25 °C, we evaluated growth and enzyme production by the presence of the phytic acid halo degradation (Enzymatic Index-EI) surrounding the colonies. Forty-seven species produced phytase, and the fungi Penicillium minioluteum (CF279) and Muscodor sp. (UBSX) showed higher degradation halos, 2.41 and 4.46, respectively. Considering the Muscodor sp. as the main source of phytase, high enzymatic levels were obtained when the fungus was grown under submerged fermentation with initial pH of 5.0 using wheat bran as additional carbon source for 144 h, at 125 rpm and 30 °C. The Optimization of Aspergillus ***4 Tannase Production under Submerged Fermentation

Tannase is a hydrolytic enzyme that is involved in the biodegradation of tannins and it has biotechnological potential in the pharmaceutical, chemical, food and beverage industries. Microorganisms, especially filamentous fungi, are important tannase producers. The aims of this work were to find a potential tannase producer and to improve the cultivation conditions. Three Aspergillus species (A. japonicus 246A, A. tamarii 3 and Aspergillus sp. GM4) were investigated in different culture media (Adams, Czapeck, Khanna, M5 and Vogel) and inducers (1% and 2% tannic acid;1% green tea;1% methyl gallate;1% gallic acid). Aspergillus sp. GM4 and Adams medium were selected. The tannase production by Aspergillus sp. GM4 in Adams medium was induced in the presence of 2% (w/v) tannic acid and gallic acid as carbon sources, while green tea was not able to induce tannase production. The Plackett-Burman screening design was performed with the variables MgSO4, KH2PO4, yeast extract, tannic acid, agitation rate and salt solution. The variables MgSO4 and agitation rate were selected for the optimization of tannase production using a Central Composite Rotatable Design. Under optimized conditions, a 2.66-fold increase in the enzyme production was observed with small modifications in the medium composition.

Nutrient content affecting Spodoptera frugiperda and Dalbulus maidis occurrence in corn

This work aims to study the effect of the nutrient contents in the leaves of corn on the incidence of Spodopterafrugiperda （Smith）（Lepidoptera： Noctuidae） and Dalbulus maidis （Delong ＆ Wolcott）（Homoptera： Cicadellidae）. The treatments were represented by unfertilized corn plants and corn plants fertilized with 250 kg of the formulation 4-14-8 （N- P-K）＋ 100 kg of ammonium sulfate （AS）/ha and 500 kg of the formulation 4-14-8 （N-P-K）＋200 kg of AS/ha, arranged in randomized blocks with four replicates. Attacks by S. frugiperda and D. maidis were higher as the nitrogen, sulfur, calcium and copper content in the leaves of corn increased. The effects of calcium and copper found in this study were not consistent with that of lower susceptibility associated with higher levels of these nutrients mentioned in the literature. We believe that there was a cumulative effect of the nutrients altogether, building up a nutritiously better vegetable which in turn had less reserve for defense, or that the content of both nutrients in the leaves were not high enough to induce such effects.

Hepatic encephalopathy(HE)is a cognitive disturbance characterized by neuropsychiatric *** occurs in acute and chronic hepatic disease and also in patients with portosystemic *** presence of these portosystemic shunts allows the passage of nitrogenous substances from the intestines through systemic veins without liver ***,the embolization of these shunts has been performed tocontrol HE manifestations,but the presence of portal vein thrombosis is considered a *** this presentation we show a cirrhotic patient with severe HE and portal vein thrombosis who was submitted to embolization of a large portosystemic *** report:a 57 years-old cirrhotic patient who had been hospitalized many times for persistent HE and hepatic coma,even without precipitant *** had a wide portosystemic shunt and also portal vein *** abdominal angiography confirmed the splenorenal shunt and showed other *** larger shunt was embolized through placement of microcoils,and the patient had no recurrence of overt *** was a little increase of esophageal and gastric varices,but no endoscopic treatment was *** portosystemic shunts are frequent causes of recurrent HE in cirrhotic patients,portal vein thrombosis should be considered a relative contraindication to perform a shunt ***,in particular cases with many shunts and severe HE,we found that one of these shunts can be safely embolized and this procedure can be sufficient to obtain a good HE *** conclusion,we reported a case of persistent HE due to a wide portosystemic shunt associated with portal vein *** the patient had other shunts,she was successfully treated by embolization of the larger shunt.

XEN^(■) implant and trabeculectomy medium-term quality of life assessment and comparison of results

AIM: To evaluate and compare the quality of life of patients submitted to XEN~? implant or trabeculectomy and the relationship with potentially involved ***: A cross-sectional study of patients with advanced open-angle glaucoma who underwent implantation of XEN~?(group 1) and trabeculectomy(group 2) between October 2015 and February 2017. The studied variables were: age, gender, follow-up time, need of topical anti-hypertensive therapy, visual acuity and intraocular pressure(IOP). The quantification of the quality of life was attained through the Glaucoma Symptom Scale(GSS) questionnaire. RESULTS: Totally 34 eyes(34 patients) were included, 17 in each group. The mean GSS scores for group 1 were 42.6±6.8(median, 47; p25, 36.5; p75, 48.5) and for group 2 it was 41.6±7.0(median, 43; p25, 36.5; p75, 47.0; P=0.34). There was a strong negative correlation between the need for topical anti-hypertensive drugs and the GSS result in both groups(r=-0.88, P<0.01, r=-0.59, P=0.01, respectively) and a moderate negative correlation with IOP in group 1(r=-0.50, P=0.03).CONCLUSION: The analysis demonstrates the noninferiority of medium-term quality of life of one group in relation to the other(XEN~? implant and trabeculectomy). The number of topical anti-hypertensive drugs and IOP negatively influenced the quality of life.

Araucaria angustifolia Aboveground Roots Presented High Arbuscular Mycorrhizal Fungal Colonization and Diversity in the Brazilian Atlantic Forest

30 different arbuscular mycorrhizal fungi （AMF） species, distributed in different genera such as Glomus, Acaulospora, Scutellospora, Entrophospora, Ambisporu, Kuklospora, Gigaspora, and Archeospora, have been identified in the root zone of Araucaria angustifolia, known as Brazil Pine. During our AMF survey in this ecosystem, our attention was called to the presence of many superficially growing Araucaria roots. Our hypothesis was that these roots were colonized with AMF because of the presence of AMF spores in organic material aboveground. Samples of these superficial roots and the organic substrate they were growing on were evaluated for their mycorrhizal status. DNA was extracted from the AMF-colonized superficial roots and submitted to polymerase chain reaction （PCR） amplification using the NS31-AM1 primer pair, followed by cloning and sequencing. We found that the root colonization percentages were between 31% and 52%, and the number of AMF spores in the substrate ranged from 27 to 164 spores per 50 g dry substrate. The phylogenetic analyses and tree construction using maximum parsimony （MP） and neighbor-joining （N J） methods identified 13 different species of the phylum Glomeromycota belonging to the genera Glomus, Funneliformis, Rhizophagus, Gigaspora, Acaulospora, and Archaeospora, and five isolates were identified only at the genus level. To our knowledge, this is the first report on Araucaria angustifolia with roots growing aboveground, producing runner roots that develop on dead tree trunks and organic material. The higher colonization of the aboveground roots than those commonly found in belowground Araucaria roots suggests that they may present active metabolic uptake of nutrients.

Small wastewater treatment plants (WWTP) are frequently located, by necessity, in remote and isolated sites, which increases the difficulty of its energy supply. Some of them are located near the seaside, in environmental sensible zones, and due to tourism activity of these sites, seasonal effects related with population size fluctuation can occur, which can originate certain inefficiencies concerning WWTP design and energy supply. The objective of this paper is to describe a step by step procedure for evaluation of the wind potential of sites that are dependent of in-situ energy generation, as well as, a case study on the utilization of the potential wind energy in Magoito WWTP (5000 p.e.). The employed methodology comprised the collection of one year of in-situ wind data and its validation by comparison with historical data of about 10 years of a nearby anemometric station. The data provided by the two anemometric stations was statistically treated. It allowed the analysis of the results from the two stations are promissory in terms of wind availability and velocity. Finally, it comprised the simulation of the local wind conditions for a considerable larger area in order to find the best site for locating a wind turbine.
