To better eluc.date epigenetic.mec.anisms that c.rrelate with the dynamic.gene expression program observed upon T-c.ll differentiation, we investigated the genomic.landsc.pe of histone modific.tions in naive and memory c.8＋ T c.lls. Using a c.lP-Seq approac. c.upled with global gene expression profiling, we generated genome-wide histone H3 lysine 4 （H3K4me3） and H3 lysine 27 （H3K27me3） trimethylation maps in naive, T memory stem c.lls, c.ntral memory c.lls, and effec.or memory c.lls in order to gain insight into how histone arc.itec.ure is remodeled during T c.ll differentiation. We show that H3K4me3 histone modific.tions are assoc.ated with ac.ivation of genes, while H3K27me3 is negatively c.rrelated with gene expression at c.nonic.l loc. and enhanc.rs assoc.ated with T-c.ll metabolism, effec.or func.ion, and memory. Our results also reveal histone modific.tions and gene expression signatures that distinguish the rec.ntly identified T memory stem c.lls from other c.8＋ T-c.ll subsets. Taken together, our results suggest that c.8＋ lymphoc.tes undergo c.romatin remodeling in a progressive fashion. These findings have major implic.tions for our understanding of peripheral T-c.ll ontogeny and the formation of immunologic.l memory.

In the c.ntral parts of the Himalaya,the Higher Himalayan c.ystalline (HHc. high-grade roc.s are mainly exhumed,as a c.nsequenc. of duc.ile flow at deeper level c.ntrolled by folding and broad duc.ile top-to-SW shearing between the Main c.ntral Thrust (Mc.) in the south and the South Tebetan Detac.ment System (STDS) in the *** rec.rds folding history of pre-Himalayan (D1) to Himalayan deformations (D2/D3/D4).The roc.s of the HHc.are thoroughly transposed by D2 deformation into Sm (S1+S2).The extent of transposition and a well-developed NE-plunging L2 lineation indic.te intense strain during D2 throughout the studied portion of the *** flow c.ntinued,resulting in rotation of F1 and F2 folds due NE-direc.ion and generation of NW-SE plunging F3 folds within the HHc.S-c.fabric., asymmetric.augen,asymmetric.pressure shadows,detac.ment of quartz-feldspathic.layer etc.are the shear fabric.developed during D2 duc.ile deformation that show top-to-SW *** HHc.then formed a c.ustal wedge that was rapidly exhumed at deeper level attributed to duc.ile flow. Atherosc.erosis is a c.ronic.inflammatory disease that is c.arac.erized by the build-up of lipid-ric. plaques in the arterial walls. The standard treatment for patients with atherosc.erosis is statin therapy aimed to lower serum lipid levels. Despite its widespread use, many patients taking statins c.ntinue to experienc. ac.te events. Thus, to develop improved and alternative therapies, we previously reported on mic.oRNA-145 (miR-145 mic.lles) and its ability to inhibit atherosc.erosis by targeting vasc.lar smooth musc.e c.lls (VSMc.). Importantly, one dose of miR-145 mic.lles signific.ntly abrogated disease progression when evaluated two weeks post-administration. Thus, in this study, to evaluate how long the sustained effec.s of miR-145 mic.lles c.n be maintained and towards identifying a dosing regimen that is prac.ic.l for patients with c.ronic.disease, the therapeutic.effec.s of a single dose of miR-145 mic.lles were evaluated for up to two months in vivo. After one and two months post-treatment, miR-145 mic.lles were found to reduc. plaque size and overall lesion area c.mpared to all other c.ntrols inc.uding statins without c.using adverse effec.s. Furthermore, a single dose of miR-145 mic.lle treatment inhibited VSMc.transdifferentiation into pathogenic.mac.ophage-like and osteogenic.c.lls in plaques. Together, our data shows the long-term effic.c. and sustained effec.s of miR-145 mic.lles that is amenable using a dosing frequenc. relevant to c.ronic.disease patients.

Hepatoc.llular c.rc.noma in non-alc.holic.steatohepatitis:c.rrent knowledge and implic.tions for management

With the prevalenc. of hepatitis c.virus expec.ed to dec.ine, the proportion of hepatoc.llular c.rc.noma(Hc.) related to non-alc.holic.steatohepatitis(NASH) is antic.pated to inc.ease exponentially due to the growing epidemic.of obesity and diabetes. The annual inc.denc. rate of developing Hc. in patients with NASH-related c.rrhosis is not c.early understood with rates ranging from 2.6%-12.8%. While multiple new mec.anisms have been implic.ted in the development of Hc. in NASH; further prospec.ive long-term studies are needed to validate these findings. Rec.nt evidenc. has shown a signific.nt proportion of patients with non-alc.holic.fatty liver disease and NASH progress to Hc. in the absenc. of c.rrhosis. Liver resec.ion and transplantation represent c.rative therapeutic.options in selec. NASHrelated Hc. patients but have plac.d a signific.nt burden to our healthc.re resourc.s and utilization. c.rrently NASH-related Hc. is the fastest growing indic.tion for liver transplant in Hc. c.ndidates. Inc.eased efforts to implement effec.ive sc.eening and preventative strategies, partic.larly in non-c.rrhotic.NASH patients, are needed to reduc. the future impac. imposed by NASH-related Hc..
